Histoplasma capsulatum and Caenorhabditis elegans: a simple nematode model for an innate immune response to fungal infection.

Histoplasma capsulatum is a primary fungal pathogen of mammals responsible for histoplasmosis. During pathogenesis H. capsulatum yeast proliferate in phagosomes of macrophages. This extensive host/pathogen interaction involves a complex cascade of responses in both organisms. In the mammalian host, infection results in complex branched immunity that is initiated with an innate response and later induces an adaptive response but each response is difficult to resolve during fungal infection. Therefore, in an effort to identify less complex systems and to gain understanding of the host innate res

When Shigella tells the cell to hang on.

OspE, a Shigella type III effector binds to integrin-like kinase and enhances cell adhesion to better disseminate and colonize the intestinal epithelium. Because of the existence of OspE orthologues in other enteropathogens such as enteropathogenic Escherichia coli or Salmonella sp., maintenance of cell adhesion appears as a widespread strategy for bacteria that interact with the intestinal epithelium.

The genetics and cell biology of Wolbachia-host interactions.

Wolbachia are gram-negative bacteria that are widespread in nature, carried by the majority of insect species as well as some mites, crustaceans, and filarial nematodes. Wolbachia can range from parasitic to symbiotic, depending upon the interaction with the host species. The success of Wolbachia is attributed to efficient maternal transmission and manipulations of host reproduction that favor infected females, such as sperm-egg cytoplasmic incompatibility (CI). Much remains unknown about the mechanistic basis for Wolbachia-host interactions. Here we summarize the current understanding of Wolb

Human coagulation factor X-adenovirus type 5 complexes poorly stimulate an innate immune response in human mononuclear phagocytes.

UNLABELLED: One of the first lines of host defense against many viruses in vertebrates is the innate immune system, which detects pathogen-a … acrophages in vitro and in vivo. Given the importance of studies using animals to better understand host-pathogen interactions, we asked if human FX (hFX)-armored HAd type 5 (HAd5) was capable of activating innate immune sensors in primary human mononuclear phagocytes. To this end, we assayed human mononuclear phagocytes for their ability to be stimulated by hFX-armored HAd …

High-Resolution Detection of Chlamydia Proteins by Expansion Microscopy.

The pathogenic bacterium Chlamydia replicates via an unusual developmental cycle that takes place within a membrane-bound inclusion inside an infected host cell. Immunofluorescence microscopy has revealed many details about the two specialized developmental forms and the recruitment of host organelles to the inclusion, but lacks the high resolution of electron microscopy (EM). In this chapter, we describe the use of an innovative microscopy method, called Tenfold Robust Expansion (TREx) microscopy, to visualize proteins in Chlamydia-infected cells at nanoscale resolution. We present a detailed

Viral deISGylation holds the key to the metabolic kingdom.

The interferon-induced modification of cellular and viral proteins by ISG15 inhibits viral infection and is reversed by virus-encoded deISGylases, but the broad physiological impacts of this host-pathogen clash remain unclear. In this issue of Immunity, Zhu et al. reveal that ISG15 modification broadly restricts key metabolic pathways that deISGylation relieves to promote viral replication.

From gatekeeper to target: MAPK cascades as control circuits at the insect-microbe interface.

Mitogen-activated protein kinase (MAPK) signaling cascades are conserved modules that orchestrate multiple processes, including immunity, stress adaptation, and tissue homeostasis. In insects, MAPK pathways function as central regulatory circuits, coordinating defense responses and resistance mechanisms against diverse pathogens. MAPK cascades couple damage sensing in the gut epithelium to intestinal regeneration, pathogen/microbiota stability, cellular and humoral immune responses, and immune homeostasis. Consequently, a wide range of pathogens (parasites, viruses, and bacteria) target, suppr

Development and characterization of chicken lung organoids for in vitro modeling of avian influenza virus-host cell interaction.

High pathogenicity avian influenza viruses pose a growing threat to poultry, livestock, wildlife, and humans as they undergo accelerated expansion of host and geographical ranges. Since 2020, these viruses have driven a panzootic characterized by extensive viral diversification and spillover into species previously considered to be resistant. There is currently a lack of physiologically relevant in vitro models that can be used to screen the rapidly changing viral landscape. To address this need, we describe the first chicken lung organoids derived from adult stem cells of specific pathogen fr

Herpesviruses and autophagy: An intracellular conflict.

Herpesviruses are a class of viruses capable of causing various diseases in humans and animals. These pathogens have the ability to remain latent within the host cell for extended periods, often remaining dormant until immune function is compromised. Extensive research indicates that autophagy, as a key intracellular defense mechanism, is closely associated with the herpesvirus life cycle. Following cellular invasion, herpesviruses evade immunity and sustain infection by modulating autophagy formation, blocking autophagy-lysosome fusion, or selectively degrading key immune molecules via autoph

[Research Progress on Quorum Sensing-Regulated Oral Microbial Interaction Networks in Periodontitis].

Quorum sensing (QS) is a mechanism by which bacteria communicate and coordinate group behaviors through the secretion of signaling molecules, playing a critical role in oral microbial interactions and the pathogenesis of periodontitis. This review summarizes recent advances in QS-mediated regulation of oral microbial interaction networks in periodontitis, focusing on the molecular mechanisms that modulate biofilm formation, virulence factor expression, and pathogen community dynamics. In addition, novel QS-targeted therapeutic strategies are discussed. Although periodontitis is fundamentally a

Host mitochondrial DNA in bacterial pathogenesis: Friend, Foe, or Both?

Mitochondria, traditionally recognized for their energy-producing function, also play a vital role in immune responses by regulating host–bacteria interactions. This narrative review examines emerging evidence on how host mitochondrial DNA (mtDNA) influences bacterial infections, emphasizing its active involvement in immune defense and disease progression. We first discuss how mitochondrial dynamics and stress responses support infection control, followed by an exploration of mtDNA as an intrinsic danger signal. The review highlights key immune pathways and inflammatory mechanisms triggered by

Bacteria-phage coevolution drives variation in bacterial wilt disease incidence via resistance-virulence trade-offs.

Bacteria-phage coevolution often results in correlated fitness effects on partner species. Whether coevolutionary changes impact the ecology of the surrounding communities is unclear. Here we link coevolution between the phytopathogenic bacterium Ralstonia pseudosolanacearum and its phage parasites to bacterial wilt disease patterns across four geographically disconnected tomato fields. We find that bacteria and phages are locally adapted between and within fields. Phage infectivity was highest on sympatric bacteria, and bacteria showed greater phage resistance when isolated from healthy than

The rhinovirus puzzle: Linking virus and host factors to childhood asthma.

Rhinoviruses (RVs) are the most common respiratory pathogens and a significant cause of asthma exacerbations and wheezing illnesses in children. In addition, RV wheezing illnesses during the preschool years strongly predict asthma development. This review includes updates on RV epidemiology and virus diversity, highlighting species-specific differences and antigenic variability that present significant challenges for vaccine design. Host responses to RV involve canonical antiviral responses originating in airway epithelial cells, followed by activation of adaptive immunity. In patients with as

Gut microbiota and the immune system: A regulatory axis through host MicroRNA modulation.

The human gastrointestinal tract harbors trillions of microorganisms that collectively exert profound influences on host physiology, particularly immune function. Emerging evidence reveals that gut microbiota modulates immune responses through intricate interactions with host microRNAs (miRNAs), small non-coding RNAs that post-transcriptionally regulate gene expression. This review synthesizes current understanding of how gut bacteria influence host immunity via miRNA-dependent mechanisms. We examine two primary pathways: first, the capacity of gut microbes to regulate host miRNA expression pr
