Effect of Intermittent Warming on the Quality and Lipid Metabolism of Blueberry (Vaccinium corymbosum L., cv. Duke) Fruit.

The change of lipid metabolism is a key point of blueberry fruit after refrigeration. This study was conducted to evaluate the effects of intermittent warming (IW) of "DuKe" blueberry fruit on its shelf life at 20 ± 0.5°C following 30 days of refrigeration. IW-treated fruit showed higher contents of phosphatidylcholine, linoleic acid, and oleic acid but lower contents of phosphatidic acid and palmitic acid compared to controls. Protective effects on the cell membrane were also reflected as inhibition of the activity of phospholipase D and lipoxygenase. The blueberry fruit showed a lower decay

Metabolomic analysis of untargeted bovine uterine secretions in dairy cows with endometritis using ultra-performance liquid chromatography/quadrupole time-of-flight mass spectrometry.

Endometritis is among the most common bovine uterine diseases; as a cause of infertility, it affects the progress of the cattle industry. In this study, we used a novel technique based on ultra-performance liquid chromatography/quadrupole time-of-flight mass spectrometry for comparative metabolomics of uterine secretions in healthy cows and cows with endometritis, classified based on clinical symptoms. Univariate and multivariate statistical analyses revealed significant differences between the two groups (n = 12). Compared with healthy uterine secretion samples, in the clinical endometritis s

Inhibition of 15-hydroxy prostaglandin dehydrogenase activity in rabbit gastric antral mucosa by 13-hydroperoxyoctadecadienoic acid.

The effect of 13-hydroperoxyoctadecadienoic acid (13-HPODE), a hydroperoxy adduct of linoleic acid (LA), on the activities of prostaglandin (PG) synthesizing and catabolizing enzymes in rabbit gastric antral mucosa was examined. 13-HPODE had no effect on the synthesis of PGE2, PGF2 alpha and PGD2 from exogenous arachidonic acid in the microsomal fraction of the gastric mucosa at concentrations ranging from 5-20 microM. On the other hand, at 1-10 microM, it inhibited the activity of 15-hydroxy PG dehydrogenase (PGDH), which catalyzes the initial step of catabolism of PGs, in a dose-dependent ma

Intracellular and extracellular untargeted metabolomics reveal the effect of acute uranium exposure in HK-2 cells.

Uranium exposure poses a serious threat to the health of occupational populations and the public. Although metabolomics is a promising research approach to study the toxicological mechanisms of uranium exposure, in vitro studies using human cells are scarce. Applying cultured cell metabolomics, we exhaustively analyzed the intracellular and extracellular differential metabolites upon uranium exposure and characterized the possible biological effects of uranium exposure on human kidney cells. Uranium exposure significantly induced disturbance in the amino acid biosynthesis and linoleic acid met

Impact of a high-fat diet on the fatty acid composition of the retina.

Structure and function of the retina mainly rely on its fatty acid (FA) composition. Evidence from epidemiological studies and from animal experiments indicates that FA composition of the retina is influenced by the diet. Mice under chronic high-fat diet (HFD) develop metabolic syndrome, a risk factor for diabetes that is associated with structural and functional alterations of the retina. Here, we studied the impact of chronic exposure of mice to HFD on retinal FA composition. C57BL/6 J male mice were fed either a chow diet or a HFD for 11 weeks. As expected, HFD induced weight gain, adiposit

Composition of fatty acids in a high-fat diet affects adipose tissue inflammation by inducing calreticulin on adipocytes and activating group 1 innate lymphoid cells.

Obesity-induced adipose tissue inflammation plays a critical role in the development of metabolic diseases. For example, NK1.1+ group 1 innate lymphoid cells (G1-ILCs) in adipose tissues are activated in the early stages of inflammation in response to a high-fat diet (HFD). In this study, we examined whether the composition of fatty acids affected adipose inflammatory responses induced by an HFD. Mice were fed a stearic acid (C18:0)-rich HFD (HFD-S) or a linoleic acid (C18:2)-rich HFD (HFD-L). HFD-L-fed mice showed significant obesity compared with HFD-S-fed mice. Visceral and subcutaneous fat

Expression Profiles of Fatty Acid Transporters and the Role of n-3 and n-6 Polyunsaturated Fatty Acids in the Porcine Endometrium.

Fatty acids (FAs) are important for cell membrane composition, eicosanoid synthesis, and metabolic processes. Membrane proteins that facilitate FA transport into cells include FA translocase (also known as CD36) and FA transporter proteins (encoded by SLC27A genes). The present study aimed to examine expression profiles of FA transporters in the endometrium of cyclic and early pregnant gilts on days 3 to 20 after estrus and the possible regulation by conceptus signals and polyunsaturated FAs (PUFAs). The effect of PUFAs on prostaglandin (PG) synthesis and transcript abundance of genes related

Monitoring the Shelf Life of Hemp Seed Oil Stored at Two Temperatures in Different Materials via Near-Infrared (NIR) Spectroscopy.

Hempseed oil (HSO) is extremely rich in unsaturated fatty acids, especially linoleic (18:2 n-6) and α-linolenic (18:3 n-3) acids, which determine its high sensitivity to oxidative and photo-oxidative degradations that can lead to rancidity despite the presence of antioxidant compounds. The aim of this work was to evaluate which material/temperature/light solutions better preserve HSO quality during its shelf life and to test NIR as a rapid, non-destructive technique for monitoring oxidation phenomena. Futura 75 hemp seeds were cold-pressed; the oil was packed into 20 mL vials of four different

Fetal Exposure to Endocrine Disrupting-Bisphenol A (BPA) Alters Testicular Fatty Acid Metabolism in the Adult Offspring: Relevance to Sperm Maturation and Quality.

Daily exposure to bisphenols can affect reproductive functions due to their pseudo-estrogenic and/or anti-androgenic effects. Testicular lipids contain high levels of polyunsaturated fatty acids necessary for sperm maturity, motility, and spermatogenesis. Whether prenatal exposure to bisphenols alters testicular fatty acid metabolism in adult offspring is unknown. Pregnant Wistar rats were gavaged from gestational day 4 to 21 with BPA and BPS (0.0, 0.4, 4.0, 40.0 μg/kg bw/day). Despite increased body and testis weight, the total testicular cholesterol, triglyceride, and plasma fatty acids were

Dietary phytanic acid-induced changes in tissue fatty acid profiles in mice.

The aims of this research communication were to investigate the in vivo tissue accumulation of phytanic acid (PA) and any changes in the tissue fatty acid profiles in mice. Previous in vitro studies have demonstrated that PA is a milk component with the potential to cause both beneficial effects on lipid and glucose metabolism and detrimental effects on neuronal cells. However, there is limited information about its in vivo actions. In this study, mice were fed diets containing either 0.00 or 0.05% 3RS, 7R, 11R-PA, which is the isomer found in milk and the human body. After 4 weeks, adipose ti

Inhibition of GABA(A) receptor (GABAR) currents by arachidonic acid in HEK 293 cells stably transfected with alpha1beta2gamma2 GABAR subunits.

Agonist-stimulated liberation of arachidonic acid and subsequent generation of active metabolites are established components of several signal transduction pathways including pathways that regulate ion channels. We evaluated the role of arachidonic acid and some related unsaturated and saturated fatty acids in the modulation of a stably expressed recombinant gamma-aminobutyric acidA receptor (GABAR) isoform, the alpha1beta2gamma2 isoform. Whole-cell currents evoked by 10 microM GABA were inhibited in a concentration-dependent manner by arachidonic acid (0.1-100 microM). This effect of arachido

Association between methylated fatty acids and early pregnancy outcomes in dairy cows: a plasma GC‒MS assay.

Early conception failure is a major contributor to infertility in dairy cows. Fatty acids (FAs) play critical roles in maintaining normal reproductive function. This study aimed to determine Long Chain Fatty Acids (LCFAs) and their derivatives in plasma and to explore their association with early pregnancy outcome in high producing dairy cows. Blood samples were collected from 36 inseminated cows at 30 days post-insemination (dpi). Pregnancy was diagnosed at both 30 dpi and 45 dpi. Plasma LCFA levels were quantified via gas chromatography-mass spectrometry (GC-MS). Cows that experienced concep

Alteration of epidermal lipid composition as a result of deficiency in the magnesium transporter Nipal4.

Lipids in the stratum corneum play an important role in the formation of the skin permeability barrier. The causative gene for congenital ichthyosis, NIPAL4, encodes a Mg2+ transporter and is involved in increases in intracellular Mg2+ concentrations that depend on keratinocyte differentiation. However, the role of this increased Mg2+ concentration in skin barrier formation and its effect on the lipid composition of the stratum corneum has remained largely unknown. Therefore, in the present study, we performed a detailed analysis of epidermal lipids in Nipal4 KO mice via TLC and MS. Compared w

Cadmium and copper-induced metabolic and proteomic changes in the root tip during early maize growth.

In this study, the metabolic adjustments performed by maize (Zea mays L.) seminal roots exposed to 25 µM Cd2+ or 25 µM Cu2+ at pre-emergence are compared, focusing on the proteomic changes after metal exposure. Root width was increased, and root length was decreased after 72 h of metal treatment. Both metals induced H2O2 accumulation and lipid peroxidation in the root tip. These changes were accompanied by increases in lipoxygenase activity and 4-hydroxy-2-nonenal content. NMR spectroscopy revealed that the abundance of 38 water-soluble metabolites was significantly modified by Cd and Cu expos
