How kanamycin A interacts with bacterial and mammalian mimetic membranes.

Biological membranes are natural barriers to the transport of molecules and drugs within human bodies. Many antibacterial agents need to cro … interactions of kanamycin A, as a representative aminoglycoside, with both bacterial and mammalian lipid bilayers at the molecular level. Computational studies revealed strong hydrogen bonding interactions between the hydroxyl and amino groups of the aminoglycoside with the ester carbonyl and phosphate groups of the lipids. Pentameric Architecture of the SARS-CoV-2 Envelope Protein Revealed by SEC-MALS, Cryo-EM, and Molecular Dynamics.

The SARS-CoV-2 envelope (E) protein is a small viroporin that drives viral assembly, budding, and host interactions, yet its structural organization has remained elusive. Earlier nuclear magnetic resonance spectroscopy studies hint at oligomerization without direct evidence, and the construct lacks the flexible C-terminal region. To bridge this gap, we synthesized the full-length E protein to investigate its oligomeric state. Using size-exclusion chromatography coupled with multiangle light scattering, we demonstrated that the E protein assembles as a stable pentamer in solution. We then recon

Plasmon Coupling Enhanced Raman Scattering Nanobeacon for Single-Step, Ultrasensitive Detection of Cholera Toxin.

We report the development of a novel plasmon coupling enhanced Raman scattering (PCERS) method, PCERS nanobeacon, for ultrasensitive, single-step, homogeneous detection of cholera toxin (CT). This method relies on our design of the plasmonic nanoparticles, which have a bilayer phospholipid coating with embedded Raman indicators and CT-binding ligands of monosialoganglioside (GM1). This design allows a facile synthesis of the plasmonic nanoparticle via two-step self-assembly without any specific modification or chemical immobilization. The realization of tethering GM1 on the surface imparts the

How do sterols determine the antifungal activity of amphotericin B? Free energy of binding between the drug and its membrane targets.

Amphotericin B (AmB) is a well-known polyene antibiotic used to treat systemic fungal infections. It is commonly accepted that the presence … rol complexes in a lipid bilayer. The free energy profiles for the AmB-sterol association in phospholipid bilayers containing 30 mol % of sterols were calculated and thoroughly analyzed. The results obtained confirm the formation of specific AmB/ergosterol complexes and are used to determine the energetic and structural origin of the enhanced affinity of AmB for …

Increasing Lipid Nanoparticle mRNA Copy Number Improves Transfection Potency.

Lipid nanoparticles (LNPs) are the leading platform for delivery of RNA-based therapeutics. There is considerable interest in optimizing the transfection potency of these systems. Standard formulation protocols involve mixing lipids in ethanol with a buffer containing mRNA at pH 4, followed by dialysis against a pH 7.4 saline buffer to remove ethanol and raise the pH. The dialysis process results in fusion of mRNA-lipid complexes and small vesicles formed at pH 4, driven by two processes: Conversion of the ionizable lipid from the positively charged bilayer form to the neutral oil form and fus

Designing a green-emitting viscosity-sensitive 4,4-difluoro-4-bora-3a,4a-diaza-s-indacene (BODIPY) probe for plasma membrane viscosity imaging.

Viscosity is a key characteristic of lipid membranes - it governs the passive diffusion of solutes and affects the lipid raft formation and membrane fluidity. Precise determination of viscosity values in biological systems is of great interest and viscosity-sensitive fluorescent probes offer a convenient solution for this task. In this work we present a novel membrane-targeting and water-soluble viscosity probe BODIPY-PM, which is based on one of the most frequently used probes BODIPY-C10. Despite its regular use, BODIPY-C10 suffers from poor integration into liquid-ordered lipid phases and la

The covalent modification of spectrin in red cell membranes by the lipid peroxidation product 4-hydroxy-2-nonenal.

Spectrin strengthens the red cell membrane through its direct association with membrane lipids and through protein-protein interactions. Spectrin loss reduces the membrane stability and results in various types of hereditary spherocytosis. However, less is known about acquired spectrin damage. Here, we showed that alpha- and beta-spectrin in human red cells are the primary targets of the lipid peroxidation product 4-hydroxy-2-nonenal (HNE) by immunoblotting and mass spectrometry analyses. The level of HNE adducts in spectrin (particularly alpha-spectrin) and several other membrane proteins was

Effects of halothane and propofol on purified brain protein kinase C activation.

BACKGROUND: Protein kinase C (PKC) has been implicated as a target for general anesthetic action in the central nervous system. Previous rep … and propofol stimulated purified brain PKC activity in vitro assayed with physiologically relevant lipid bilayers in the absence or presence of Ca2+. This effect appears to be mediated through the lipid-binding regulatory domain of PKC. The potencies of halothane and propofol in stimulating PKC in vitro are consistent with submaximal activation of PKC at clinic …

Distinct effects of progesterone and cholesterol on lipid membranes: insights from biophysical experiments and molecular dynamics simulations.

Steroid hormones, including progesterone, are known to exert genomic, non-genomic and non-specific effects. However, their influence on membrane biophysics remains unclear. In this study, we investigate the distinct membrane-modulating behaviour of progesterone compared to cholesterol, employing a multidisciplinary approach that combines fluorescence microscopy, steady-state spectroscopy, and atomistic molecular dynamics simulations. Our results demonstrate that, whereas cholesterol promotes lipid packing and stabilises phase-separated domains, progesterone disrupts phase separation, reduces l

Interactions of lipopolysaccharide with lipid membranes, raft models - a solid state NMR study.

Lipopolysaccharide (LPS) is a major component of the external leaflet of bacterial outer membranes, key pro-inflammatory factor and an important mediator of host-pathogen interactions. In host cells it activates the complement along with a pro-inflammatory response via a TLR4-mediated signalling cascade and shows preference for cholesterol-containing membranes. Here, we use solid state (13)C and (31)P MAS NMR to investigate the interactions of LPS from three bacterial species, Brucella melitensis, Klebsiella pneumoniae and Escherichia coli, with mixed lipid membranes, raft models. Heterogeneous Ganglioside-Enriched Nanoclusters with Different Densities in Membrane Rafts Detected by a Peptidyl Molecular Probe.

The specific features of the lateral distribution of gangliosides play key roles in cell-cell communications and the onset of various diseases related to the plasma membrane. We herein demonstrated that an artificial peptide identified from a phage-displayed library is available as a molecular probe for specific ganglioside nanoclustering sites in caveolae/membrane rafts on the cell surface. Atomic force microscopy studies indicated that the peptide specifically binds to the highly enriched monosialoganglioside GM1 nanodomains of reconstituted lipid bilayers composed of GM1, sphingomyelin, cho…

Functional studies of cochleate assemblies of an oligo-acyl-lysyl with lipid mixtures for combating bacterial multidrug resistance.

The cationic antimicrobial oligo-acyl-lysyls (OAKs) interact with lipid mixtures mimicking the composition of bacterial cytoplasmic membranes. We have reported the ability of one such OAK, C(12)K-7α(8), to cluster anionic lipids and to promote a structural change with lipid bilayers to form rolled cylindrical structures or cochleates, without requiring divalent cations for their assembly. These assemblies can be exploited for drug delivery, permitting their synergistic use with antibiotics in systemic therapy to increase efficacy and reduce toxicity. Our previous studies of the biophysical pro…

Role of Membrane Lipids in the Regulation of Erythrocytic Oxygen-Transport Function in Cardiovascular Diseases.

The composition and condition of membrane lipids, the morphology of erythrocytes, and hemoglobin distribution were explored with the help of laser interference microscopy (LIM) and Raman spectroscopy. It is shown that patients with cardiovascular diseases (CVD) have significant changes in the composition of their phospholipids and the fatty acids of membrane lipids. Furthermore, the microviscosity of the membranes and morphology of the erythrocytes are altered causing disordered oxygen transport by hemoglobin. Basic therapy carried out with the use of antiaggregants, statins, antianginals, bet

Rigid amphipathic fusion inhibitors, small molecule antiviral compounds against enveloped viruses.

Antiviral drugs targeting viral proteins often result in prompt selection for resistance. Moreover, the number of viral targets is limited. Novel antiviral targets are therefore needed. The unique characteristics of fusion between virion envelopes and cell membranes may provide such targets. Like all fusing bilayers, viral envelopes locally adopt hourglass-shaped stalks during the initial stages of fusion, a process that requires local negative membrane curvature. Unlike cellular vesicles, however, viral envelopes do not redistribute lipids between leaflets, can only use the energy released by
