Lack of association between leptin G2548A gene polymorphism and Behçet's disease.

BACKGROUND: Behçet's disease is a chronic, multisystem, inflammatory disease characterized by the predominance of T-helper 1 cytokines. The disease is also characterized by infiltration of lymphocytes and neutrophils into the affected tissues. Because cytokines are involved in the regulation of lymphocyte and phagocyte functions, they may play an important role in the pathogenesis of Behçet's disease. Leptin, a member of the gp 130 family of cytokines, induces a strong T-helper 1 response and is regarded as a proinflammatory inducer. Recent studies have shown that serum leptin concentration wa

HLA alleles in pre-menopausal breast cancer patients from western India.

BACKGROUND & OBJECTIVES: Cytotoxic function of Natural Killer (NK) cells is regulated by the products of HLA class I genes. Associations between HLA alleles and risk for cancers have been suggested earlier. Several reports further emphasize the need to examine influence of HLA genotypes on risk for malignant disorders. Therefore, we undertook this study to assess the possibility of association of HLA-class I alleles in pre-menopausal breast cancer patients. METHODS: Eighty one pre-menopausal breast cancer patients and 160, age and ethnicity matched healthy women from western India were studie

Deletions of the homeobox gene SHOX (short stature homeobox) are an important cause of growth failure in children with short stature.

Short stature, with an incidence of 3 in 100, is a fairly frequent disorder in children. Idiopathic short stature refers to patients who are short due to various unknown reasons. Mutations of a human homeobox gene, SHOX (short stature homeobox), have recently been shown to be associated with the short stature phenotype in patients with Turner syndrome and most patients with Léri-Weill dyschondrosteosis. This study addresses the question of the incidence and type of SHOX mutations in patients with short stature. We analyzed the SHOX gene for intragenic mutations by single strand conformation po

Relaxed replication of mtDNA: A model with implications for the expression of disease.

Heteroplasmic mtDNA defects are an important cause of human disease with clinical features that primarily involve nondividing (postmitotic) tissues. Within single cells the percentage level of mutated mtDNA must exceed a critical threshold level before the genetic defect is expressed. Although the level of mutated mtDNA may alter over time, the mechanism behind the change is not understood. It currently is not possible to directly measure the level of mutant mtDNA within living cells. We therefore developed a mathematical model of human mtDNA replication, based on a solid foundation of experim

The role of the SCA2 trinucleotide repeat expansion in 89 autosomal dominant cerebellar ataxia families. Frequency, clinical and genetic correlates.

The spinocerebellar ataxia type 2 (SCA2) is caused by a trinucleotide (CAG) expansion in the coding region of the ataxin 2 gene on chromosome 12q.89 families with autosomal dominant cerebellar ataxia (ADCA) types I, II and III, and 47 isolated cases with idiopathic late onset cerebellar ataxia (ILOCA), were analysed for this mutation. The identification of the SCA2 mutation in 31 out of 38 families with the ADCA I phenotype, but in none of those with ADCA II, ADCA III or ILOCA confirms the specificity of this mutation. A clinical comparison of the ADCA I patients with the three known mutations

Genetics of Indian childhood cirrhosis.

Indian Childhood Cirrhosis (ICC) is a unique syndrome with characteristic clinical, epidemiological and histopathological features which is a major cause of mortality in India in children 1 to 4 years of age. The aetiopathogenesis of this invariably fatal disease is still obscure. Various theories of its aetiopathogenesis include genetic, viral, metabolic, toxic, autoimmune or a combination of factors. The present article deals with a brief review of literature to elucidate the possible genetic mechanisms involved. In earlier reports autosomal recessive (AR) mode of inheritance was suggested.

Genetic markers in Welsh gypsies.

Data are presented on polymorphic genetic markers of 84 Welsh gypsies, members of an inbred population characterised by a high incidence of phenylketonuria and other recessively inherited diseases. Sixteen polymorphic loci were studied: the ABO blood group distribution was comparable to that of the surrounding population, the B gene frequency being 7-5%, a lower frequency than most earlier studies in gypsies. The haptoglobin 1 gene frequency (23-7%) was very different from that of the British population, resembling the frequency found in India and some previously reported gypsy groups. The dat…

On the mutation rate of neurofibromatosis.

A genetic study of 124 cases of neurofibromatosis was performed. The contingent of probands was mainly represented by a Russian population, most of the individuals being born in the European part of the RSFSR. Both parents of the probands were examined in only 58 cases, the proportion of sporadic cases in this group being 0.79, as compared to 0.77 for the whole group under study. The existing data evaluated by a direct method are not yet sufficient for a decisive estimation of the penetrance, which, however, cannot be under 80%. XG: the forgotten blood group system.

The XG blood group system is best known for its contributions to the fields of genetics and chromosome mapping. This system comprises two antigens, Xg(a) and CD99, that are not antithetical but that demonstrate a unique phenotypic relationship. XG is located on the tip of the short arm of the X chromosome with exons 1 to 3 present in the pseudoautosomal region of the X (and Y) chromosome(s) and exons 4 to 10 located only on the X chromosome. Xg(a) demonstrates a clear X-linked pattern of inheritance. MIC2, the gene encoding the CD99 antigen, is found in the pseudoautosomal region of both the X

[Polymorphism analysis of G199A, Ncol in ANK1 and Memphis I in SLC4A1 genes in Mexican healthy individuals and subjects affected with hereditary spherocytosis].

BACKGROUND: In Mexico, Hereditary Spherocytosis (HS) is the main cause of hereditary hemolytic anemia, due to mutations of one or more genes involved in the erythrocyte membrane, making it difficult to identify the primary gene. OBJECTIVE: With the purpose of estimating the use of the polymorphisms G199A and NcoI of ANK1 gene, and Memphis I of SLC4A1 gene, as genetic markers to screen this disease, we searched the allelic and genotypic frequencies in 45 DNA samples of HS patients and 28 from healthy individuals. RESULTS: Allelic and genotypic frequencies were similar in both studied groups f

Maternal gestational mercury exposure in relation to cord blood T cell alterations and placental gene expression signatures.

The immunotoxic impacts of mercury during early life is poorly understood. We investigated the associations between gestational mercury exposure and frequency of cord blood T cells as well as placental gene expression. Frequency of natural Treg cells was positively associated with prenatal and postpartum mercury toenail concentrations. Frequency of NKT and activated naïve Th cells was positively associated with prenatal toenail mercury concentrations and number of maternal silver-mercury dental amalgams, respectively. Placental gene expression analyses revealed distinct gene signatures associa

Analysis of IL-1A(-889) and TNFA(-308) gene polymorphism in Brazilian patients with generalized aggressive periodontitis.

Generalized aggressive periodontitis (GAP) comprises a group of periodontal diseases characterized by the rapid destruction of periodontal tissues which affect young individuals who generally present no systemic disorders. Polymorphisms in the interleukin-1 (IL-1) and tumor necrosis factor-alpha (TNF-alpha) genes have been associated with an increased severity of chronic periodontitis. The objective of the present study was to evaluate the association between IL-1A (-889) and TNFA (-308) gene polymorphisms and GAP. One hundred nonsmoking subjects were selected, including 30 with GAP and 70 wit

Paternal genetic structure of an endogamous Saraiki population from Pakistan inferred from rapidly mutating Y-STRs.

BACKGROUND: Y-chromosomal short tandem repeats (Y-STRs) are widely used in forensic genetics, but conventional panels show limited diversity and poor resolution among paternal relatives in endogamous populations. Rapidly mutating (RM) Y-STRs offer higher mutation rates to overcome these limitations. AIM: To evaluate the forensic and population genetic utility of 13 RM Y-STRs in the endogamous Saraiki population of Pakistan. SUBJECTS AND METHODS: Thirteen RM Y-STR loci were analysed in 100 unrelated Saraiki males. Haplotype diversity, discrimination capacity, allele frequencies, and gene dive

Alternative Reproductive Tactic Frequencies Vary with Stream Size for an Appalachian Salamander.

AbstractSpatial variation in allele frequencies can provide clues about the mechanisms responsible for the maintenance of genetic polymorphisms. In some populations of two-lined salamanders, males exhibit two alternative reproductive tactics determined by a Y-linked genetic polymorphism: searching males locate and court females in terrestrial habitats, and guarding males defend mates at aquatic nesting sites. Here, we study the fine-scale spatial variation in the frequency of these tactics by genotyping >1,000 total larvae from five pairs of small and large stream reaches in the Appalachian Mo

GWAS With a Global Population of Wheat Stripe Rust Identified High-Confidence Avirulence Gene Candidates.

Wheat stripe rust, caused by Puccinia striiformis f. sp. tritici (Pst), is a globally destructive disease characterised by rapid virulence evolution. Despite its significance, the genetic basis of avirulence variation at a global scale remains poorly understood. In this study, we integrated virulence phenotypes against 18 Yr resistance genes with genome-wide variation from a worldwide collection of Pst isolates to resolve the population-genomic architecture of avirulence. We identified multiple avirulence-associated genomic regions and defined 69 candidate effectors, with 11 high-confidence ca

Optimizing hard cutoff quality control metrics for IBD segment inference and genotyping accuracy in low-coverage sequencing data.

Effective quality control (QC) is critical for reliable identity-by-descent (IBD) inference from low-pass whole-genome sequencing (LPWGS), where genotype uncertainty under low coverage can bias relatedness estimates. We conducted a comprehensive grid search to evaluate genotype posterior probability (GP) and Bayes factor (BF) thresholds across multiple coverage levels, assessing performance by false positive rate, false negative rate, and mean squared error (MSE) of total IBD sharing. At moderate-to-high coverage (> 0.10 ×), GP > 0.99 maintained stable performance and additional BF filtering p
