Elucidating the regulatory role of the Shine-Dalgarno sequence in structural gene expression of the malolactic enzyme operon using a GFP/mCherry model.

Malolactic fermentation (MLF) in wine relies on Oenococcus oeni, in which the mleA and mleP genes are translationally coupled. To explore the post-transcriptional regulation involved, we constructed parallel GFP/mCherry reporter systems that mimic both the native overlapping (translationally coupled) structure and an engineered non-overlapping (non-coupled) structure. Through site-directed SD2 mutagenesis coupled with computational prediction of 16 S rRNA hybridization energy (ΔG) and minimum free energy (MFE) of mRNA secondary structure, we quantified the effect of intergenic sequence feature

Induction of oxidative stress and antioxidative mechanisms in Phaseolus vulgaris after Cd application.

Oxidative stress has been shown to be of great importance in the toxicity of several metals (copper, zinc, ...). In this study, the relationship of cadmium phytotoxicity and antioxidative reactions in bean (Phaseolus vulgaris L.) plants was investigated. Eleven-day-old seedlings were exposed to an environmentally realistic concentration of cadmium (2 microM CdSO(4)). Several biochemical and physiological parameters were influenced even by these low concentrations. At the biochemical level, the antioxidative defence mechanism was significantly activated after 24 h of cadmium exposure. Some enzy

Multiple effects of 5,5'-diphenylhydantoin on the thyroid hormone system.

The studies described above indicate the likelihood of a significant effect of DPH on cellular functions that are regulated by T3 at concentrations of DPH that occur during treatment of patients with Dilantin. Thus, it is possible that sensitive measures of cellular thyroid status in man, if available, might indicate that DPH treatment causes a mild hypothyroid state which may be partially compensated for by an activity of DPH as a partial thyroid hormone agonist. This review illustrates the complex array of interactions of DPH with multiple elements of the thyroid hormone system ranging from

The NADPH metabolic network regulates human αB-crystallin cardiomyopathy and reductive stress in Drosophila melanogaster.

Dominant mutations in the alpha-B crystallin (CryAB) gene are responsible for a number of inherited human disorders, including cardiomyopathy, skeletal muscle myopathy, and cataracts. The cellular mechanisms of disease pathology for these disorders are not well understood. Among recent advances is that the disease state can be linked to a disturbance in the oxidation/reduction environment of the cell. In a mouse model, cardiomyopathy caused by the dominant CryAB(R120G) missense mutation was suppressed by mutation of the gene that encodes glucose 6-phosphate dehydrogenase (G6PD), one of the cel

Hexachlorobenzene, a dioxin-type compound, increases malic enzyme gene transcription through a mechanism involving the thyroid hormone response element.

Hexachlorobenzene (HCB) is a dioxin-type chemical that acts mainly through the aryl hydrocarbon receptor. Chronic exposure of rats to HCB increases the activity of malic enzyme (ME). In this report, we show that this increase is correlated with an induction of ME messenger RNA (mRNA) levels, with the maximal HCB effect achieved after 9 days of intoxication. This effect is specific for ME, as other liver enzymes, such as glyceraldehyde-3-phosphate dehydrogenase, phosphoenol pyruvate carboxykinase, and mitochondrial alpha-glycerol-3-phosphate dehydrogenase, are not affected by HCB. The induction

Response of hepatic mitochondrial alpha-glycerophosphate dehydrogenase and malic enzyme to constant infusions of L-triiodothyronine in rats bearing the Walker 256 carcinoma. Evidence for divergent postreceptor regulation of the thyroid hormone response.

To characterize the hepatic response to L-triiodothyronine (T3) in an experimental nonthyroidal disease, we determined the activity of hepatic mitochondrial alpha-glycerophosphate dehydrogenase (alpha-GPD) and cytosol malic enzyme (ME) as a function of the saturation of the nuclear T3 receptor during constant T3 infusions in rats bearing the Walker 256 carcinoma. Groups of control and tumor-bearing rats were infused by minipumps (Alza Corp., Palo Alto, CA) with vehicle, 1.2 or 4.5 micrograms T3/100 body wt per day for 3 d. The range for serum T3 was 47.2 +/- 4.1 to 165 +/- 17.3 ng/dl for the c

Multi-omics reveals the microbial succession-driven metabolic regulation during sequential fermentation of fresh Spirulina.

Fermented Spirulina has superior sensorial and bioactive properties, but the impact of microbial succession on metabolic regulations of these attributes remains unclear. This study investigates metabolic regulations via integrated multi-omics analyses that link microbial succession with coordinated shifts in volatile and non-volatile metabolites. Sequential fermentation enhanced the sensory score of Spirulina from 40.09 to 89.29 points and improved its in vitro hypoglycemic and hypolipidemic activities as depicted by higher α-glucosidase inhibition (90.2%), pancreatic lipase inhibition (36.5%)
