OBJECTIVES: Biologic therapies targeting interleukin IL-17, IL-23 and IL-12/23 have substantially improved outcomes in patients with moderate-to-severe psoriasis. In real-world settings, treatment effectiveness depends not only on drug efficacy but also on patient adherence, which is essential for the appropriate interpretation of treatment persistence. The objectives of the present study were to evaluate adherence to biologic therapy with IL-17 inhibitors, IL-23 inhibitors and IL-12/23 inhibitors in patients with moderate-to-severe psoriasis in real-world clinical practice and to identify fac

INTRODUCTION: Interleukin-17 inhibitors (IL-17i) and interleukin-23 inhibitors (IL-23i) are advanced therapeutic options for moderate-to-severe psoriasis. In real-world settings, biologic persistence is commonly used as a proxy for effectiveness and safety, and a treatment-free status following biologic discontinuation may provide insights into disease remission. This study aimed to assess persistence and treatment-free status for IL-17i versus IL-23i among biologic-naïve patients with psoriasis in Japan.
PATIENTS AND METHODS: This retrospective cohort study analyzed data from the Japanese Me

Chronic graft-versus-host disease (cGvHD) is a major cause of late morbidity after allogeneic hematopoietic stem-cell transplantation (alloHSCT), particularly in severe fibrosing forms. Although IFNγ-related cytokines have been implicated in cGvHD, their contributions to disease progression, survival, relapse, and response to immunosuppression remain unclear. In this retrospective study, 552 alloHSCT recipients with serum samples collected at cGvHD onset (or day 180 in controls) were analyzed for IFNγ-related cytokines. Among 14 cytokines, IL-12p40 and IL-18 emerged as divergent signatures. Bo

Autism spectrum disorder (ASD) is a complicated neurodevelopmental syndrome characterized by abnormalities in social communication, lack of interests, and repetitive behaviors. Increasing evidence from recent studies indicates that neuroinflammation and immunological dysregulation play essential roles in the pathogenesis of ASD. This review consolidates current knowledge on two anti-inflammatory cytokines of the IL-1 family, interleukin-38 (IL-38) and interleukin-37 (IL-37), which have recently emerged as essential modulators of neuroimmune mechanisms in ASD, highlighting their emerging roles

Adenomyosis involves the infiltration of endometrial glands and stroma deep into the uterine tissue, causing disruption to the endometrial-myometrial interface (EMI). The role of interleukin-17 (IL-17) has been extensively studied in endometriosis, but its involvement in adenomyosis remains unclear. This study aimed to investigate the expression of IL-17 in eutopic and ectopic endometrium (adenomyosis) of individuals with adenomyosis at the level of EMI. Paired tissues of eutopic endometrium and adenomyoma were collected from 16 premenopausal women undergoing hysterectomy due to adenomyosis. T

Systemic juvenile idiopathic arthritis (sJIA) is a childhood rheumatic disease of unknown origin. Dysregulated innate immunity is implicated in disease pathology. We investigated if IL-1 inhibition affects circulating cytokines and monocyte gene expression. CD14+ monocytes from patients in the RAPPORT trial were analyzed by RT-PCR for expression of IL1B and transcription factors associated with monocyte activation. Serum IL-1ra decreased with treatment, and IL-18BP transiently increased. Serum levels of IL-1β, IL-6, IL-10 and IL-18 were unchanged. IRF5 and STAT6 were decreased, and PPARG was i

Mucin is a major component of mucus in gastrointestinal mucosa. Increase of specific sialomucins having Sda blood group antigen, NeuAcα2-3(GalNAcβ1-4)Galβ1-4GlcNAcβ-, is considered to be associated with expulsion of the parasitic intestinal nematode Nippostrongylus brasiliensis. In this study, we examined the relationship between interleukin (IL)-13 pathway and expression of Sda-sialomucins in small intestinal mucosa with N. brasiliensis infection. Nematode infection induced marked increases in small intestinal mucins that reacted with anti-Sda antibody in wild type (wt) mice. However, this in

OBJECTIVE: Interleukin 1 Receptor 1 (IL1R1) and its ligand, IL1β, are upregulated in cardiovascular disease, obesity, and infection. Previously, we reported a higher level of IL1R1 transcripts in platelets from obese individuals of the Framingham Heart Study (FHS), but its functional effect in platelets has never been described. Additionally, IL1β levels are increased in atherosclerotic plaques and in bacterial infections. The aim of this work is to determine whether IL1β, through IL1R1, can activate platelets and megakaryocytes to promote atherothrombosis.
APPROACH AND RESULTS: We found that

Estradiol-17β (E2) is a potent regulator of early pregnancy and the estrous cycle in pigs. Production of E2 occurs in the porcine myometrium, but the factors involved in its regulation are unknown. In this in vitro study, it was investigated whether interleukin (IL)-1β, IL-6, and tumor necrosis factor (TNF)-α affect the release of E2 from the porcine myometrium on Days 10 to 11, 12 to 13, and 15 to 16 of pregnancy and the estrous cycle. The expression of the cytochrome P450 family 19 (CYP19) gene and the presence of the aromatase cytochrome P450 protein in the myometrium confirmed the ability
