Yield prediction in a peanut breeding program using remote sensing data and machine learning algorithms.

Peanut is a critical food crop worldwide, and the development of high-throughput phenotyping techniques is essential for enhancing the crop's genetic gain rate. Given the obvious challenges of directly estimating peanut yields through remote sensing, an approach that utilizes above-ground phenotypes to estimate underground yield is necessary. To that end, this study leveraged unmanned aerial vehicles (UAVs) for high-throughput phenotyping of surface traits in peanut. Using a diverse set of peanut germplasm planted in 2021 and 2022, UAV flight missions were repeatedly conducted to capture image

Functional analysis of NtWRKY70 regulating tobacco defense against Rhizoctonia solani AG3-TB.

Rhizoctonia solani AG3-TB is a destructive necrotrophic pathogen causing tobacco target spot disease. WRKY70 acts as a key regulator integrating salicylic acid (SA) and jasmonic acid (JA) signals in plant immunity. Our previous study found that NtWRKY70 in Nicotiana tabacum L. is significantly induced by R. solani AG3-TB infection; however, its functional mechanism remains elusive. In this study, bioinformatics analysis showed that NtWRKY70 is a nuclear localized protein with conserved domains and transcriptional autoactivation activity. Further investigations using virus-induced gene silencin

Maize Aldehyde Decarbonylase 1 Gene (ZmCER1) Positively Regulates Salt and Drought Tolerance by Improving Wax Synthesis and Reactive Oxygen Species Detoxification.

Maize (Zea mays L.) is a vital global crop whose productivity is severely threatened by abiotic stresses. Epicuticular waxes provide a hydrophobic barrier that protects land plants from environmental stresses. However, the role of key wax biosynthetic enzymes, such as aldehyde decarbonylase CER1, in maize stress adaptation remains unclear. In this study, we performed a functional characterization of ZmCER1 in maize. Our results show that the overexpression of ZmCER1 in both Arabidopsis and maize substantially improved tolerance to these abiotic stresses. Under stress conditions, the transgenic

LcCOL7 and LcCOL8 Negatively Regulate Plant Flowering Independent of Day Length.

CONSTANS-LIKE (COL) genes are pivotal regulatory components in the photoperiodic flowering pathway of plants. These genes can be regulated by both photoreceptors and the circadian clock, modulating plant flowering responses under specific day lengths by regulating florigen levels. However, the COL gene family in Litchi chinensis Sonn. has not yet been characterized. In this study, we identified eight COL family members in litchi and classified them into three subgroups based on phylogenetic analysis. The analysis of cis-regulatory elements within the promoters of LcCOLs revealed a wide distrib

Climate Change and Nitrogen-Fixing Legumes: Investigating Stress-Modulated Dynamics of Carbon Fixation and Root Nodulation.

In the Anthropocene era, climate change is increasingly subjecting the crops to overlapping abiotic stressors such as drought, elevated temperatures, and air pollution, thereby disrupting their physiological integrity and functional performance. This review synthesises current knowledge on responses of N2-fixing plants to such stressors, focusing on core physiological processes and symbiotic nitrogen fixation via nodulation. The intricate interdependence between these traits is explored through the lens of altered source-sink relationships, which are highly sensitive to multifactorial environm

Dissecting flowering time and flower color in Carum carvi utilizing a long-read draft genome and a GBS-based QTL mapping.

Caraway (Carum carvi L.) is a major essential oil crop with biennial and annual flowering types. As basic research resource, we developed a draft genome assembly using long-read ONT sequencing and provide a structural and functional gene annotation for an annual caraway inbred line. To elucidate the genetic control of flowering, a genotyping-by-sequencing (GBS) was conducted for an F2 population (N = 187) generating 731 strictly filtered SNPs. A linkage map was constructed spanning 663 cM across 10 linkage groups with in total 634 (full map) or 259 (thinned map) SNPs. Contrary to its dominance

Genetic composition and heterozygosity of gibberellin-induced 2n gametes evaluated by SSR markers in Populus.

BACKGROUND: The artificial induction of unreduced (2n) gametes, due to first division restitution (FDR) or second division restitution (SDR), is an important method to produce triploids in higher plants. It is crucial to evaluate the genetic composition and heterozygosity transmission of induced 2n gametes with different formation mechanisms. RESULTS: Here, we produced 110 triploids by pollinating the female inflorescence of Populus alba × P. glandulosa with gibberellin-induced 2n pollen of Populus bolleana. The genetic composition and heterozygosity of the induced 2n pollen were inferred fro

Transcriptomic analysis reveals mechanism of light-sensitive albinism in tea plant Camellia sinensis 'Huangjinju'.

BACKGROUND: Camellia sinensis 'Huangjinju' is an albino tea variety developed recently in China. Young leaves of 'Huangjinju' demonstrate bright yellow when cultivated under natural sunlight, but regreens under reduced light intensity. To elucidate the physiological and molecular mechanisms of this light-sensitive albinism, we compared leaf pigmentation, metabolites, cellular ultrastructure and transcriptome between plants cultured under natural sunlight and shade. RESULTS: Shading treatment doubled the chlorophyll concentration and regreened albino leaves; carotenoid also increased by 30%. E

Natural and induced loss of function mutations in SlMBP21 MADS-box gene led to jointless-2 phenotype in tomato.

Abscission is the mechanism by which plants disconnect unfertilized flowers, ripe fruits, senescent or diseased organs from the plant. In tomato, pedicel abscission is an important agronomic factor that controls yield and post-harvest fruit quality. Two non-allelic mutations, jointless (j) and jointless-2 (j-2), controlling pedicel abscission zone formation have been documented but only j-2 has been extensively used in breeding. J was shown to encode a MADS-box protein. Using a combination of physical mapping and gene expression analysis we identified a positional candidate, Solyc12g038510, as

The wheat homolog of putative nucleotide-binding site-leucine-rich repeat resistance gene TaRGA contributes to resistance against powdery mildew.

Powdery mildew, one of the most destructive wheat diseases worldwide, is caused by Blumeria graminis f. sp. tritici (Bgt), a fungal species with a consistently high mutation rate that makes individual resistance (R) genes ineffective. Therefore, effective resistance-related gene cloning is vital for breeding and studying the resistance mechanisms of the disease. In this study, a putative nucleotide-binding site-leucine-rich repeat (NBS-LRR) R gene (TaRGA) was cloned using a homology-based cloning strategy and analyzed for its effect on powdery mildew disease and wheat defense responses. A three-season field study on the in-situ remediation of Cd-contaminated paddy soil using lime, two industrial by-products, and a low-Cd-accumulation rice cultivar.

To mitigate the serious problem of Cd-contaminated paddy soil, we investigated the remediation potential of combining in-situ immobilization with a low-Cd-accumulation rice cultivar. A three-season field experiment compared the soil pH, available Cd and absorption of Cd by three rice cultivars with different Cd accumulation abilities grown in Cd-contaminated paddy soil amended with lime (L), slag (S), and bagasse (B) alone or in combination. The three amendments applied alone and in combination significantly increased soil pH, reduced available Cd and absorption of Cd by rice with no effect on

Microarray analysis of the interaction between the aphid Rhopalosiphum padi and host plants reveals both differences and similarities between susceptible and partially resistant barley lines.

The bird cherry-oat aphid (Rhopalosiphum padi L.) is an important pest on cereals causing plant growth reduction without specific leaf symptoms. Breeding of barley (Hordeum vulgare L.) for R. padi resistance shows that there are several resistance genes, reducing aphid growth. To identify candidate sequences for resistance-related genes, we performed microarray analysis of gene expression after aphid infestation in two susceptible and two partially resistant barley genotypes. One of the four lines is a descendant of two of the other genotypes. There were large differences in gene induction bet

Recruitment of entomopathogenic nematodes by insect-damaged maize roots.

Plants under attack by arthropod herbivores often emit volatile compounds from their leaves that attract natural enemies of the herbivores. Here we report the first identification of an insect-induced belowground plant signal, (E)-beta-caryophyllene, which strongly attracts an entomopathogenic nematode. Maize roots release this sesquiterpene in response to feeding by larvae of the beetle Diabrotica virgifera virgifera, a maize pest that is currently invading Europe. Most North American maize lines do not release (E)-beta-caryophyllene, whereas European lines and the wild maize ancestor, teosin

Soy-RepViT: a lightweight deep learning framework for fine-grained recognition of soybean growth habits.

Soybean growth habit is generally classified into determinate, indeterminate, and semi-determinate types, and its accurate recognition is important for breeding selection, high-density cultivation, and agronomic management. However, the semi-determinate type is intermediate between determinate and indeterminate types and shows continuous transitional characteristics with the indeterminate type in stem elongation, upper internode length, and pod distribution, making fine-grained discrimination between these two types difficult. Practical deployment also requires lightweight, efficient models. T

Overexpression of the Vitis amurensis gene VabHLH015 in Arabidopsis thaliana and grape calli suppresses cold tolerance.

Cold stress represents a significant environmental challenge that constrains the development of the grapevine industry. The basic helix-loop-helix (bHLH) transcription factor family plays pivotal regulatory roles in plant growth, development, and stress responses. This study identifies a novel member of the bHLH family, referred to as VabHLH015, isolated from Vitis amurensis 'Zuoshan-1'. The physicochemical properties, conserved domains, and evolutionary relationships of VabHLH015 were thoroughly analyzed. Additionally, Arabidopsis thaliana and Vitis vinifera 'Chardonnay' calli overexpressing

Epigenetic Regulation of Divergent Co-Expression Between Whole-Genome Duplication and Transposed Duplication Genes and Its Impact on Catechin Accumulation in Camellia sinensis.

BACKGROUND/OBJECTIVES: Whole-genome duplication (WGD) and transposed duplication (TRD) are two principal evolutionary drivers of plant genome expansion, yet the molecular mechanisms underlying their divergent co-expression patterns remain poorly characterized. METHODS: In this study, based on the reference genome of tea plant (Camellia sinensis 'Yunkang 10', YK10), we integrated publicly available transcriptomic, ATAC-seq, H3K27ac ChIP-seq, whole-genome bisulfite sequencing (WGBS), and SNP data from eight tissues and performed a multi-layered analysis of co-expression divergence across 4071 W

Integrated morpho-physiological and biochemical responses of wheat genotypes to terminal heat stress and their role in determining grain yield.

Terminal heat stress is a major constraint to wheat production under global climate change, necessitating the identification of key adaptive traits and tolerant genotypes. This study evaluated 13 bread wheat (Triticum aestivum L.) genotypes under optimal and heat-stress conditions, focusing on morphological, physiological, and biochemical traits at the grain-filling stage. Significant genotypic variability was observed across all measured traits, indicating substantial diversity for heat adaptation. Based on the heat susceptibility index, genotypes TAW 119, DBW 187, and HD 2932 were identified

Advances and challenges in plant molecular marker technologies and their applications in the artificial intelligence empowered era.

Plant molecular marker technologies have reshaped crop genetics and breeding by making it possible to analyse genome-wide variation with a precision that phenotype-based selection, even in experienced programmes, cannot reach in routine practice. This review summarises recent progress in marker platforms from classical RFLP and SSR systems to high-throughput SNP genotyping, with emphasis on KASP, multiple nucleotide polymorphism and multi-gene panel technologies, and on sequencing-based methods such as GBS, GBTS and Hyper-seq that often serve as an upstream discovery layer for targeted assays

Integrated physiological, transcriptomic and metabolomic analyses reveal the mechanism of UV-B radiation enhancement to alleviate drought stress damage in Tibetan hulless barley (qingke).

Drought (D) and ultraviolet-B (UV-B) radiation present severe, frequently co-occurring constraints on crop productivity, particularly in high-altitude environments such as the Qinghai-Xizang Plateau. Although drought severely impairs plant growth and photosynthetic capacity, UV-B radiation can counterintuitively mitigate drought-induced damage; however, the molecular and metabolic mechanisms underpinning this stress cross-talk remain poorly understood. Here, using integrated physiological, transcriptomic, and metabolomic profiling, we examine how Tibetan hulless barley (qingke) responds to ind
