Plant Stress Signaling in a Cross-Kingdom Context: Metabolic Control of Redox and ROS-Ca2+-Hormone Interactomes.

Biological systems maintain functional integrity under environmental stress through tightly coordinated signaling networks, yet the mechanisms that preserve signal fidelity in the presence of stochastic oxidative and ionic fluctuations across taxa remain incompletely understood. Although redox regulation, coupling between reactive oxygen species (ROS) and Ca2+, and hormone-mediated feedback loops have been extensively characterized within individual kingdoms, a cross-kingdom framework linking these pathways through metabolite-mediated modulation is lacking. Here, a cross-kingdom compilation of

Physiological and metabolic reprogramming under stress combination in plants.

In nature, plants are rarely challenged by a single, isolated, environmental stress factor. Instead, they continuously experience a dynamic range of abiotic and biotic constraints, generating conditions of multifactorial stress combination (MFSC). Driven by anthropogenic activities, many stressors are increasing in their frequency, intensity, and duration, exposing plants to complex conditions that go well beyond two interacting factors. A recent conceptual advance is the recognition that plant responses to MFSC cannot be predicted by simply summing up plant responses of the different individu

HSF1 phase transition mediates stress adaptation and cell fate decisions.

Under proteotoxic stress, some cells survive whereas others die. The mechanisms governing this heterogeneity in cell fate remain unknown. Here we report that condensation and phase transition of heat-shock factor 1 (HSF1), a transcriptional regulator of chaperones1,2, is integral to cell-fate decisions underlying survival or death. During stress, HSF1 drives chaperone expression but also accumulates separately in nuclear stress bodies called foci3-6. Foci formation has been regarded as a marker of cells actively upregulating chaperones3,6-10. Using multiplexed tissue imaging, we observed HSF1

Pancreatic beta-cell failure and diabetes in mice with a deletion mutation of the endoplasmic reticulum molecular chaperone gene P58IPK.

The endoplasmic reticulum (ER) transmits apoptotic signals in the pancreas during ER stress, implicating ER stress-mediated apoptosis in the development of diabetes. P58(IPK) (DNAJC3) is induced during ER stress and functions as a negative feedback component to inhibit eIF-2alpha signaling and attenuate the later phases of the ER stress response. To gain insight into a more comprehensive role of P58(IPK) function, we generated deletion mutant mice that showed a gradual onset of glucosuria and hyperglycemia associated with increasing apoptosis of pancreatic islet cells. Lack of P58(IPK) had no

Comprehensive genomic analysis of the maize ACP gene family reveals ZmACP1-mediated salt stress tolerance in Arabidopsis.

This study identifies 11 ZmACP genes in maize, revealing their role in salt stress adaptation, with ZmACP1 overexpression enhancing salt tolerance in Arabidopsis. Soil salinity is an escalating global threat that compromises crop yields. Acyl carrier proteins (ACPs) are essential cofactors of type II fatty acid synthase that determine acyl-chain length and unsaturation, yet their evolutionary diversification and functional relevance to salt stress remain poorly understood in maize (Zea mays L.). In this study, we identified 11 nonredundant ZmACP genes encoding proteins of 105-141 amino acids t

Chronic predator stress in female mice reduces primordial follicle numbers: implications for the role of ghrelin.

Chronic stress is a known suppressor of female reproductive function. However, attempts to isolate single causal links between stress and reproductive dysfunction have not yet been successful due to their multi-faceted aetiologies. The gut-derived hormone ghrelin regulates stress and reproductive function and may therefore be pivotal in the neuroendocrine integration of the hypothalamic-pituitary-adrenal (HPA) and -gonadal (HPG) axes. Here, we hypothesised that chronic stress disrupts ovarian follicle maturation and that this effect is mediated by a stress-induced increase in acyl ghrelin and

Posttranscriptional reprogramming controls MASLD progression through chronic ER stress adaptation.

Metabolic dysfunction-associated steatohepatitis (MASH) and its progression to hepatocellular carcinoma remain major clinical challenges. Chronic endoplasmic reticulum (ER) stress, induced by sustained high-fat diet (HFD) intake, promotes hepatic inflammation, lipid accumulation, and hepatocellular dysfunction during MASH pathogenesis. While transcriptional responses are well characterized, the posttranscriptional mechanisms underlying hepatocyte adaptation to chronic ER stress remain poorly understood. Using an integrative approach combining transcriptomics, ribosome profiling, cytoplasmic po

Introducing Concurrent Imaging and Unidimensional Analytics for Plant Stress Responses.

Advancements in phenotyping technologies, including object imaging, high-throughput monitoring, and soft computing, are pivotal for understanding plant responses to environmental stresses. These technologies enable detailed analyses of morphological, physiological, and structural adaptations under abiotic and biotic stresses, such as drought. Current work using multimodal and multi-perspective image processing methods can capture the essential processes that enhance plant resilience and counteract stress by identifying morphological and biochemical indicators. However, the dynamic and complex

Emotional pain in non-human mammals: from neural circuitry and opioid signaling to dysthymia-like states, grief-like behaviors, and physiological dysregulation.

This review aims to analyze the neurobiological, pathophysiological, and ethological foundations of emotional pain in non-human mammals, with particular emphasis on the transition from adaptive responses to pathological states such as animal dysthymia and Takotsubo syndrome. In the review, four conditions that can activate emotional systems will be discussed, such as pain derived from surgery or injury, stressful conditions that do not cause pain (e.g., unknown noises or smells), separation stress where an animal is isolated from the group (e.g., dam-calf separation), and death of a familiar a

Cholesterol dysregulation and microglial NLRP3 inflammasome activation after ischemic stroke: implications for neuroinflammation and functional recovery.

Secondary neuroinflammation is an important determinant of tissue injury, repair, and neurological recovery after ischemic stroke, yet the lipid-metabolic environment in which this response develops remains incompletely integrated into current mechanistic models. This narrative review synthesizes PubMed-indexed, peer-reviewed literature published between 2010 and 2026 on post-stroke cholesterol metabolism, microglial lipid handling, NLRP3 inflammasome signaling, immunometabolic remodeling, therapeutic intervention, and recovery-related outcomes. Ischemic injury substantially remodels the local

Transcriptomic analysis reveals distinct expression patterns in focal cortical dysplasia.

OBJECTIVE: Focal cortical dysplasia (FCD) is a common cause of drug-resistant epilepsy in children and adults and exhibits substantial clinical and histopathological heterogeneity. However, morphological classification alone does not fully explain variability in epileptogenicity, disease severity, or treatment response. METHODS: We aimed to define molecular mechanisms underlying FCD heterogeneity using integrated transcriptomic and regulatory analyses. RNA and small RNA sequencing (RNA-seq) were conducted on cortical tissue resected during epilepsy surgery from 66 patients with histopathologi

Computational Identification of Novel Transcriptional Regulators and Functional Gene Clusters in Lactococcus lactis Using Integrated Bioinformatics Approaches.

Lactococcus lactis is an essential industrial "cell factory" used extensively in food fermentation and biotechnology. However, a critical biological question regarding the regulatory mechanisms of the microorganism's adaptation process remains unresolved: how does the bacterium transcriptionally coordinate the trade-off between primary metabolism and cell-surface remodeling during environmental stress and competence? To date, a unified, global model of its gene regulatory networks (GRNs) that accounts for this transition remains lacking. To address this fragmentation and eliminate selection bi

Multi-omics analysis reveals the mechanism of indirect hepatotoxicity of triptolide upon LPS stimulation.

PURPOSE: Triptolide (TP), an active compound derived from the traditional Chinese herb Tripterygium wilfordii Hook F, is notable for its therapeutic properties. However, hepatotoxicity remains its primary adverse effect. There is a new perspective about TP hepatotoxicity: hepatic hypersensitivity triggered by lipopolysaccharide (LPS) stimulation. However, the global molecular alterations underlying this synergistic effect remain poorly defined. METHODS: We employed an integrated proteomics and metabolomics approach to systematically characterize the molecular landscape in the livers of mice t

miRNA and mRNA Profiling Links Connexin Deficiency to Deafness via Early Oxidative Damage in the Mouse Stria Vascularis.

Pathogenic mutations in the non-syndromic hearing loss and deafness 1 (DFNB1) locus are the primary cause of monogenic inheritance for prelingual hearing loss. To unravel molecular pathways involved in etiopathology and look for early degeneration biomarkers, we used a system biology approach to analyze Cx30-/- mice at an early cochlear post-natal developmental stage. These mice are a DFNB1 mouse model with severely reduced expression levels of two connexins in the inner ear, Cx30, and Cx26. Integrated analysis of miRNA and mRNA expression profiles in the cochleae of Cx30-/- mice at post-natal

High-viscosity hydrogels in cartilage injury and osteochondral defect repair: mechanisms, design strategies, and clinical translation.

Cartilage injury and osteochondral defects remain major clinical challenges owing to the limited intrinsic regenerative capacity of cartilage and the inability of current treatments to restore durable hyaline tissue. Conventional hydrogels often fail under joint loading because of insufficient mechanical stability, poor interfacial integration, and limited biological responsiveness. In this review, high-viscosity hydrogels (HVHs) are considered within a performance-based framework that integrates post-deployment cohesion and structural retention, viscoelastic energy dissipation, and reversible

Age modulates liver responses to asparaginase-induced amino acid stress in mice.

Asparaginase is an amino acid-depleting agent used to treat blood cancers. Metabolic complications due to asparaginase affect liver function … ls. Unbiased exploration of the effect of asparaginase on the liver transcriptome revealed that the integrated stress response (ISR) was the only molecular signature shared across the ages, corroborating similar eukaryotic initiation factor 2 phosphorylation responses to asparaginase at all ages. Juvenile livers exhibited steatosis and iron accumulation following asparagina …

JUND-driven stress-responsive astrocytes promote neuronal apoptosis via enhanced gap junction signaling in autism spectrum disorder.

BACKGROUND: Dysregulated stress responses are increasingly implicated in the pathophysiology of autism spectrum disorder (ASD). Astrocytes, which are highly vulnerable to stress, critically support neuronal survival; however, their specific role in ASD remains poorly defined. METHODS: We analyzed single nucleus RNA sequencing data from postmortem cortices of ASD and control donors. Our analysis included enrichment analysis of astrocyte subpopulations and integrated pySCENIC-based GRN reconstruction, TF profiling, regulon-DEG analysis, and cell-cell communication. Mechanistic studies involved

The Reductive Stress Nexus: Integrated Pathways of Metabolic Dysregulation, Senescence and Blood-Brain Barrier Failure in Neurodegenerative Disease and Brain Aging.

Reductive stress refers to a pathological shift toward a more reduced state in one or more defined redox couples, including NAD+/NADH, NADP+/NADPH, and GSSG/GSH, within specific cellular or subcellular compartments. Low-molecular-weight (LMW) thiols, including cysteine, cysteamine, glutathione, homocysteine, and hydrogen sulfide, contribute to cellular redox buffering, thiol-disulfide exchange, and redox-dependent metabolic regulation. Under chronic activation of antioxidant enzymatic systems, expansion of the reduced thiol pool may shift redox homeostasis toward a hyperreduced state, suppress

The role of osteocalcin in regulating the acute stress response.

Osteocalcin (OCN), a bone-derived hormone, considered as an indicator of bone turnover. Beyond its canonical role in bone metabolism, OCN may have many other functions as well. Studies have shown that it may also regulate glucose and lipid metabolism, cognitive function, sexual function, and more. Recently, OCN has become one interesting hormone with potential effects on acute stress response (ASR), which is essential for vertebrates' survival. This review aims to comprehensively summarize the progress on the role of OCN in the pathophysiology of ASR and to thoroughly analyze the molecular mec

Antisemitism as a Chronic Cardiometabolic, Endocrine, and Glutamatergic Stressor Requiring Psychometric Monitoring.

We are seeing an exploding expansion of antisemitic attacks worldwide, raising concerns about their potential impact on biological stress regulation and health. Antisemitism is a historically persistent and structurally embedded form of social exclusion that may contribute to chronic psychosocial stress exposure. Building on research into intergenerational trauma, including neuroendocrine alterations in Holocaust survivors and their descendants, this commentary integrates psychometric, empirical, and conceptual approaches to propose a biologically grounded framework linking antisemitism to end
