Purpose: One of the most well-known medications for treating delusional infestation (DI) is pimozide. Many patients may be reluctant to initiate treatment unless a medication has anti-pathogenic properties, as they feel otherwise it does not address their concerns regarding infestation. In this article, we explore the evidence that pimozide has a range of antipathogenic effects and how this fact can aid in patient care. Materials and methods: A scoping literature review was performed using The National Library of Medicine (PubMed). The search terms used were pimozide AND anti-microbial OR anti

Naturally, a wide range of genetic and environmental variables predominate, such as bacterial, viral, and parasite infective entities that have been identified as carcinogenic bioagents. Many helminth and protozoan parasitic diseases are liable to cause human cancer. Conveniently, three trematode parasites viz. Schistosoma haematobium, Opisthorchis viverrini, and Clonorchis sinensis have been reported to be intrinsically linked with human cancer. Similar studies for other parasitic infections are still imprecise and need further validation. Plasmodium falciparum is known to cause holoendemic B…

Economic losses and adverse effects on animals' health and productivity due to gastrointestinal parasites constitute a significant challenge for expanding and improving the small ruminant industry in developing countries. This study aimed to determine the diversity and geographic distribution of gastrointestinal parasites infecting small ruminants in Grenada. Seven hundred and thirteen fecal samples from 159 sheep and 554 goats from 64 farms in Grenada, Carriacou, and Petite Martinique were collected for coprological examination. Of 713 sheep and goat samples, 640 were positive for gastrointes…

Blood-stage Plasmodium falciparum parasites rely on a non-photosynthetic plastid, the apicoplast, for survival, making it an attractive target for antimalarial intervention. Like the mitochondrion, the apicoplast cannot be generated de novo and must be inherited by daughter parasites during cell division. This inheritance relies on coordinated apicoplast positioning and fission, but the molecular mechanisms controlling these processes remain poorly understood. Here, we identify a previously uncharacterized P. falciparum protein (Pf3D7_0613600), which we name PfAnchor, as a key regulator of api…

Quantifying intestinal parasite load in freshwater fish requires the development of reliable, minimally invasive methods. We evaluated the use of a very thin and flexible endoscope to detect and quantify acanthocephalan parasites in 25 European chubs (Squalius cephalus). The presence and number of parasites, as estimated using the endoscope, were correlated with dissection-based counts, especially in the anterior part of the intestine accessible via the insertion tube. Accuracy declined in highly infected fish due to overlapping parasites, but endoscopy still provides an accurate evaluation of…

While the interaction between humans and their parasites is well studied today, taking a long view of infection throughout human evolution helps to place the current picture in context and identify trends in infection over time. After considering how early technologies may have facilitated the transmission of parasites to humans, we examine the association between humans and parasites through time using archaeological and genetic evidence. Techniques such as microscopy, immunoenzymatic assays and DNA analysis have identified a range of protozoa, helminths and ectoparasites in our ancestors. Ev…

The movement of molecules across the membranous barriers of a cell is fundamental to cellular homeostasis in every living organism. This vit … molecule under energy expenditure or against an established electrochemical gradient. Apicomplexan parasites invest up to 5% of their genome to encode for membrane transporters; however, only a few have been fully characterised, and their specific mechanisms remain largely unknown. Recent advances in structural biology and the application of state-of-the-a …

Ubiquitin C-terminal hydrolase 37 (UCH37 also known as UCHL5) is a conserved deubiquitinating enzyme (DUB) with dual roles in proteasomal degradation and chromatin remodeling in humans. Its Plasmodium falciparum ortholog, PfUCH37, is unusual in that it possesses both DUB and deneddylating activities. While PfUCH37 is enriched in proteasome preparations, its direct interaction and broader functions in Plasmodium remain unclear, particularly given the absence of the chromatin remodeling complex INO80 homologs. This study utilizes transgenic parasites and proteomics to identify PfUCH37-associatin…

Dogs are the most popular pets worldwide. Close contact between dogs and people increases the risk of transmission of various zoonotic parasitic infections. Given the importance of veterinary medicine in preserving the One Health concept, the aim of this research was to identify intestinal parasites that may have zoonotic potential and to evaluate risk factors (individual and environmental). The research was conducted in Serbia in 2022 and 2023 on 382 owned dogs, using qualitative methods of coprological examination with a concentration on parasitic elements. The overall prevalence of intestin…

Immune-mediated bowel diseases (IMBD), notably ulcerative colitis and Crohn's disease, impose a substantial global health burden due to their intricate etiology and escalating prevalence. The nexus between intestinal parasites and the gut microbiome in IMBD is a dynamic and complex field of study. Several studies have evidenced the capacity of intestinal parasites to modulate the gut microbiome, inducing alterations in microbial diversity, abundance, and metabolic activity. These changes are crucial in influencing the immune response and contributing to the development of IMBDs. Simultaneously…

OBJECTIVES: Recent work has demonstrated that automated fluorescence flow cytometry (FLC) is a potential alternative for the detection and quantification of Plasmodium parasites. The objective of this study was to apply this novel FLC method to detect and quantify Babesia parasites in venous blood and compare results to light microscopy and polymerase chain reaction methods.
METHODS: An automated hematology/malaria analyzer (XN-31; Sysmex) was used to detect and quantify B microti-infected red blood cells from residual venous blood samples (n = 250: Babesia positive, n = 170; Babesia negative…

A signature remains elusive of naturally-acquired immunity against Plasmodium falciparum . We identified P. falciparum in a 14-month cohort of 239 people in Kenya, genotyped at immunogenic parasite targets expressed in the pre-erythrocytic (circumsporozoite protein, CSP) and blood (apical membrane antigen 1, AMA-1) stages, and classified into epitope type based on variants in the DV10, Th2R, and Th3R epitopes in CSP and the c1L region of AMA-1. Compared to asymptomatic index infections, symptomatic malaria was associated with a reduced reinfection by parasites bearing homologous CSP-Th2R (adju…

Genome-wide screening in apicomplexan species has transformed our understanding of these parasitic protozoa. Kimmel et al. report a 'knock sideways' system and provide a powerful use case for its feasibility in a gene-by-gene screening in Plasmodium falciparum. Carefully deployed, a novel toolkit helps to dissect the biological uniqueness of an important parasite.

An associated microbiome of any host helps it in different metabolic processes ranging from the decomposition of food to the maturation of gametes. Organisms with a parasitic mode of life, though present at nutritious sites inside their host, maintain their own microbiome. Nevertheless, the comprehensive characterization and functionality of microbiome in parasitic organisms remain understudied. We selected two nematode parasites of Kashmir Merino sheep viz;Haemonchus contortus and Trichuris ovis based on their higher prevalence, difference in mode of nutrition, habitation site and effect on h…

Parasites in which freshwater snails are intermediate hosts pose a serious threat to human health worldwide. We show here that freshwater snails can potentially be controlled by leech predation; in principle, this approach could significantly reduce snail-borne parasitic diseases (SBPDs). Specifically, glossiphoniid leeches, Helobdella austinensis and congener species, consume freshwater snails indiscriminately, while other common leeches do not. A single adult H. austenensis, for example, can consume up to its weight in snails, e.g. Physella acuta, per day. Our predator-prey models suggest th…
