Perivascular myofibroblasts and microvascular occlusion in hypertrophic scars and keloids.

Microvessels in normal skin, granulation tissue, hypertrophic scar, keloid, and mature scar from human subjects were studied by transmission electron microscopy. Comparative observations suggested that most microvessels in hypertrophic scar and keloid are occluded or partially occluded, apparently owing to an excess of endothelial cells. Endothelial cell contraction was also supported by the observations, and perivascular satellite cells (pericytes), some of which were identified as myofibroblasts, were observed in hypertrophic scars and keloids. Among findings from statistical analyses were t…

Spongiform polioencephalomyelopathy caused by a murine retrovirus. II. Ultrastructural localization of virus replication and spongiform changes in the central nervous system.

The development of murine retrovirus induced spongiform polioencephalomyelopathy was studied sequentially by electron microscopy. During the initial 30 days, viral infection of the central nervous system, as evidenced by viral budding from membranes, was limited to the endothelial cells and pericytes. Viral particles were observed in the lumen of blood vessels, extracellular spaces and astrocytic endfeet surrounding blood vessels, but no morphological evidence of productive infection was found in astrocytes or neurons during early development of vacuolation. The earliest lesions in the neuropi…

When Barriers Break: Tight Junction Regulation and Dynamic Alterations of Barrier Integrity in Neurological Injury.

The blood-brain barrier and blood-spinal cord barrier (BBB/BSCB) are essential protective components for the healthy functioning of the central nervous system (CNS). While these barriers protect the CNS from peripheral factors, such as immune cells and blood products, they can become disrupted in pathological conditions and injury. The neurovascular unit (NVU) is composed of endothelial cells (ECs), pericytes, astrocytes, microglia, and neurons, all of which contribute to proper function and the maintenance of the BBB/BSCB. Tight junctions (TJs) unite cellular components and are modulated by b…

The Mechanism of Ferroptosis and Blood-Brain Barrier Damage in Cerebrovascular Diseases.

The blood-brain barrier (BBB) is a vital protective structure that prevents harmful substances in the blood from entering the central nervou … ption. Ferroptosis affects multiple key brain cell types, including endothelial cells, glial cells, pericytes, and neurons, potentially leading to BBB dysfunction in cerebrovascular diseases. This article explores the role of ferroptosis in different types of cerebrovascular diseases and its effects on various cells. It covers the latest research in this ar …

Hexokinase-2-glycolytic overload axis in brain disorders exacerbated by diabetes.

Subjects with diabetes have an increased risk of dementia, Parkinson's disease, and stroke. In the central nervous system, there is increased inflammation, blood-brain barrier permeability, amyloid-β and α-synuclein deposition, and decreased neuronal synaptogenesis and survival. The role of hexokinases in controlling glucose metabolism therein is often overlooked. We propose that hexokinase-2 dysfunction-driven glycolytic overload is critically involved. This increases glycolytic intermediates in astrocytes, microglial cells, microvessel endothelial cells, and pericytes, stimulating mitochondr…

Single-cell epigenetic landscape, microenvironment interactions, and gene regulatory modules of non-functioning pituitary adenomas.

The epigenetic landscape and tumor microenvironment (TME) interactions of non-functioning pituitary adenomas (NFPAs), benign tumors with high morbidity and recurrence rates, are not well characterized. We completed single-nucleus (sn) multiomics assays on 4 gonadotrope NFPAs (34,819 cells) and 11 non-diseased postmortem control pituitaries (51,535 cells), finding decreased proportions of tumor-associated endothelial cells and pericytes and increased proportions of macrophages. We identified bidirectional tumor-macrophage crosstalk comprising nine ligand-receptor interactions and experimentally…

Semaglutide attenuates neuroinflammation in male mice.

Glucagon-like peptide-1 receptor agonists (GLP-1RAs) have shown promise in preclinical models of neurodegeneration, with emerging evidence s … associated transcriptional signatures specifically in microglia, endothelial cells, and a subset of pericytes. Mechanistically, we identify a subset of Glp1r-expressing neurons in the dorsal vagal complex that, upon semaglutide treatment, regulate genes involved in anti-inflammatory signaling. Kir7.1 is the physiological target for hormones and steroids that regulate uteroplacental function.

Preterm birth is detrimental to the well-being of both the mother and the newborn. During normal gestation, the myometrium is maintained in a quiescent state by progesterone. As a steroid hormone, progesterone is thought to modify uterine and placental morphology by altering gene expression, but another direct mode of action has long been suspected. Here, we reveal the nongenomic molecular mechanism of progesterone as the activation of human and murine inwardly rectifying potassium channel Kir7.1, which is expressed in myometrium and placental pericytes during late gestation. Kir7.1 is also ac…

CircRNAs in the tumor microenvironment: new frontiers in cancer progression and therapy.

The tumor microenvironment (TME), a dynamic ecosystem which including immune cells, cancer-associated fibroblasts (CAFs), endothelial cells, pericytes and acellular components, is orchestrating cancer progression through crosstalk between malignant cells and stromal components and increasingly recognized as a therapeutic frontier. Within this intricate network, circular RNAs (circRNAs) have emerged as pivotal regulators due to their unique covalently closed structures, which confer exceptional stability and multifunctional capabilities. This regulation is mediated through multiple mechanisms, …

Placenta-Derived Mesenchymal Stromal-Like Cells Promote 3D-Engineered Muscle Tissue Differentiation and Vessel Network Maturation.

Placental-derived stromal-like cells (PLX-PAD) have been shown to facilitate muscle tissue recovery after injury and stimulate angiogenesis. This work assesses the impact of PLX-PAD cells on the vascularization and maturation of engineered skeletal muscle tissue. Specifically, their effects in direct co-culture with endothelial cells, pericytes, and myoblasts seeded within microporous 3D scaffolds are characterized. Additionally, the impact of hypoxic PLX-PAD cell-conditioned medium (CM) on vascularization and muscle differentiation of engineered tissue is monitored. Co-culture of PLX-PAD with…

Flow in fetoplacental-like microvessels in vitro enhances perfusion, barrier function, and matrix stability.

Proper placental vascularization is vital for pregnancy outcomes, but assessing it with animal models and human explants has limitations. We introduce a 3D in vitro model of human placenta terminal villi including fetal mesenchyme and vascular endothelium. By coculturing HUVEC, placental fibroblasts, and pericytes in a macrofluidic chip with a flow reservoir, we generate fully perfusable fetal microvessels. Pressure-driven flow facilitates microvessel growth and remodeling, resulting in early formation of interconnected and lasting placental-like vascular networks. Computational fluid dynamics…

Circadian rhythms in the blood-brain barrier: impact on neurological disorders and stress responses.

Circadian disruption has become more prevalent in society due to the increase in shift work, sleep disruption, blue light exposure, and trav … tes. The BBB is a highly selective dynamic interface composed of CNS endothelial cells, astrocytes, pericytes, neurons, and microglia that form the neurovascular unit (NVU). Circadian rhythms modulate BBB integrity through regulating oscillations of tight junction proteins, assisting in functions of the NVU, and modulating transporter functions. Circadian d …

The Role of Cytokines in Neutrophil Development, Tissue Homing, Function and Plasticity in Health and Disease.

Neutrophils are crucial innate immune cells and comprise 50-70% of the white blood cell population under homeostatic conditions. Upon infection and in cancer, blood neutrophil numbers significantly increase because of the secretion of various chemo- and cytokines by, e.g., leukocytes, pericytes, fibroblasts and endothelial cells present in the inflamed tissue or in the tumor microenvironment (TME). The function of neutrophils in cancer has recently gained considerable attention, as they can exert both pro- and anti-tumorigenic functions, dependent on the cytokine milieu present in the TME. Her…

Cerebral Hemangiopericytoma Manifesting as Epilepsia Partialis Continua: A Case Report.

UNLABELLED: Cerebral hemangiopericytomas are very rare mesenchymal tumours arising from pericytes surrounding the blood vessels in the brain. Most patients present with headaches, focal neurological findings and focal seizures with or without generalisation. Our patient chiefly complained of an uncontrollable movement of her right hand that was initially fleeting but later became continuous. Her symptoms were initially described as tremors. We found an intracranial tumour as a cause of her symptoms, suspected the tumour to be a meningioma and performed surgical extirpation which resulted in sy…
