Comparison of the effects of a selective muscarinic receptor antagonist and hyoscine (scopolamine) on motion sickness, skin conductance and heart rate.

AIMS: Hyoscine (scopolamine), which is effective in the prophylaxis of motion sickness, shows similar binding affinities to all of the five known muscarinic receptor sub-types. The effectiveness of hyoscine was compared with zamifenacin (UK-76654), which binds selectively to the muscarinic M3 and m5 receptors. METHODS: Eighteen subjects received hyoscine hydrobromide 0.6 mg, zamifenacin 20 mg, or placebo (double-blind cross-over design). Sessions were 1 week apart and the drug (oral) was given 90 min prior to a motion sickness test. Motion sickness was elicited by cross-coupled stimulation on…

Prevention of motion sickness in flight maneuvers, aided by transfer of adaptation effects acquired in the laboratory: ten consecutive referrals.

Ten flyers, grounded because of nausea and vomiting, were referred as potential candidates for adaptation to cross-coupled angular accelerations in a slow-rotation room; such adaptation has been shown to "transfer" to flight maneuvers. There was no opportunity to attempt treatment in two candidates. Among the remaining eight, five regained flight status (62.5%); follow-up periods of those five candidates ranged from 10 to 27 months. In one of the three remaining candidates, a satisfactory level of adaptation was achieved but more than 4 months elapsed before his assignment to a duty squadron.

Dataset of subjective human responses and objective metrics related to motion sickness collected and computed on a winter research voyage.

The SA Agulhas II, a South African (SA) polar supply and research vessel, embarked on a Winter Research Cruise in July of 2022 with 83 passengers aboard. Subjective human responses related to motion sickness were collected from 63 participants through paperbound questionnaires and, in parallel, from 15 participants through a mobile application that is part of a novel human cyber-physical system. Additionally, the human cyber-physical system enabled participant location tracking using near-field communication technology. The motion sickness responses are answers to questions that describe a par…

Effect of passengers' active head tilt and opening/closure of eyes on motion sickness in lateral acceleration environment of cars.

This study examined the effect of passengers' active head-tilt and eyes-open/eyes-closed conditions on the severity of motion sickness in the lateral acceleration environment of cars. In the centrifugal head-tilt condition, participants intentionally tilted their heads towards the centrifugal force, whereas in the centripetal head-tilt condition, the participants tilted their heads against the centrifugal acceleration. The eyes-open and eyes-closed cases were investigated for each head-tilt condition. In the experimental runs, the sickness rating in the centripetal head-tilt condition was sign…

Correlation of phasic and tonic skin-conductance responses with severity of motion sickness induced by viewing an optokinetic rotating drum.

50 subjects viewed an optokinetic rotating drum for 12 min. Subjective ratings of motion sickness were obtained during the development of motion sickness. The tonic and phasic skin-conductance responses were recorded from the sites of finger, palm, and forehead during the baseline and drum-rotation periods. Analysis showed subjects' subjective ratings of motion sickness gradually increased during the 12 min. of the drum-rotation period. Both phasic and tonic skin conductance also gradually increased during drum rotation for all subjects. Pearson correlations for 50 subjects showed their rating…

A new goldfish model to evaluate pharmacokinetic and pharmacodynamic effects of drugs used for motion sickness in different gravity loads.

This paper proposes a new goldfish model to predict pharmacodynamic/pharmacokinetic effects of drugs used to treat motion sickness administered in differing gravity loads. The assumption of these experiments is that the vestibular system is dominant in producing motion sickness and that the visual system is secondary or of small import in the production of motion sickness. Studies will evaluate the parameter of gravity and the contribution of vision to the role of the neurovestibular system in the initiation of motion sickness with and without pharmacologic agents. Promethazine will be studied…

[Effects of pikamilon on the brain water-electrolyte balance and in models of responses to the noise and vibration exposure, and their combination with motion sickness].

Electric impedance measurements on awake rabbits with electrodes implanted into cortex, thalamus, and hypothalamus showed that modeling of the noise and general (wide-band) vibration, as well as their combination with sea-sickness action, leads to disturbances in the aqueous-electrolyte balance in brain. This is manifested by extracellular and (less pronounced) intracellular edema development. Picamilon administration (10 mg/kg, i.v.) completely prevented development of the vibration-induced intracellular edema and markedly reduced the development of damage in all other cases studied.

The effect on motion sickness and oculomotor function of GR 38032F, a 5-HT3-receptor antagonist with anti-emetic properties.

1. The 5-hydroxytryptamine (5-HT3) receptor antagonist, GR 38032F, which possesses potent anti-emetic properties in vomiting induced by cancer chemotherapeutic drugs, has been tested to determine its value in the prophylaxis of motion sickness induced by cross-coupled stimulation. The double-blind trial compared GR 38032F with both a placebo (lactose) and with hyoscine. In addition, studies of ocular pursuit and saccadic eye movements were carried out following the administration of each drug. 2. The prophylactic effect of GR 38032F on motion-induced nausea was indistinguishable from that of p…

[Effect of pharmacological agents to prevent the motion sickness syndrome on the cardiovascular system in simulating the early period of adaptation to weightlessness].

The application of pharmacological agents, preventing the motion sickness syndrome in modelling the early stages of adaptation to weightlessness by means of 6-hour antiorthostatic influence (-15 degrees), does not exert a negative action on the human cardiovascular system. The given agents normalize partially cerebral circulation, eliminating changes induced by antiorthostatic influence.

Vestibular habituation is associated with distinct autonomic signatures and greater reduction in visually induced motion sickness: A randomized controlled trial.

This study compared the efficacy of short-duration, high-intensity versus stepwise, progressively lengthened optokinetic drum-chair training in alleviating visually induced motion sickness (VIMS) and modulating autonomic function. Ninety-six VIMS-susceptible university students were randomly assigned to either a short-duration (S-D) group (one training session daily for 3 consecutive days, with each session terminated upon the first report of pronounced nausea) or a stepwise (S-W) group underwent a 6-day graduated protocol (one session daily, duration: 60 s/120 s/180 s). Efficacy was assessed …

Motion sickness-susceptible participants exposed to coherent rotating dot patterns show excessive N2 amplitudes and impaired theta-band phase synchronization.

Visually induced motion sickness (VIMS) can occur via prolonged exposure to visual stimulation that generates the illusion of self-motion (vection). Not everyone is susceptible to VIMS and the neural mechanism underlying susceptibility is unclear. This study explored the differences of electroencephalographic (EEG) signatures between VIMS-susceptible and VIMS-resistant groups. Thirty-two-channel EEG data were recorded from 12 VIMS-susceptible and 15 VIMS-resistant university students while they were watching two patterns of moving dots: (1) a coherent rotation pattern (vection-inducing and pot…
