Genetic variants in cyclooxygenase-2: Expression and risk of gastric cancer and its precursors in a Chinese population.

BACKGROUND & AIMS: To screen the genetic variants in cyclooxygenase-2 (COX-2), and evaluate their effects on COX-2 expression and risk of gastric cancer (GC) and its precursors, a population-based study was conducted in Linqu County, Shangdong Province, an area of China, a high-risk area of GC. METHODS: Genotypes were determined by polymerase chain reaction (PCR)-based denaturing high-performance liquid chromatography analysis in 248 GC cases and 1523 subjects with precancerous gastric lesions. COX-2 expression was detected by immunohistochemical analysis of biopsy specimens of 593 subjects s

Genetic polymorphisms in the angiotensin II receptor gene and their association with open-angle glaucoma in a Japanese population.

PURPOSE: The local renin-angiotensin system (RAS) is present in the ciliary body and plays a role in regulating aqueous humor dynamics and thus intraocular pressure (IOP). The purpose of this study was to determine whether gene polymorphisms in the RAS increase the risk of development of glaucoma in the Japanese. METHODS: A case-control study was performed in 698 Japanese subjects: 190 patients with primary open-angle glaucoma (POAG), 268 patients with normal-tension glaucoma (NTG), and 240 normal subjects. Ten polymorphisms in seven genes-AGT/Thr174Met and AGT/Met235Thr; REN/I8-83G-->A; ACE/

Informed consent and biobanks: a population-based study of attitudes towards tissue donation for genetic research.

AIMS: The procurement and usage of tissue samples has begun receiving increasing legal and ethical attention. The authors' aim was to develop an empirically based understanding of public attitudes to the use of tissue for research. METHOD: A questionnaire was distributed to a randomized sample (n=1,000) of the population in the age group 18-85 in Västerbotten County, Sweden. RESULTS: The response rate was approximately 60%. There was a general acceptance of genetic research based on biobank material (71%) but, though it is often a prerequisite for tissue-based research, a majority (62%) woul

Aedes aegypti

Impact of genetic polymorphisms on the smoking-related risk of periodontal disease: the population-based study SHIP.

Periodontitis is a bacterial inflammatory disease leading to attachment loss with the consequence of tooth loss. There exists a multifactorial risk pattern including bacterial challenge, smoking, age, sex, diabetes, socio-economic and genetic factors. Smoking has the highest impact on the course of the disease modulated by all the other factors. Here, we report the relationship between smoking and the polymorphisms of genetic polymorphisms inflicted in the pathogenesis. In a randomly selected population-based study, 1083 subjects were typed for the polymorphisms of the IL-1 genotype, Fcgamma RI

Investigation of the genetic influence of the OPG, VDR (Fok1), and COLIA1 Sp1 polymorphisms on BMD in the Irish population.

Low bone mineral density (BMD) is a major risk factor for the development of osteoporosis and there is strong evidence to suggest that the procurement and preservation of peak BMD is genetically determined. In an effort to identify factors responsible for susceptibility to low BMD in the Irish population, we investigated its possible association with polymorphisms in the Osteoprotegerin (OPG) gene, Type I collagen alpha 1 (COLIA1) Sp1 binding site and vitamin D receptor (VDR) start codon. Following a systematic screening of the regulatory and coding regions of the OPG gene, we identified a nov

Trypanosoma cruzi: role of host genetic background in the differential tissue distribution of parasite clonal populations.

Chagas' disease, caused by the protozoan parasite Trypanosoma cruzi, has quite a variable clinical presentation, ranging from asymptomatic to severe chronic cardiac and/or gastrointestinal disease. The reason for that is not completely understood, but both parasite and host genetic traits are certainly involved. Recently, we have demonstrated clinically and experimentally that the genetic variability of T. cruzi is one of the determinants of the pattern of tissue involvement in Chagas' disease. We then decided to turn our attention to the role of host genetic background. To study this, we comp

Cardiovascular risk factors and Alzheimer's disease: a genetic association study in a population aged 85 or over.

Increasing evidence suggests a relation between vascular disorders and late-onset Alzheimer's disease (AD). We performed an association analysis of low-density lipoprotein receptor-related protein (LRP), lipoprotein lipase (LPL), and angiotensin converting enzyme (ACE) genes, known to be involved in vascular disorders, and AD. Genotyping was carried out in 113 patients with clinically defined Alzheimer's disease (NINCDS-ADRDA criteria) and 203 non-demented controls in a prospective, population-based study of people aged 85 years or over (Vantaa 85+ Study). Corresponding analysis was performed

Minisatellite marker analysis of Trypanosoma brucei: reconciliation of clonal, panmictic, and epidemic population genetic structures.

The African trypanosome, Trypanosoma brucei, has been shown to undergo genetic exchange in the laboratory, but controversy exists as to the role of genetic exchange in natural populations. Much of the analysis to date has been derived from isoenzyme or randomly amplified polymorphic DNA data with parasite material from a range of hosts and geographical locations. These markers fail to distinguish between the human infective (T. b. rhodesiense) and nonhuman infective (T. b. brucei) "subspecies" so that parasites derived from hosts other than humans potentially contain both subspecies. To overco

The incidence of different cystic fibrosis mutations in the Scottish population: effects on prenatal diagnosis and genetic counselling.

We present an analysis of the frequency of 16 different cystic fibrosis (CF) mutant alleles in the Scottish population. Each allele was detected in DNA amplified by the polymerase chain reaction (PCR) either directly on polyacrylamide gels, on agarose gels after restriction enzyme digestion, or by using allele specific oligonucleotides. Among 506 CF chromosomes, of predominantly Scottish origin, the frequencies of the different mutations were delta F508 0.71, G551D 0.05, G542X 0.04, R117H 0.01, 1717-1G----A 0.01, A455E + delta I507 + R553X + R560T + W1282X + 621 + 1G----T combined 0.03, unpubl

Genetic analysis of cover test measures and AC/A ratio in human populations with varying incidences of strabismus.

The genetic analyses of indices relating to distance alignment and accommodative convergence are presented. This approach is important in understanding the contribution of genetic factors to observed individual differences for these measures. Abnormalities of either of these two components results in a tendency towards either a convergent or divergent position of the eyes (relative to the position of fusional demand) and thus places an additional load or strain on the other components of the binocular mechanism which must compensate for this potential disruption. If compensation is inadequate

Oxidative balance score, genetic susceptibility, and the risk of pancreatic cancer: a population-based cohort study.

BACKGROUND: Pancreatic cancer (PC) is one of the deadliest cancers, largely due to its prolonged asymptomatic progression. Identifying and mitigating risk factors is critical for reducing PC incidence. Although oxidative balance score (OBS) is a potential modifiable factor for preventive interventions, its association with PC risk remains unclear. METHODS: The study included 157,527 participants from the UK Biobank, all of whom were free of PC at baseline. OBS was calculated using 16 diet-lifestyle components derived from a validated self-reported questionnaire. A higher OBS indicates a profi

Implications and ramifications of using the CDC tier 1 genetic screening concept in East Asian populations.

PURPOSE: The US Centers for Disease Control and Prevention defined the Centers for Disease Control and Prevention Tier 1 (CDCT1) genomics applications, advocating mass screening for 3 hereditary conditions with significant public health impacts. On this premise, we investigated how genome sequencing (GS) and ethnic differences affect genetic screening in East Asia. Additionally, we incorporated 2 specific genes, NOTCH3 (HGNC:7883) and GJB2 (HGNC: 4284), linked to cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy (MIM: 125310) and hereditary hearing loss

Analysis of Inbreeding, Population Structure, and Genetic Diversity in the Kumamoto Sub-Breed of Japanese Brown Cattle.

Introduction: The Kumamoto sub-breed of Japanese Brown cattle is a small population facing increasing inbreeding and declining effective population size driven by the intensive use of a limited number of elite sires. Methods: We analyzed 811 Japanese Brown cows genotyped using a 30K SNP array, retaining 19,745 SNPs after quality control. We calculated and compared ten genomic inbreeding estimators, including SNP-by-SNP and segment-based measures. Population structure was first assessed by rearing region using principal component analysis (PCA) and was then evaluated using ADMIXTURE-based clust

Genetic associations with alcohol drinking initiation and cessation in sub-Saharan African and African ancestry populations.

Alcohol misuse is a major global health concern with significant genetic contributions, yet studies in African populations remain limited. We conducted genome-wide association meta-analyses for alcohol drinking initiation and cessation across multiple cohorts comprising individuals from sub-Saharan Africa (N = 20,936) and those with African ancestry from UK Biobank (N = 7,726). Our analyses identified two genome-wide significant loci associated with drinking cessation ( FYB2 and FTO regions) and several suggestive associations overlapping with genes implicated in alcohol metabolism, behavioral

Genetic diversity and population structure of Coffea arabica in Saudi Arabia revealed by whole-genome resequencing.

Gene banks are essential for conserving plant genetic resources, yet limited genomic characterization constrains their effective use in breeding. Coffea arabica L., an economically important crop in Saudi Arabia, remains poorly characterized at the genome-wide level. We performed whole-genome resequencing of 78 Saudi C. arabica accessions conserved at the Ministry of Environment, Water and Agriculture (MEWA) Seed Center and PGR Bank, generating 86 sequencing libraries, including eight technical replicates. Reads were aligned to the Cara_r6 reference genome, yielding 468, 133 variants (412, 043

Functional characterization of genetic variants associated with rare inherited renal disorders in the Sri Lankan population.

Rare inherited renal disorders (RIRD) are genetically heterogeneous conditions that contribute substantially to morbidity and mortality, particularly among pediatric and young adult populations. Although next-generation sequencing has improved diagnostic yield, functional interpretation of novel variants and variants of uncertain significance remains limited, especially in underrepresented populations such as Sri Lankans. This study aimed to functionally characterize genetic variants identified through whole-exome sequencing (WES) in Sri Lankan patients with clinically diagnosed RIRD. In RT-qP

The contribution of sexual selection and genetic constraints to phenotypic divergence among natural populations of Drosophila subquinaria.

Evolutionary trajectories are determined by selection and the underlying genetic architecture of additive genetic variances and covariances for the traits. To better understand divergence in multivariate phenotypes among natural populations, we need population-specific estimates of selection and genetic architecture of the traits involved; such extensive data are challenging to acquire and few examples exist. Here we do this using males from six populations of the mushroom-feeding fly Drosophila subquinaria. We take advantage of a naturally occurring pattern of reproductive character displacem

Genetic and clinical investigation of insulin-degrading enzyme in Parkinson's disease within the Chinese Han population.

INTRODUCTION: Growing evidence suggests a mechanistic link between type 2 diabetes mellitus and Parkinson's disease (PD), with insulin-degrading enzyme (IDE) implicated in both insulin and amyloid-β metabolism, as well as α-synuclein degradation. However, the role of IDE in PD pathogenesis remains insufficiently defined. This study aimed to investigate the association of IDE gene polymorphisms and serum IDE levels with sporadic PD in a Chinese Han population. METHODS: Fourteen single nucleotide polymorphisms (SNPs) within the IDE gene were genotyped in 463 patients with sporadic PD and 576 ag
