In vitro antiviral activity of a peptide-nucleic acid solution against the human immunodeficiency virus and influenza A virus.

A peptide-nucleic acid solution which had previously been reported to show in vivo efficacy in several viral infections (i.e. influenza, hepatitis, mumps, encephalitis, etc) was tested in three independent laboratories, including the US National Institutes of Health by specific in vitro methods for HIV and Influenza A. The results of these studies demonstrated significant anti-viral activity of the peptide-nucleic acid solution against the Human Immunodeficiency Virus (HIV) and the Influenza A virus.

Metabolic activation and nucleic acid binding of acetaminophen and related arylamine substrates by the respiratory burst of human granulocytes.

Following stimulation with phorbol myristate acetate, human granulocytes were found to incorporate acetaminophen, p-phenetidine, p-aminophenol, and p-chloroaniline into cellular DNA and RNA. Phenacetin was not incorporated into nucleic acid or metabolized by such activated granulocytes. None of the substrates gave nucleic acid binding if the granulocyte cultures were not induced to undergo the respiratory burst. Additional studies on the binding of acetaminophen to DNA and RNA were made by use of both ring-14C-labeled and carbonyl-14C-labeled forms of this substrate. The finding that equivalen

Hierarchical Nucleic Acid Amplification-Driven DNA Hydrogel Interface for Label-Free, Ultratrace PFOA Monitoring.

Per- and polyfluoroalkyl substances (PFAS), particularly perfluorooctanoic acid (PFOA), represent persistent environmental contaminants that require sensitive and reliable analytical methods for monitoring. However, the efficient conversion of molecular recognition events into amplified analytical signals remains challenging. Herein, we report a label-free electrochemical impedance spectroscopy (EIS) sensing platform based on a target-triggered DNA hydrogel signal amplification interface for ultratrace PFOA detection. Upon specific recognition of PFOA by an aptamer probe, a strand displacement

Tetrahedral Framework Nucleic Acid-Based Small RNA Delivery: Prospects and Challenges in Bone and Cartilage Regeneration.

Bone and cartilage defects represent major clinical challenges, with an increasingly urgent need for effective repair and regeneration. In recent years, small RNA-based regulatory strategies, such as microRNAs (miRNAs) and small interfering RNAs (siRNAs), have emerged as promising candidates for promoting bone and cartilage regeneration. However, achieving efficient and safe targeted delivery remains a critical challenge. Tetrahedral framework nucleic acids (tFNAs), a type of three-dimensional DNA nanomaterial with precise structure and excellent biocompatibility, serve dual functions as both …

Lipid Nanoparticle Database towards structure-function modeling and data-driven design for nucleic acid delivery.

Lipid nanoparticles (LNPs) are the leading nonviral nucleic acid delivery technology, but LNP structure-function data remains fragmented and nonstandardized. Unlike protein engineering which is anchored by the centralized Protein Data Bank, the LNP field lacks a unified repository for systematic analysis. To address this, we develop Lipid Nanoparticle Database (LNPDB) ( https://lnpdb.molcube.com ), an integrated database and web tool that consolidates structural and functional data for 19,528 LNPs. LNPDB standardizes LNP featurization by encoding lipid composition, experimental methods, and fu

Advancements in Gene Delivery using Nucleic Acid Loaded Nanoparticles for Region Specific Delivery in Alzheimer's Disease.

Alzheimer's disease (AD) is a progressive and the most common neurodegenerative condition, having a deleterious effect on memory, eventually leading to death. In the recent past, gene therapy has emerged as a promising and revolutionary treatment for AD. This study demonstrated that nucleic acid-loaded nanoparticles which deliver small interfering RNA through lipid nanoparticles successfully reduced Alzheimer's disease-related symptoms in preclinical models by decreasing amyloid-β levels and enhancing cognitive abilities. However, every rose has its thorn, as the output of gene therapy is cons

Virus-like particles based on plant viruses and bacteriophages: emerging strategies for the delivery of nucleic acid therapeutics.

Nucleic acids have emerged as a robust modality for the treatment of various diseases that are considered undruggable in the context of small-molecule therapeutics. However, their clinical translation is hindered by the lack of safe and effective delivery across extracellular and intracellular barriers. Mammalian viral vectors and synthetic non-viral carriers have long dominated the delivery landscape, but these raise concerns about safety and immunogenicity, driving the search for alternative strategies. Recently, non-mammalian viral vectors (based on plant viruses or bacteriophages) and viru…

Association between nucleic acid COVID-19 vaccines and acute myocardial infarction in adults: a systematic review.

BACKGROUND: Post-marketing surveillance has documented cardiovascular adverse events following COVID-19 vaccination, including acute myocardial infarction (AMI); however, evidence regarding causal associations remains contradictory. OBJECTIVE: To determine whether a causal association exists between nucleic acid-based COVID-19 vaccines (mRNA and DNA platforms) and AMI in adults aged 18-80 years. METHODS: A systematic review following PRISMA 2020 guidelines searched PubMed, Cochrane CENTRAL, and Google Scholar for studies evaluating mRNA vaccines (Pfizer-BioNTech, Moderna) and DNA-based vacci

Detection of Locked Nucleic Acid and 2'-O-Methoxyethyl Gapmers from Mouse Samples Using ELISA.

Antisense oligonucleotide (ASO)-mediated therapy is promising for the treatment of a variety of genetic disorders, as primarily seen for Duchenne muscular dystrophy. As more ASOs advance in therapeutic development and enter clinical trials, it becomes necessary to have a means of quantifying their amounts in biological samples post-treatment. This information will be valuable for evaluating the safety and pharmacokinetic profiles of ASOs, and also in deciding how the efficacy of these drugs can be improved. Gapmers are a class of ASOs characterized by having a central DNA segment that is surro

Development of a rapid detection kit for chicken infectious anemia virus nucleic acid (RPA-LFD method).

Chicken infectious anemia virus (CIAV) is a major immunosuppressive pathogen that causes huge economic losses in the poultry industry. The virus persists in the host for an extended period, making it difficult to eliminate and highlighting the need for rapid and accurate detection methods. In this study, we developed a recombinase polymerase amplification-lateral flow dipstick (RPA-LFD) kit for on-site detection of CIAV. Optimal primers and probes were designed and screened based on conserved regions of the full-gene sequences of 251 CIAV strains, ensuring high specificity, with no cross-react

Decoding heavy metal tolerance in rice: Nucleic acid-based technologies shaping global food security.

Global rice production is critically threatened by heavy metal contamination, particularly cadmium (Cd) and arsenic (As), which compromises yield, diminishes grain nutritional quality, and exposes billions of consumers to nephrotoxic and carcinogenic risks. Conventional remediation strategies (soil amendments, water management, phytoremediation) are prohibitively expensive, temporally protracted, and fundamentally reactive, while conventional breeding is constrained by linkage drag, polygenic trait architecture, and absence of natural alleles that restrict toxic metal uptake from essential min

Sequence-validated trigger gating for false-positive control in point-of-care isothermal nucleic acid detection.

Nonspecific by-products generated during isothermal amplification can be propagated by downstream signal amplifiers, causing false-positive visual readouts and limiting the reliability of point-of-care nucleic acid tests. Here, we report a sequence-validated trigger-gating strategy for catalytic hairpin assembly (CHA) that improves false-positive control in isothermal visual RNA detection. In this design, upstream amplification products must pass sequence-dependent validation before they can release the trigger strand required to activate entropy-driven CHA, thereby limiting the carry-through
