Z-Guggulsterone Inhibits Triple-Negative Breast Cancer Progression by Blocking Autophagosome-Lysosome Fusion via OGT-Mediated SNAP29 O-GlcNAcylation.

Triple-negative breast cancer (TNBC) remains a clinical challenge due to its heterogeneity and lack of targeted therapies. This study aimed to evaluate the antitumor effects and underlying mechanisms of Z-guggulsterone (Z-GS), a natural product, in TNBC. Human TNBC cell lines (MDA-MB-231 and MDA-MB-468) were treated with Z-GS to assess proliferation, cell-cycle progression, apoptosis, and reactive oxygen species (ROS) accumulation. Autophagic flux and the OGT-SNAP29 signaling axis were investigated via Western blotting, immunofluorescence, molecular docking, and molecular dynamics simulation.

AdipoR1-AMPK axis suppresses breast cancer across molecular subtypes via multimodal cell death pathways, including ferroptosis and apoptosis.

Adipokines secreted by adipocytes have emerged as critical modulators of cancer progression, particularly in obesity-associated malignancies. However, their therapeutic relevance and the tumor types responsive to adipokine pathways remain unclear. To identify adipokine-driven cancers and assess the therapeutic potential of adipokine signaling, we conducted a pan-cancer transcriptome analysis of the expression of various adipokine receptors in 31 tumor types. AdipoR1 was most frequently amplified and overexpressed in breast cancer across molecular subtypes. In the functional analysis, AdipoR1 s

Microfluidic capture and spatiotemporal analysis: Chemical mechanisms of tumor exosome-mediated malignant cell transformation.

Tumor metastasis is the primary cause of death from malignant tumors. The elucidation of its molecular mechanism is of great significance for breakthroughs in targeted therapy. In this study, a microfluidic chip platform integrating "dynamic dilution-precise capture-mechanical stimulation-in situ analysis" was constructed. Through the design of bionic narrow channels, efficient separation of exosomes and functional research at the single-cell level were achieved. On the one hand, the mechanical microenvironment within the chip was utilized to regulate the secretion of tumor cell exosomes (incr

Tunable Integrin-Ligand Coupling Strength Modulates Cellular Adaptive Mechanosensing.

Cells sense and respond to the matrix by exerting traction force through binding of integrins to an integrin-specific ligand. Here, Arg-Gly-Asp (RGD) peptide is covalently conjugated to the double-stranded DNA (dsDNA) and stem-loop DNA (slDNA) tethers with a tension tolerance of 43pN and immobilized on a PEG substrate. Unlike dsDNA, which is ruptured under high tension, leading to the removal of RGD, slDNA remains bound even when ruptured. Our results suggest that cells adapt their adhesion state by modulating actin filament polymerization and cofilin phosphorylation, effectively balancing the

Role of Acorus calamus extract in reducing exosome secretion by targeting Rab27a and nSMase2: a therapeutic approach for breast cancer.

BACKGROUND: Exosomes are extracellular vesicles released by cells that mediate intercellular communication and actively participate in cancer progression, metastasis, and regulation of immune response within the tumour microenvironment. Inhibiting exosome release from cancer cells could be employed as a therapeutic against cancer. METHODS AND RESULTS: In the present study, we have studied the effects of Acorus calamus in inhibiting exosome secretion via targetting Rab27a and neutral sphingomyelinase 2 (nSMase2) in HER2-positive (MDA-MB-453), hormone receptor-positive (MCF-7) and triple-negati

In vitro anticancer effect of venom from Cuban scorpion Rhopalurus junceus against a panel of human cancer cell lines.

In Cuba the endemic species of scorpion Rhopalurus junceus has been used in traditional medicine for cancer treatment. However, there is little scientific evidence about its potential in cancer therapy. The effect of a range of scorpion venom concentrations (0.1, 0.25, 0.5, 0.75 and 1mg/ml) against a panel of human tumor cell lines from epithelial (Hela, SiHa, Hep-2, NCI-H292, A549, MDA-MB-231, MDA-MB-468, HT-29), hematopoietic origins (U937, K562, Raji) and normal cells (MRC-5, MDCK, Vero) was determined by the MTT assay. Additionally, the effect of venom on tumor cell death was assayed by Fl

Targeted Delivery of Gliotoxin-Loaded Nanocarriers Heightens Therapeutic Potential in Hypoxic Environment of Triple-Negative Breast Cancer Cells.

Triple-negative breast cancer (TNBC) presents significant therapeutic challenges owing to its aggressive nature and the lack of targeted treatments. The hypoxic tumor microenvironment further ameliorates resistance and promotes cancer stem cell maintenance. Gliotoxin, a potent anticancer fungal secondary metabolite, was selected as a therapeutic agent. However, toxicity studies indicate that gliotoxin induces respiratory toxicity, limiting its application in cancer therapy. To address these problems, this study introduces a targeted drug delivery system employing folic acid-functionalized gold

Silver-selenium hybrid nanocomposite with combined cytotoxic and metabolic reprogramming effects in triple-negative breast cancer.

Triple-negative breast cancer (TNBC) remains one of the most aggressive breast cancer subtypes due to its high metastatic potential, poor response to conventional therapies, and frequent relapse after chemotherapy. In this work, we report a novel hybrid nanosystem that integrates, within a single mesoporous silica nanoplatform, two inorganic nanoparticles with complementary antitumoral activities: silver nanoparticles confined inside a mesoporous silica shell and selenium nanoparticles nucleated on the external surface. The core@shell Ag@MSN nanosystem was functionalized with transferrin to gu

Ki-67 promotes circulating tumor cell intravasation and metastasis in breast cancer.

Ki-67, a canonical proliferation marker, represents a pivotal prognostic and predictive biomarker in breast cancer. Here, by profiling 88,208 single-cell transcriptomes of circulating tumor cells (CTCs) matched with primary and metastatic lesions in breast cancer mouse models, we uncover a striking enrichment of cell-cycle genes in CTCs, particularly in CTC clusters. Using in vivo CRISPR screens, we identify Ki-67 as an essential regulator of CTC intravasation, whose knockout reduces metastasis. Mechanistically, Ki-67 depletion does not curb proliferation but suppresses genes involved in maint

Reduction of primary tumor growth and metastasis in models of triple-negative breast cancer following tilorone administration via type I interferon signaling.

BACKGROUND: People with triple-negative breast cancer (TNBC) typically have poorer survival than those with other breast cancer subtypes, with fewer treatment options available. Type I interferon (IFN) signaling has been reported as an important regulator of metastasis and therapeutic response in TNBC, but the off-target toxicity of IFN therapies has limited progression to clinical use. Augmenting Bone Morphogenetic Protein (BMP) signaling has also been associated with anti-metastatic effects, but BMP therapies are not clinically available presently. Tilorone is an anti-viral drug that induces

Synthesis, DNA-binding affinity and cytotoxicity of the dinuclear platinum(II) complexes with berenil and amines ligands.

A series of platinium(II) complexes of formula [Pt2L4(berenil)2]Cl4.4HCl.2H2O where L is piperidine (1), 4-picoline (2), 3-picoline (3) or isopropylamine (4) was prepared and their cytotoxicity have been tested against the growth of human breast cancer cells. Evaluation of the cytotoxicity of these compounds employing a MTT assay and inhibition of [3H]thymidine incorporation into DNA in both MDA-MB-231 and MCF-7 breast cancer cells demonstrated that these compounds were more active than cisplatin. Data from the ethidium displacement assay indicated that these compounds show moderate specificit

Pharmacological Vitamin C-induced high H2O2 generation mediates apoptotic cell death by caspase 3/7 activation in breast cancer tumor spheroids.

BACKGROUND: Pharmacological vitamin C (Vit-C), or high-dose Vit-C has recently gained attention as a potential cancer therapeutic. However, the anticancer activity of Vit-C has not been investigated in realistic 3D models of human cancers, especially with respect to breast cancer (BC), and its potential benefits remain under debate. Herein, we investigate the activity and mechanism of action of pharmacological Vit-C on two BC tumor spheroids. METHODS: We developed two distinct types of BC tumor spheroids from MDA-MB-231 and MCF-7 cells. The spheroids underwent treatment with a range of concen

Cannabidiol-loaded-injectable depot formulation for the treatment of triple-negative breast cancer: design, development, in-vitro and in-ovo evaluation of its anticancer activity.

Triple-negative breast cancer (TNBC) is an invasive and difficult-to-treat carcinoma that represents 15-20 % of breast malignancies and is frequently diagnosed in younger women. Chemotherapy is the mainstay treatment approach. Cannabidiol (CBD), the main non-psychoactive cannabinoid, has shown a potential anticancer activity in TNBC, enhancing the effect of conventional antineoplastics. This research aims to develop in situ forming implants (ISFIs) as a long-acting depot formulation of CBD with potential application in TNBC. This formulation is intended to be administered in the tumor site dur

Adenovirus-mediated overexpression of dominant negative epidermal growth factor receptor-CD533 as a gene therapeutic approach radiosensitizes human carcinoma and malignant glioma cells.

PURPOSE: Epidermal growth factor receptor (EGFR) and other members of the ErbB family of receptor tyrosine kinases (RTK) mediate autocrine growth regulation in a wide spectrum of human tumor cells. We have previously demonstrated that in stably transfected mammary carcinoma cells a dominant negative (DN) mutant of EGFR, EGFR-CD533 is a potent inhibitor of EGFR and its cytoprotective signaling after exposure to ionizing radiation. In the present study, we further investigate the capacity of a genetic approach, using replication-incompetent adenovirus (Ad)-mediated transfer of EGFR-CD533 (Ad-EGF

MAPK11 in breast cancer cells enhances osteoclastogenesis and bone resorption.

Breast cancer cells frequently metastasize to bone and induce osteolytic bone destruction in patients. These metastases cause severe bone pain, high risk of fractures and hypercalcemia, and are essentially incurable and fatal. Recent studies show that breast cancer cells in bone activate osteoclastogenesis and bone resorption. However the underlying mechanism is poorly understood. This study shows that the p38 MAPK (p38) isoform MAPK11 (p38β) is expressed in breast cancer cells. By using specific small hairpin RNAs for MAPK11, we demonstrated that p38β-mediated p38 activity in breast cancer ce

Rational Development of Activatable Prodrugs of the GSTP1 Inhibitor NBDHEX: Turn-On NIR Fluorogenic Drug Delivery with Selective Anticancer Activity.

Glutathione-S-transferase pi (GSTP1), overexpressed in cancer cells, is known to inactivate many electrophilic anticancer drugs by glutathionylation. While NBDHEX is a potent and selective inhibitor of GSTP1 with good anticancer activity, it suffers from poor aqueous solubility and bioavailability. Herein, we report the reactive oxygen species (ROS)-responsive fluorogenic prodrug RK-251 of NBDHEX, by coupling it to a dual positively charged NIR fluorophore (QCy7) and a ROS-responsive unit. ROS-mediated activation of RK-251 in aqueous medium resulted in rapid uncaging of NBDHEX, with turn-on NI

Selective mitochondrial dysfunction and redox collapse induced by an indole chalcone in breast cancer cells.

Breast cancer (BC) is a major health concern and remains one of the most frequently diagnosed malignancies among women worldwide. The metabolism of tumour cells, particularly their energy metabolism and ability to adapt to changing environmental conditions, has long been a focus of research. We previously reported that the synthetic chalcone derivative ZK-CH-11d (11d) induces apoptosis through the mitochondrial pathway. In this study, we demonstrate a significant and selective effect of 11d on the energy metabolism of tumour cells, without affecting non-cancer cells. We highlight alterations i

HLA class I-specific nucleolin peptides induce therapeutic T cells in triple-negative breast cancer patients.

Triple-negative breast cancer (TNBC) is an aggressive subtype with poor prognosis and limited treatment options owing to the lack of expression of common therapeutic targets such as hormone receptors and HER2. Recent advances in immunopeptidomics have enabled the identification of tumor-associated antigen peptides that can elicit tumor-specific immune responses. We identified four novel HLA class I-restricted peptides derived from nucleolin (NCL), a protein overexpressed in TNBC that is associated with poor outcomes. We predicted and validated pNCL-01 through pNCL-04 because of its strong bind
