Objective primary mediastinal B-cell lymphoma (PMBCL) is a distinct, aggressive lymphoma predominantly affecting young adults. While dose-intensive regimens are often recommended, their associated long-term toxicities and the necessity of consolidative radiotherapy remain major concerns. Given that PMBCL was explicitly excluded from the landmark POLARIX trial, we sought to evaluate the real-world efficacy and safety of first-line Pola-R-CHP in this specific population. Methods this single-center study analyzed 16 consecutive patients with newly diagnosed PMBCL treated with six cycles of Pola-R

Antibody-drug conjugates (ADCs) require antibodies with both high specificity and efficient internalization, features often overlooked by conventional discovery pipelines that rely on preselected antigens and recombinant proteins. Here, we describe an integrated phenotypic platform that combines target-unbiased live-cell biopanning with in situ chemical cross-linking and mass spectrometry to concurrently identify internalizing antibodies and their membrane-bound cognate antigens in a native cellular context. Using this approach, we identified 2E7, an antibody with rapid internalization and spe

Urothelial carcinoma is a common malignancy of the urinary tract and occurs predominantly in elderly men. Although surgery, chemotherapy, and radiotherapy remain the main treatment options, their efficacy in advanced disease is often limited, particularly in elderly patients with comorbidities or poor surgical tolerance. Antibody-drug conjugates (ADCs) combined with immune checkpoint inhibitors have recently emerged as a promising therapeutic strategy. We report the case of a 79-year-old man with advanced urothelial carcinoma who declined radical cystectomy and received disitamab vedotin plus

Immune checkpoint B7-H3 is an emerging target for immunotherapy. DS-7300a is an advanced B7-H3-targeting antibody-drug conjugate (ADC) warheaded with the topoisomerase I inhibitor DXd. DS-7300a has demonstrated clinical activity, but molecular biomarkers to predict its therapeutic response remain elusive. TP53 is one of the most mutated tumor suppressor genes across cancers, and effective therapies are urgently needed for TP53-deficient cancers. Using prostate cancer (PCa) as a model system, we reported that DS-7300a's anti-tumor efficacy is highly dependent on functional p53 in cancer cells,

The purpose of this investigation was to formulate and evaluate a chemically optimized [225Ac]Ac-labeled radioimmunoconjugate targeting the insulin-like growth factor-2 receptor (IGF2R) for targeted alpha therapy of osteosarcoma, a disease with unchanging survival rates for over 40 years. The anti-IGF2R antibody IF3 was thiol-selectively conjugated with Macropa-PEG4-maleimide and subsequently radiolabeled with 225Ac by incubation at 37 °C for 60 min. Radiochemical purity, serum stability, receptor binding kinetics, and internalization were evaluated in vitro. Biodistribution, dosimetry (MIRD s

Patients with myelodysplastic syndrome (MDS)/acute myeloid leukemia (AML) with high-risk features including TP53 mutations have poor outcomes because of the lack of effective therapies. The atypical chemokine surface receptor C-C motif chemokine receptor-like 2 (CCRL2) is overexpressed in MDS and secondary AML compared with healthy hematopoietic cells, and we recently found that TP53-mutated MDS/AML and AML with erythroid features express the highest levels of this receptor across MDS/AML subtypes. To illustrate the therapeutic potential of CCRL2 as a therapeutic target, we developed an anti-C

The DEBBRAH study showed promising results for patients with HER2-expressing (including HER2-low) metastatic breast cancer who received the antibody-drug conjugate (ADC) trastuzumab deruxtecan for leptomeningeal disease. ADCs represent a novel tool in the precision biomarker-based armamentarium, including in the tumor-agnostic setting, for targeting highly cytotoxic chemotherapy into cancer cells.

The treatment of breast cancer remains a significant clinical challenge, with limited responsiveness to conventional therapies. Photodynamic therapy, using the natural photosensitizer Hypericin, offers a promising modality, but its efficacy is limited by poor solubility, poor selectivity, and non-specific binding. To address these limitations, we developed a targeted nanoplatform leveraging the biocompatibility of gold nanoparticles and targeting capabilities of a monoclonal antibody. This study aimed to synthesize and characterize a novel nanoplatform comprising eco-friendly, green-synthesize

Antibody-photosensitizer conjugates (APCs) have emerged as promising platforms for targeted photodynamic therapy (PDT); however, most conventional APCs employ non-cleavable linkers and remain constitutively photoactive, generating fluorescence and phototoxicity irrespective of their biological location. This "always-on" behavior compromises imaging specificity and increases the risk of off-target phototoxicity. Here, we report an enzyme-responsive, cleavage-activatable cetuximab-photosensitizer conjugate (Q-Cet) that enables tumor-selective photodynamic activation and tumor-restricted immunoge

PURPOSE: Folate receptor α (FRα) is expressed in most ovarian cancers. However, only patients with high expression levels are eligible for Elahere, an FRα-targeted microtubule inhibitor antibody-drug conjugate (ADC). Efficacy limitations and safety concerns underscore the need to develop next-generation FRα-targeted ADC to treat tumors expressing variable levels of FRα and incorporate different payloads to reduce safety risks. Herein, we present the characterization of AZD5335, a novel FRα-targeted topoisomerase-1 inhibitor ADC.
EXPERIMENTAL DESIGN: The efficacy of AZD5335 was assessed and co

BACKGROUND: Diffuse large B-cell lymphoma (DLBCL) is the most common and aggressive subtype of non-Hodgkin lymphoma, with R-CHOP as the standard first-line treatment. However, many patients experience relapse or refractory disease, prompting the need for new therapeutic approaches. Recent advances, including chimeric antigen receptor (CAR) T-cell therapies, antibody-drug conjugates (ADCs), bispecific antibodies (bsAbs), immunomodulators, and Exportin-1 (XPO-1) inhibitors have transformed treatment strategies.
METHODS: A comprehensive literature review was conducted using PubMed up to January

INTRODUCTION: The DESTINY-B01 trial led to trastuzumab deruxtecan (T-DXd) approval for human epidermal growth factor receptor-2 (HER2+) metastatic breast cancers (mBCs), with efficacy further confirmed by DESTINY-B03, demonstrating improved progression-free and overall survival versus trastuzumab emtansine. Despite its efficacy, T-DXd had notable adverse events (AEs), including interstitial lung disease, necessitating real-world studies on safety and tolerability. Findings from such studies may help guide treatment selection and inform risk-benefit discussion in routine clinical practice.

Chemotherapy remains a standard treatment option for metastatic triple-negative breast cancer (TNBC) but is associated with limited survival. Although some targeted antibody-drug conjugates have demonstrated clinical benefits and are considered standard therapy, persistent unmet medical needs remain due to varying accessibility. The OptiTROP-Breast01 phase 3 trial assessed sacituzumab tirumotecan (sac-TMT) versus chemotherapy in patients with locally recurrent or metastatic TNBC who had received two or more prior therapies, including at least one for metastatic disease. Patients were randomize

Sacituzumab govitecan (SG) is a TROP2-directed antibody-drug conjugate with a payload of SN-38, a topoisomerase inhibitor. SG monotherapy has demonstrated activity in multiple solid tumors, including metastatic breast and bladder cancer. Rare tumors of the genitourinary tract often exhibit aggressive clinical behavior yet lack effective treatment options. This article describes the design and rationale for an open-label, non-randomized, phase 2 trial of SG monotherapy or concomitant SG plus atezolizumab in locally advanced/metastatic bladder/urinary tract adenocarcinoma/urachal adenocarcinoma,

KX-01 (tirbanibulin, Klisyri®) is a recently FDA-approved drug for treating actinic keratosis, with a distinct dual mechanism of action combining microtubule disruption and non-ATP-competitive Src inhibition. This unique mechanism and novel chemotype highlight KX-01's potential as a payload for antibody-drug conjugates. In this study, we synthesized and evaluated KX-01 derivatives to enhance anticancer potency and explore functional groups suitable for antibody conjugation. Notably, replacing the morpholine group with an N-benzoylpiperazine scaffold resulted in an analog with significantly imp

In recent years, immunotherapy has been increasingly incorporated into the clinical care of both pediatric and adult patients with B-cell acute lymphoblastic leukemia (B-ALL) to improve outcomes. Four main categories of immunotherapies are used in B-ALL: (1) unconjugated monoclonal antibodies (mAbs), (2) antibody-drug conjugates (ADCs), (3) T-cell-engaging antibodies, and (4) CAR T cells. Although mAbs such as rituximab are primarily used in adults, the other modalities have demonstrated efficacy in both pediatric and adult patients with B-ALL. Among ADCs, inotuzumab ozogamicin (InO) has prove

Tuberculosis (TB) remains among the leading infectious causes of death. Due to the limited number of antimicrobials in the TB drug discovery pipeline, interest has developed in host-directed approaches to improve TB treatment outcomes. C-C motif chemokine-like receptor 2 (CCRL2) is a unique seven-transmembrane domain receptor that is upregulated by inflammatory signals and mediates leucocyte migration. However, little is known about its role in TB infection. Here, we show that Mycobacterium tuberculosis (Mtb) infection increases CCRL2 protein expression in macrophages in vitro and alveolar mac

BACKGROUND: Thymic epithelial tumors (TETs), including thymoma and thymic carcinoma, are rare thoracic tumors of the anterior mediastinum. For those with advanced disease, platinum-based chemotherapy is used as first-line treatment. However, there is no standard regimen established for TET at progression after initial therapy, and treatment options for advanced/recurrent TETs are limited. Trop-2, a transmembrane glycoprotein, is overexpressed in solid tumors including thymomas and thymic carcinomas. The treatment of patients with relapsed or refractory lymphoid neoplasms represents a significant clinical challenge. Here, we identify the pro-survival BCL-2 protein family member MCL-1 as a resistance factor for the BCL-2 inhibitor venetoclax in non-Hodgkin lymphoma (NHL) cell lines and primary NHL samples. Mechanistically, we show that the antibody-drug conjugate polatuzumab vedotin promotes MCL-1 degradation via the ubiquitin/proteasome system. This targeted MCL-1 antagonism, when combined with venetoclax and the anti-CD20 antibodies obinutuzumab or rituximab, results in tumor regressions

BACKGROUND: Antibody-drug conjugates (ADCs) have been the preferred regimens for human epidermal growth factor receptor 2 (HER2)-positive metastatic breast cancer (MBC) after trastuzumab. Unfortunately, there is little data showing which ADCs should be chosen for those patients whose treatment with tyrosine kinase inhibitors (TKIs) failed. This study aims to analyze the efficacy and safety between novel anti-HER2 ADCs and trastuzumab emtansine (T-DM1) for those with TKIs failure.
MATERIALS AND METHODS: HER2-positive MBC using ADCs from January 2013 to June 2022 were included, and all of them
