Advances in HBV infection and replication systems in vitro.

BACKGROUND: Hepatitis B virus (HBV) is a DNA virus belonging to the Hepadnaviridae family that has limited tissue and species specificity. Due to the persistence of HBV covalently closed circular DNA (cccDNA) in host cells after HBV infection, current antiviral drugs cannot eradicate HBV. Therefore, the development of an active cell culture system supporting HBV infection has become the key to studying HBV and developing effective therapeutic drugs. MAIN BODY: This review summarizes the significant research achievements in HBV cell culture systems in vitro, including embryonic hepatocytes and

Targeting Human-Cytomegalovirus-Infected Cells by Redirecting T Cells Using an Anti-CD3/Anti-Glycoprotein B Bispecific Antibody.

The host immune response to human cytomegalovirus (HCMV) is effective against HCMV reactivation from latency, though not sufficient to clear the virus. T cells are primarily responsible for the control of viral reactivation. When the host immune system is compromised, as in transplant recipients with immunosuppression, HCMV reactivation and progressive infection can cause serious morbidity and mortality. Adoptive T cell therapy is effective for the control of HCMV infection in transplant recipients. However, it is a highly personalized therapeutic regimen and is difficult to implement in routi

Evaluation of host Hsp(s) as potential biomarkers for the diagnosis of tuberculous meningitis.

OBJECTIVES: Diagnosis of tuberculosis meningitis (TBM) remains challenging in tuberculosis (TB) endemic countries. The need for TB biomarkers arises, in part, from the difficulty of accurately diagnosing TBM with the available methods. PATIENTS AND METHODS: To explore the potential of host Hsps (Hsp 25, Hsp 60, Hsp 70 and Hsp 90) as an alternative marker in TBM diagnosis, we evaluated cerebrospinal fluid (CSF) sample of TBM (n=49), Pyogenic Meningitis (PM) (n=20), Viral Meningitis (VM) (n=09), Fungal Meningitis (FM) (n=04) and non infectious control (n=79) patients using indirect ELISA. The immune response to the parasitic nematode Trichinella and the ways to escape it. From experimental studies to implications for human infection.

The review describes different aspects of the host immune response to Trichinella, not only at the intestinal level on which most of the studies have focused until now, but also in the muscles which represent the final target of host invasion. The role of antibodies, T cells, mast cells, eosinophils and neutrophils, respectively, in immune reaction to this nematode is considered, in the light of the recent data derived from experimental models, both "in vivo" and more recently "in vitro" and when available, from clinical observations. A section is also devoted to the principal escape mechanism

Role of distinct type-IV-secretion systems and secreted effector sets in host adaptation by pathogenic Bartonella species.

The α-proteobacterial genus Bartonella comprises a large number of facultative intracellular pathogens that share a common lifestyle hallmarked by hemotrophic infection and arthropod transmission. Speciation in the four deep-branching lineages (L1-L4) occurred by host adaptation facilitating the establishment of long lasting bacteraemia in specific mammalian reservoir host(s). Two distinct type-IV-secretion systems (T4SSs) acquired horizontally by different Bartonella lineages mediate essential host interactions during infection and represent key innovations for host adaptation. The Trw-T4SS c

Phylogenomic Diversity Elucidates Mechanistic Insights into Lyme Borreliae-Host Association.

Host association-the selective adaptation of pathogens to specific host species-evolves through constant interactions between host and pathogens, leaving a lot yet to be discovered on immunological mechanisms and genomic determinants. The causative agents of Lyme disease (LD) are spirochete bacteria composed of multiple species of the Borrelia burgdorferi sensu lato complex, including B. burgdorferi (Bb), the main LD pathogen in North America-a useful model for the study of mechanisms underlying host-pathogen association. Host adaptation requires pathogens' ability to evade host immune respons

Experimentally induced cutaneous leishmaniasis: are L3T4+ T cells that promote parasite growth distinct from those mediating resistance?

A considerable body of evidence from various laboratories indicates that specific T cell responses generated during infection with Leishmania parasites play an important role both in the resolution and progression of cutaneous leishmaniasis. Recent data, summarized in this article, indicate that resolution of lesions and promotion of disease not only result from the activity of functionally distinct parasite-specific L3T4+ T cells but could also be mediated by functionally similar L3T4+ T cells differing only in their fine antigenic specificity. This contention is based on observations which s

The macrophage polarization-ferroptosis axis as a therapeutically targetable immunometabolic framework in periodontitis.

Periodontitis is a chronic inflammatory disease sustained by microbial dysbiosis, maladaptive host immunity, oxidative stress, and progressive destruction of periodontal supporting tissues. Macrophage polarization and ferroptosis have usually been discussed as separate pathological processes, but increasing evidence suggests that they may be functionally connected within the periodontal inflammatory microenvironment. This review evaluates the macrophage polarization-ferroptosis axis as an emerging and therapeutically targetable immunometabolic framework in periodontitis. We summarize how pro-i

Conserved ancillary residues situated proximally to the VIM-2 active-site affect its metallo β-lactamase activity.

Verona-integron-metallo-β-lactamase (VIM-2) is one of the most widespread class B β-lactamase responsible for β-lactam resistance. Although active-site residues help in metal binding, the residues nearing the active-site possess functional importance. Here, to decipher the role of such residues in the activity and stability of VIM-2, the residues E146, D182, N210, S207, and D213 were selected through in-silico analyses and substituted with alanine using site-directed mutagenesis. The effects of substitution mutations were assessed by comparing the changes in β-lactam susceptibility pattern of

Bioinformatics approach to understand the mode of microbial pathogenesis of Chlamydia trachomatis and their implications in gynecologic malignancy.

Chlamydia trachomatis has a say on the target gene i.e., modulating the expression of target gene in the host so that it is given protection from the immune cells and so its survival and replication are not arrested by the host. The current study reports a wide range of C. trachomatis proteins that target the cellular as well as sub-cellular components of the host in gynecologic malignancy. Various bioinformatics tools was used to conduct an in-depth analysis on nuclear and eukaryotic sub cellular localization signal to find the sequences of the predicted proteins of C. trachomatis strain G. The virus infecting the parasitoid Leptopilina boulardi exerts a specific action on superparasitism behaviour.

Parasites often induce behavioural changes in their host. However, it is not necessarily easy to determine whether these changes are representative of an adaptation of the parasite (parasite manipulation), an adaptive response of the host or a side-effect of infection. In a solitary parasitoid of Drosophila larvae (Leptopilina boulardi), viral particles (LbFV) modify the host acceptance behaviour of infected females by increasing their tendency to superparasitize. This behavioural alteration allows for the horizontal transmission of the virus within superparasitized Drosophila larvae. To add s

Secreted neutral metalloproteases of Bacillus anthracis as candidate pathogenic factors.

To evaluate the pathogenic potential of Bacillus anthracis-secreted proteases distinct from lethal toxin, two neutral zinc metalloproteases were purified to apparent homogeneity from the culture supernatant of a non-virulent delta Ames strain (pXO1-, pXO2-). The first (designated Npr599) is a thermolysin-like enzyme highly homologous to bacillolysins from other Bacillus species. The second (designated InhA) is a homolog of the Bacillus thuringiensis immune inhibitor A. These proteases belong to the M4 and M6 families, respectively. Both enzymes digested various substrates, including extracellu

Nonspecific patterns of vector, host and avian malaria parasite associations in a central African rainforest.

Malaria parasites use vertebrate hosts for asexual multiplication and Culicidae mosquitoes for sexual and asexual development, yet the literature on avian malaria remains biased towards examining the asexual stages of the life cycle in birds. To fully understand parasite evolution and mechanism of malaria transmission, knowledge of all three components of the vector-host-parasite system is essential. Little is known about avian parasite-vector associations in African rainforests where numerous species of birds are infected with avian haemosporidians of the genera Plasmodium and Haemoproteus. H

Variation within and among host species in engorgement of larval trombiculid mites.

We recovered larval trombiculid mites (i.e. chiggers), vectors of scrub typhus, from small mammal hosts in Taiwan, and compared the relative degree of engorgement (RDE) of the dominant chigger (Leptotrombidium imphalum) from different hosts. Naturally occurring chiggers recovered from Rattus losea and Bandicota indica were 1·4x and 1·3x as engorged as those from Apodemus agrarius. Within each host species, RDE was negatively related to chigger loads, but was mostly unrelated to gender or to body or reproductive condition of hosts. We documented significant variation in chigger engorgement both

Live attenuated tetravalent dengue virus host range vaccine is immunogenic in African green monkeys following a single vaccination.

UNLABELLED: The causative agent of dengue fever, dengue virus (DENV), is transmitted by mosquitoes, and as distribution of these insects has expanded, so has dengue-related disease. DENV is a member of the Flaviviridae family and has 4 distinct serotypes (DENV-1, -2, -3, and -4). No lasting cross protection is afforded to heterologous serotypes following infection by any one of the individual serotypes. The presence of nonneutralizing antibodies to one serotype can facilitate the occurrence of more-severe dengue hemorrhagic fever through immune enhancement upon infection with a second serotype

Migration of some roundworm species in experimentally infected white mice.

The migration of four roundworm species (Ascaris suum, Toxocara canis, T. cati and Toxascaris leonina) was studied in various organs of experimentally infected white mice. The hatching of T. leonina and T. cati larvae in the paratenic host is more rapid (24-36 h) than the hatching of A. suum and T. canis larvae (1-6 days). The deposition of larvae in host organs is different in the individual species. Maximum number of larvae in the liver: T. leonina and A. suum on day 4, T. canis on day 2 and T. cati on day 1. Maximum number of larvae in the lungs: A. suum and T. leonina on day 7, T. cati on

Insights into goatpox virus and sheeppox virus genomes from pangenome graphs.

The capripoxviruses comprise three species: goatpox virus (GTPV), sheeppox virus (SPPV) and lumpy skin disease virus (LSDV). They are large … lighted loci under strong constraint and lineage-specific structural changes that may contribute to host specificity. Together, these results demonstrated how graph-based genome models complement gene-based analyses in resolving poxvirus genome evolution and provide a resource for improved comparative and population genomic studies of large DNA viruses.

A Highly Protective Live-Attenuated Vaccine Generated by Targeted Deletion of the Mycobacterium bovis Virulence Factor VapC40.

Type II toxin-antitoxin (TA) systems are significantly expanded in the Mycobacterium tuberculosis complex; however, the functional role of the VapBC40 system in Mycobacterium bovis (M. bovis) pathogenesis remains poorly characterized. This study aimed to investigate the role of VapBC40 in mycobacterial virulence and evaluate its potential as a target for rational vaccine attenuation. We performed evolutionary analysis and yeast two-hybrid assays to characterize VapBC40 system specificity, conducted in vitro macrophage infection models and in vivo murine studies to assess virulence contribution
