Cutaneous melanoma is the most lethal skin cancer, and risk is strongly shaped by pigmentary phenotype. The red-hair color phenotype, marked by red hair, fair skin, freckling, and poor tanning, carries elevated melanoma risk driven largely by loss-of-function variants in the melanocortin 1 receptor gene (MC1R). This risk is not explained by reduced ultraviolet protection alone. Impaired MC1R signaling shifts melanogenesis from photoprotective eumelanin toward pheomelanin, a pigment associated with oxidative stress and partly ultraviolet-independent melanomagenesis. MC1R may also influence mela

BACKGROUND: Polyploidy is frequently observed in cancer cells and is closely associated with chromosomal instability, which can lead to cancer progression. Polyploid cancers are more aggressive than diploid cancers, and polyploidy has been shown to be a prognostic marker for hepatocellular carcinoma (HCC). However, polyploidy is challenging to diagnose. Currently, no clinically implementable methods are available for diagnosing polyploidy in cancer.
METHODS: We established a method for assessing polyploidization in HCC using deep-learning-based artificial intelligence image recognition models

BACKGROUND AND PURPOSE: Accurate identification of lung cancer subtypes in medical images is of great significance for the diagnosis and treatment of lung cancer. Despite substantial progress in existing methods, they remain challenging due to limited annotated datasets, large intra-class differences, and high inter-class similarities.
METHODS: To address these challenges, we propose a Frequency Domain Transformer Model (FDTrans) to identify patients' lung cancer subtypes using the TCGA lung cancer dataset. We add a pre-processing process to transfer histopathological images to the frequency

Microsatellites are small, repetitive sequences found all across the human genome. Microsatellite instability is the phenomenon of variations in the length of microsatellites induced by the insertion or deletion of repeat units in tumor tissue (MSI). MSI-type stomach malignancy has distinct genetic phenotypes and clinic pathological characteristics, and the stability of microsatellites influences whether or not patients with gastric mesothelioma react to immunotherapy. As a result, determining MSI status prior to surgery is critical for developing treatment options for individuals with gastric

BACKGROUND: Dentinogenesis imperfecta (DI) is a rare debilitating hereditary disorder affecting dentin formation and causing loss of the overlying enamel. Clinically, DI sufferers have a discolored and weakened dentition with an increased risk of fracture. The aims of this study were to assess genotype-phenotype findings in three families with DI-II with special reference to mutations in the DSPP gene and clinical, histological, and imaging manifestations.
METHODS: Nine patients participated in the study (two from family A, four from family B, and three from family C). Buccal swab samples wer

The experience in free radical biology and medicine shows the crucial role of redox signalling in carcinogenesis. The cells and tissues of healthy mammals are characterised by a low level of reactive oxygen species (ROS) and some constant (reference) level of reducing equivalents. Increasing of ROS above the critical level provokes genomic instability. The present study describes universal methodology for direct imaging of tissue redox activity in carcinogenesis, which allows a differentiation of cancer development from normal condition. The experiments were conducted on: neuroblastoma-bearing

Distal myopathies (DM) are clinically and genetically heterogeneous neuromuscular disorders, and identifying a molecular genetic cause may remain challenging in a subset of cases. Moreover, DM may be misdiagnosed as hereditary neuropathies due to overlapping clinical features. Here, we report a novel structural variant in FLNC associated with DM identified through genome sequencing (GS). Two affected relatives initially presented independently with referral diagnoses of Charcot-Marie-Tooth disease and amyotrophic lateral sclerosis. Clinical re-evaluation led to a change of the diagnosis to DM.

The impact of sex hormones on the brain has been widely recognized. Polycystic ovary syndrome (PCOS) is frequently accompanied by abnormal hormone. Furthermore, psychiatric disorders are among the most common comorbidities associated with PCOS. However, whether PCOS induces alterations in brain structure remains unclear. Mendelian randomization (MR) was conducted to determine this causal association and pinpoint specific brain regions prone to PCOS. Genome wide association study (GWAS) data from the FinnGen database, for PCOS phenotypes from 39,004 PCOS patients and 731,830 controls were inclu

BACKGROUND: Previous observational studies have reported a possible association between major depressive disorder (MDD) and abnormal cortical structure. However, it is unclear whether MDD causes reductions in global cortical thickness (CT) and global area (SA).
OBJECTIVE: We aimed to test the bidirectional causal relationship between MDD and CT and SA using a Mendelian randomization (MR) design and performed exploratory analyses of MDD on CT and SA in different brain regions.
METHODS: Summary-level data were obtained from two GWAS meta-analysis studies: one screening for single nucleotide po

Human organ structure and function are important endophenotypes for clinical outcomes. Genome-wide association studies (GWAS) have identified numerous common variants associated with phenotypes derived from magnetic resonance imaging (MRI) of the brain and body. However, the role of rare protein-coding variations affecting organ size and function is largely unknown. Here we present an exome-wide association study that evaluates 596 multi-organ MRI traits across over 50,000 individuals from the UK Biobank. We identified 107 variant-level associations and 224 gene-based burden associations (67 u

The neuronal ceroid lipofuscinoses (NCLs) are a group of inherited neurodegenerative disorders characterized by progressive declines in neurological functions following normal development. The NCLs are distinguished from similar disorders by the accumulation of autofluorescent lysosomal storage bodies in neurons and many other cell types, and are classified as lysosomal storage diseases. At least 13 genes contain pathogenic sequence variants that underlie different forms of NCL. Naturally occurring canine NCLs can serve as models to develop better understanding of the disease pathologies and f

Phelan-McDermid syndrome, also known as the 22q13 deletion syndrome, is a chromosomal microdeletion syndrome characterized by neonatal hypotonia, normal growth, profound developmental delay, absent or delayed speech, and minor dysmorphic features. Almost all of the 22q13 deletions published so far have been described as terminal. It is believed that the SHANK3 gene is the major candidate gene for the neurologic features of the syndrome. Here we describe a patient with a 0.72-Mb interstitial 22q13.2 deletion, intellectual disability, autistic behavior, epilepsy, mild dysmorphic features, and no

Surgical management of pheochromocytoma has evolved from a high-risk, open procedure to a nuanced, minimally invasive practice guided by multidisciplinary care. This review synthesizes contemporary advances in diagnostics, genetics, perioperative medicine, and surgical techniques. Definitive treatment remains complete resection, with minimally invasive adrenalectomy (transabdominal or retroperitoneal, laparoscopic, or robotic) now the standard for localized tumors, offering comparable oncologic outcomes when selected based on tumor size, anatomy, and expertise. Genomic characterization is inte

Pulmonary tumor thrombotic microangiopathy (PTTM) is a rapidly progressive cancer-related disease with a dismal clinical course. The patient in this report was a 43-year-old man with metastatic salivary duct carcinoma arising from the parotid gland. Combined androgen blockade therapy was administered started as first-line treatment, but failed after 5 months, followed by docetaxel plus carboplatin therapy as second-line treatment, which failed after 3 months. Genomic profiling revealed a BRAF V600E mutation, and combined BRAF and MEK inhibitor therapy was started as third-line treatment. The c

Viruses are genetically and structurally diverse, and outnumber cells by orders of magnitude. They can cause acute and chronic infections, suppress, or exacerbate immunity, or dysregulate survival and growth of cells. To identify chemical agents with pro- or antiviral effects we conducted arrayed high-content image-based multi-cycle infection screens of 1,280 mainly FDA-approved compounds with three human viruses, rhinovirus (RV), influenza A virus (IAV), and herpes simplex virus (HSV) differing in genome organization, composition, presence of an envelope, and tropism. We report the case of a 22-year-old woman presenting with an acute onset of dizziness, gait dysbalance and blurred vision. Magnetic resonance imaging included 3 Tesla and 7 Tesla imaging and revealed a T2-hyperintense, T1-hypointense, non-contrast-enhancing lesion strictly confined to the white matter affecting the right optic radiation. An extensive ophthalmologic examination yielded mild quadrantanopia but no signs of optic neuropathy. The lesion was biopsied. The neuropathological evaluation revealed a demyelinating lesion with marked tissue vacuolization and granular myelin disintegration
