A wound-inducible tobacco peroxidase gene expresses preferentially in the vascular system.

A tobacco peroxidase gene tpoxN1 was reported to be expressed within 1 h after wounding in leaves [Hiraga et al. (2000a) Plant Cell Physiol. 41: 165]. We describe here further results on the wound-induced tpoxN1 expression. The quick tpoxN1 induction occurred preferentially in stems and petioles, but was negligible in leaf blades even 8 h after wounding. Induced GUS activity was also detected rapidly after wounding in the stem of transgenic tobacco plants carrying the tpoxN1 promoter::GUS fusion gene, localized mainly in the vascular systems where it was maintained this level for 14 d or more.

Enhanced Drought Stress Tolerance by the Arbuscular Mycorrhizal Symbiosis in a Drought-Sensitive Maize Cultivar Is Related to a Broader and Differential Regulation of Host Plant Aquaporins than in a Drought-Tolerant Cultivar.

The arbuscular mycorrhizal (AM) symbiosis has been shown to improve maize tolerance to different drought stress scenarios by regulating a wide range of host plants aquaporins. The objective of this study was to highlight the differences in aquaporin regulation by comparing the effects of the AM symbiosis on root aquaporin gene expression and plant physiology in two maize cultivars with contrasting drought sensitivity. This information would help to identify key aquaporin genes involved in the enhanced drought tolerance by the AM symbiosis. Results showed that when plants were subjected to drou

The gene expression profiles of mitochondrial respiratory components in Arabidopsis plants with differing amounts of ALTERNATIVE OXIDASE1a under high intensity light.

We compared the expression of mitochondrial alternative oxidase (AOX) and other non-phosphorylating respiratory components (NPhPs) in wild type and AOX1a transgenic Arabidopsis thaliana following short-term transfer of plants to higher irradiance conditions to gain more insight into the mechanisms of AOX functioning under light. The AOX1a overexpressing line (XX-2) showed the highest amount of AOX1a transcripts and AOX1A synthesis during the entire experiment, and many NPhPs genes were down-regulated after 6-8 h under the higher light conditions. Antisense AS-12 plants displayed a compensatory

Molecular, biochemical, and organismal analyses of tomato plants simultaneously attacked by herbivores from two feeding guilds.

Previous work identified aphids and caterpillars as having distinct effects on plant responses to herbivory. We sought to decipher these interactions across different levels of biological organization, i.e., molecular, biochemical, and organismal, with tomato plants either damaged by one 3rd-instar beet armyworm caterpillar (Spodoptera exigua), damaged by 40 adult potato aphids (Macrosiphum euphorbiae), simultaneous damaged by both herbivores, or left undamaged (controls). After placing insects on plants, plants were transferred to a growth chamber for 5 d to induce a systemic response. Subseq

Changes in the translational landscape during red clover necrotic mosaic virus infection.

Viruses alter host gene expression to create a proviral environment, while the host simultaneously regulates gene expression to restrict viral spread. Owing to RNA viruses' complete reliance on the host translational machinery, it is important to assess translational control during virus infection. Therefore, we used ribosome profiling (Ribo-seq) paired with RNA-seq to observe how red clover necrotic mosaic virus (RCNMV) infection of Arabidopsis plants alters cellular gene expression at the levels of mRNA abundance and translation efficiency. We determined that at 5 days post-inoculation (dpi)

Interplay of auxin, KANADI and Class III HD-ZIP transcription factors in vascular tissue formation.

Class III HD-ZIP and KANADI gene family members have complementary expression patterns in the vasculature and their gain-of-function and loss-of-function mutants have complementary vascular phenotypes. This suggests that members of the two gene families are involved in the establishment of the spatial arrangement of phloem, cambium and xylem. In this study, we have investigated the role of these two gene families in vascular tissue differentiation, in particular their interactions with the plant hormone auxin. We have analyzed the vasculature of plants that have altered expression levels of Cl

FAST-TRACE: delineating tissue-specific cis-regulatory activities in the cadmium/zinc hyperaccumulator Sedum plumbizincicola.

FAST-TRACE is a transient expression system for rapid homologous promoter analysis in Sedum plumbizincicola, enabling identification of critical regulatory regions and a MYB transcription factor, SpTRB4, that activates the SpHMA3 promoter. Investigating the SpHMA3 promoter in Sedum plumbizincicola is essential to elucidate the molecular regulatory mechanisms underlying the above-ground metal detoxification. Given that generating stably transformed plants is time-consuming and costly, an efficient, low-cost method for the preliminary promoter activity assessment is needed. Here, we present FAST

Visualization of auxin responses during vine twining in Ipomoea nil and the functional role of the subapical hook in this process.

This study suggests that hook-shaped bending associated with properly regulated auxin response patterns in the subapical region may contribute to the maintenance of twining in morning glory vines. The twining behavior demonstrated by vine plants represents an important morphogenetic adaptation essential to their survival; however, the physiological mechanisms driving this response remain largely unknown. To elucidate the role of auxin in vine twining in morning glory (Ipomoea nil), we generated and investigated transgenic morning glory carrying the GUS reporter gene under an auxin-responsive p

A mutation in the intron splice acceptor site of a GA3ox gene confers dwarf architecture in watermelon (Citrullus lanatus L.).

Dwarf architecture is an important trait associated with plant yield, lodging resistance and labor cost. Here, we aimed to identify a gene causing dwarfism in watermelon. The 'w106' (dwarf) and 'Charleston Gray' (vine) were used as parents to construct F1 and F2 progeny. Dwarf architecture of 'w106' was mainly caused by longitudinal cell length reduction and was controlled by a single recessive gene. Whole-genome sequencing of two parents and two bulk DNAs of F2 population localized this gene to a 2.63-Mb region on chromosome 9; this was further narrowed to a 541-kb region. Within this region,

Combined transcription factor profiling, microarray analysis and metabolite profiling reveals the transcriptional control of metabolic shifts occurring during tomato fruit development.

Maturation of fleshy fruits such as tomato (Solanum lycopersicum) is subject to tight genetic control. Here we describe the development of a quantitative real-time PCR platform that allows accurate quantification of the expression level of approximately 1000 tomato transcription factors. In addition to utilizing this novel approach, we performed cDNA microarray analysis and metabolite profiling of primary and secondary metabolites using GC-MS and LC-MS, respectively. We applied these platforms to pericarp material harvested throughout fruit development, studying both wild-type Solanum lycopers

Comparative macroarray analysis of morphine containing Papaver somniferum and eight morphine free Papaver species identifies an O-methyltransferase involved in benzylisoquinoline biosynthesis.

Benzylisoquinoline alkaloids constitute a group of about 2,500 structures and are mainly produced by plants of the order Ranunculales. But only the opium poppy, Papaver somniferum, and Papaver setigerum are able to produce morphine. In this study, we started to investigate by gene expression analysis the molecular basis for this exceptional biosynthetic ability. A sequencing project from P. somniferum seedlings was initiated using a method based on the amplified fragment length polymorphism technique that resulted in 849 UniGenes. These cDNAs were analysed on macroarrays for differential expre

Transcriptome Analysis of Choke Stroma and Asymptomatic Inflorescence Tissues Reveals Changes in Gene Expression in Both Epichloë festucae and Its Host Plant Festuca rubra subsp. rubra.

Many cool-season grasses have symbiotic relationships with Epichloë (Ascomycota, Clavicipitaceae) fungal endophytes that inhabit the intercellular spaces of the above-ground parts of the host plants. The presence of the Epichloë endophytes is generally beneficial to the hosts due to enhanced tolerance to biotic and abiotic stresses conferred by the endophytes. Many Epichloë spp. are asexual, and those infections always remain asymptomatic. However, some Epichloë spp. have a sexual stage and produce a macroscopic fruiting body, a stroma, that envelops the developing inflorescence causing a synd

TGW1a locus simultaneously shortens growth duration and boosts grain yield in rice.

Reconciling the trade-off between short growth duration and high grain yield is essential for enhancing annual rice yields. We found that qTGW1a, encoding a flowering locus T-like protein, controls heading and nitrogen use efficiency (NUE) underpinning grain weight and yield in rice. TGW1a interacts with and stabilizes Ghd7 and Hd1 to boost NUE and grain yield. Natural variations in the promoter enable the ancestral allele TGW1aJZ to maintain an intermediate level of TGW1a transcription. By decoupling its linkage to the weak grain number regulator Gn1aJZ, TGW1aJZ conferred 3.67 to 8.67 days sh

The glutathione-related metabolism and the AhGST23 gene mediate drought and salt stresses tolerance in peanut (Arachis hypogaea L.).

BACKGROUND: Abiotic stresses such as drought and salinity significantly constrain peanut productivity. However, their underlying molecular response mechanisms remain unclear. RESULTS: This study identified the AhGST23 gene, together with its involvement in glutathione metabolism and the ascorbic acid-glutathione (AsA-GSH) cycle, as a key component in alleviating drought and salt stresses in peanut. Under drought (15% PEG6000 for 6 days) and salt (200 mM NaCl for 6 days) stresses, peanut seedlings exhibited a marked reduction in biomass, net photosynthetic rate, transpiration rate, and chloroph

Genome-wide identification of MGT gene family in soybean (Glycine max) and their expression analyses under magnesium stress conditions.

BACKGROUND: Magnesium (Mg) is essential for plant growth and development and plays critical roles in physiological and biochemical processes. Mg deficiency adversely affects growth of plants by limiting shoot and root development, disturbing the structure and membranes of the grana, reducing photosynthesis efficiency, and lowering net CO2 assimilation. The MGT (Magnesium transporter) family is responsible for the absorption and transportation of magnesium in plants. Although the MGT family has been identified in different plant species, research regarding the soybean MGT genes is limited. RES

Studying Plant MIF/D-DT-Like Genes and Proteins (MDLs).

Human macrophage migration inhibitory factor (MIF) is an inflammatory cytokine with chemokine-like characteristics and an upstream regulator of host innate immunity. It is a critical mediator of a variety of human diseases, such as acute and chronic inflammatory diseases, autoimmunity, atherosclerosis, and cancer. MIF is an atypical chemokine that not only signals through its cognate receptor CD74, but also interacts with the classical chemokine receptors CXCR2 and CXCR4. MIF and its homolog D-dopachrome tautomerase (D-DT)/MIF-2 are structurally unique proteins that are conserved across kingdo

NbALD1 mediates resistance to turnip mosaic virus by regulating the accumulation of salicylic acid and the ethylene pathway in Nicotiana benthamiana.

AGD2-LIKE DEFENCE RESPONSE PROTEIN 1 (ALD1) triggers plant defence against bacterial and fungal pathogens by regulating the salicylic acid (SA) pathway and an unknown SA-independent pathway. We now show that Nicotiana benthamiana ALD1 is involved in defence against a virus and that the ethylene pathway also participates in ALD1-mediated resistance. NbALD1 was up-regulated in plants infected with turnip mosaic virus (TuMV). Silencing of NbALD1 facilitated TuMV infection, while overexpression of NbALD1 or exogenous application of pipecolic acid (Pip), the downstream product of ALD1, enhanced res

Detailed analysis of the expression of an alpha-gliadin promoter and the deposition of alpha-gliadin protein during wheat grain development.

BACKGROUND AND AIMS: Alpha-gliadin proteins are important for the industrial quality of bread wheat flour, but they also contain many epitopes that can trigger celiac (coeliac) disease (CD). The B-genome-encoded alpha-gliadin genes, however, contain very few epitopes. Controlling alpha-gliadin gene expression in wheat requires knowledge on the processes of expression and deposition of alpha-gliadin protein during wheat grain development. METHODS: A 592-bp fragment of the promotor of a B-genome-encoded alpha-gliadin gene driving the expression of a GUS reporter gene was transformed into wheat.

Root-specific expression of the LeRse-1 gene in tomato is induced by exposure of the shoot to light.

The tomato gene LeRse-1 was isolated from a root hair-specific cDNA library. The amino acid sequence of the LeRse-1 product displayed a high degree of identity to that of AgMtd, a mannitol dehydrogenase-encoding gene from celery. Expression of LeRse-1 was found to be restricted to the root organs of tomato, with no detectable transcripts in stems and leaves. Transcripts of LeRse-1 were detected in root hairs, as well as other root tissues of mature plants and seedlings. Root-specific LeRse-1 expression increased upon exposure of the shoot to light. Since roots were kept in darkness, shoot-to-r

Early steps of adventitious rooting: morphology, hormonal profiling and carbohydrate turnover in carnation stem cuttings.

The rooting of stem cuttings is a common vegetative propagation practice in many ornamental species. A detailed analysis of the morphological changes occurring in the basal region of cultivated carnation cuttings during the early stages of adventitious rooting was carried out and the physiological modifications induced by exogenous auxin application were studied. To this end, the endogenous concentrations of five major classes of plant hormones [auxin, cytokinin (CK), abscisic acid, salicylic acid (SA) and jasmonic acid] and the ethylene precursor 1-aminocyclopropane-1-carboxylic acid were ana
