Development of a time-resolved fluoroimmunoassay for measuring plasma growth hormone in Nile tilapia (Oreochromis niloticus).

Growth hormone is a hormone secreted from the pituitary and is involved in the regulation of most major physiological processes such as grow … ith recombinant Gh and assembled them with a linker to form a single-chain variable fragment (scFv) gene pool. Using phage display technology, we isolated scFv DNA fragments from the pool, which encode a protein that specifically binds to tilapia Gh. We then established Eu-DTTA-based TRFIA for measuring plasma Gh in tilapia. The sensitivity of double antibo …

Soluble CD40L is associated with increased oxidative burst and neutrophil extracellular trap release in Behçet's disease.

BACKGROUND: Studies have suggested that soluble factors in plasma from patients with active (aBD) and inactive (iBD) Behçet's disease (BD) stimulate neutrophil function. Soluble CD40 ligand (sCD40L) is an important mediator of inflammation in BD. Its expression and effect on neutrophil oxidative burst and neutrophil extracellular trap (NET) release have not been characterized. In this study, we sought to investigate the role of plasma and the CD40L pathway on NET release and the oxidative burst profile in patients with aBD and iBD. METHODS: Neutrophils and peripheral blood mononuclear cells (

Proteoglycan sulfation in cartilage and cell cultures from patients with sulfate transporter chondrodysplasias: relationship to clinical severity and indications on the role of intracellular sulfate production.

Mutations in the diastrophic dysplasia sulfate transporter (DTDST) gene have been associated with a family of chondrodysplasias that includes diastrophic dysplasia (DTD), atelosteogenesis type 2 (AO2) and the lethal condition achondrogenesis type 1B (ACG1B). There is a correlation between the nature of the mutations and the clinical phenotype, but our understanding of the pathophysiology of the disorder, which involves defective sulfation of cartilage proteoglycans, is far from complete. To evaluate the degree of proteoglycan undersulfation in vivo, we have extracted chondroitin sulfate proteo

The IGF1-PGM5 Axis Inhibits Aerobic Glycolysis and Serves As a Tumor Suppressor in Nasopharyngeal Carcinoma.

Nasopharyngeal carcinoma (NPC) is a prevalent malignancy with a distinct geographical distribution. This study aimed to identify core glycolysis-related genes and investigate their functional roles in NPC pathogenesis. Integrated transcriptomic analysis was performed on multiple Gene Expression Omnibus datasets. Screening of core genes was performed computationally via machine learning algorithms on the pool of glycolysis-related differentially expressed genes (Gly-DEGs). The tumor-suppressive function of insulin-like growth factor 1 (IGF1) was demonstrated through a combination of in vitro as

Turnip mosaic virus alters phosphorus metabolism and shoot-root allocation without resource competition.

Plant viruses affect production through symptom induction in host plants. These symptoms could partially arise from nutrient deprivation: The resource competition hypothesis posits that massive viral replication deprives hosts of essential nutrients, yet direct evidence for phosphorus (P) competition is lacking. Moreover, it is reported that biotic stresses can lead to alterations on P metabolism. Using a hydroponic system enabling separate analysis of shoots and roots in adult Arabidopsis thaliana plants, we investigated whether Turnip mosaic virus (TuMV) drawed significant P internal pools l

Antioxidant Upregulation Contributes to Improvements in Diaphragm Function Following Cervical Spinal Cord Injury and Hyperbaric Oxygen Therapy.

Cervical spinal cord injury directly impacts the phrenic motor neuron pool, resulting in impaired ventilation and diaphragm muscle dysfunction. Previously, we demonstrated that hyperbaric oxygen (HBO) therapy, delivered during the acute phase following SCI, can attenuate diaphragm dysfunction. However, despite the demonstrated efficacy of HBO to improve muscle function, the molecular signaling pathways regulating this effect remain unclear. These experiments test the hypothesis that the protective effects of HBO therapy are associated with upregulation of the endogenous antioxidants SOD1, SOD2

Pregnane X receptor deletion induces a lean phenotype in chow-fed mice through a liver-ileum-muscle axis involving 6α-hydroxylated bile acids and Fgf15.

OBJECTIVES: The pregnane X receptor (Pxr; Nr1i2) plays a role in metabolic regulation. However, its contribution to systemic energy homeostasis under physiological conditions remains unclear. Here, we investigated the metabolic consequences of Nr1i2 deficiency in chow-fed mice. METHODS: Phenotyping was performed in adult whole-body Nr1i2 knockout (Nr1i2-/-) chow-fed mice and their wild-type littermates. Body composition, metabolic parameters, skeletal muscle mass and myofiber size were assessed. Ileal gene expression, circulating Fgf15 levels, and bile acid composition were analyzed. Intestin

Prime Editing of Alzheimer's Disease High-Risk APOE4 Allele by Brain-Directed Adeno-Associated Virus Vectors.

Common variants of the apolipoprotein E (APOE) gene have a major impact on the risk of developing Alzheimer's disease (AD). Relative to homozygotes with the common E3 allele, the APOE4 variant (C112R) increases risk by 3.5-fold in E3/E4 heterozygotes and 15-fold in E4 homozygotes. Since the E3 and E4 alleles differ only by a single nucleotide, gene editing of E4 to E3 is a potential strategy to reduce AD risk in E4 homozygotes. Because the APOE pool in the brain is separate from systemic APOE, editing to treat AD would ideally be directed to the brain. Following in vitro optimization of prime

Identification of genes expressed in the pectoral fins and skin of Takifugu rubripes to reveal candidate genes involved in host recognition by the crustacean ectoparasite Caligus fugu.

The initial detection of host-derived chemical cues is a key physiological event that strongly influences infection success in parasitic copepods such as Caligus fugu. Although ionotropic receptors were previously identified in C. fugu as potential host-sensing molecules, their ligands and specific roles in host recognition remain unknown. Since the infective copepodid I stage preferentially attaches to the fins of the puffer host, genes expressed at higher levels in fins may contribute to host recognition by C. fugu. In this study, differentially expressed genes between the pectoral fins and

Identification of differentially expressed mRNAs and miRNAs in spermatozoa of bulls of varying fertility.

Bulls used in artificial insemination, with apparently normal semen quality, can vary significantly in their field fertility. This study aimed to characterize the transcriptome of spermatozoa from high (HF) and low (LF) fertility bulls at the mRNA and miRNA level in order to identify potential novel markers of fertility. Holstein-Friesian bulls were assigned to either the HF or LF group (n = 10 per group) based on an adjusted national fertility index from a minimum of 500 inseminations. Total RNA was extracted from a pool of frozen-thawed spermatozoa from three different ejaculates per bull, f

Development of a molecular xenomonitoring protocol to assess filariasis transmission.

According to the World Health Organization, lymphatic filariasis (LF), a mosquito-borne neglected tropical disease (NTD), should be eliminated as a public health concern by the end of 2020. To this end, the goals of the Global Programme to Eliminate Lymphatic Filariasis (GPELF) include interrupting transmission through mass drug administration (MDA). After two decades, several countries have implemented MDA and are now ready to confirm whether transmission has been interrupted. The method for detecting the parasites in mosquito vectors known as xenomonitoring is a non-invasive tool for assessi

The molecular and cellular origin of human prostate cancer.

Prostate cancer is the most commonly diagnosed male malignancy. Despite compelling epidemiology, there are no definitive aetiological clues linking development to frequency. Pre-malignancies such as proliferative inflammatory atrophy (PIA) and prostatic intraepithelial neoplasia (PIN) yield insights into the initiating events of prostate cancer, as they supply a background "field" for further transformation. An inflammatory aetiology, linked to recurrent prostatitis, and heterologous signalling from reactive stroma and infiltrating immune cells may result in cytokine addiction of cancer cells,

Altered TFEB-mediated lysosomal biogenesis in Gaucher disease iPSC-derived neuronal cells.

Gaucher disease (GD) is caused by mutations in the GBA1 gene, which encodes the lysosomal enzyme glucocerebrosidase (GCase). The severe forms of GD are associated with neurodegeneration with either rapid (Type 2) or slow progression (Type 3). Although the neurodegenerative process in GD has been linked to lysosomal dysfunction, the mechanisms involved are largely unknown. To identify the lysosomal alterations in GD neurons and uncover the mechanisms involved, we used induced pluripotent stem cells (iPSCs) derived from patients with GD. In GD iPSC-derived neuronal cells (iPSC-NCs), GBA1 mutatio
