Investigation of the Ba2+-sensitive NH4+ transport pathways in the apical cell membrane of primary cultured rabbit MTAL cells.

BACKGROUND: Several apical ammonium (NH(4)(+)/NH(3)) transport pathways have been described in medullary thick ascending limb (MTAL) cells. The exact nature and importance of some of these pathways remain controversial. METHODS: Ammonium transport in primary cultured rabbit MTAL cells was investigated by measuring intracellular pH (pH(i)). RESULTS: To create physiological conditions, experiments were performed in the symmetrical presence of NH(4)Cl, which acidified the cells to pH(i) 6.89. When blockers of apical NH(4)(+) transport were used, the cells alkalinized due to a decreased NH(4)(+)

Emerging Role of Low-dose Loop Diuretics in the Management of Heart Failure Patients to Maintain Euvolemia.

The current scenario of employing loop diuretics in combination with guideline-directed medical therapy (GDMT) demonstrates a comprehensive approach to improving clinical outcomes in individuals with heart failure (HF). GDMT uses four types of drugs: angiotensin receptor-neprilysin inhibitors (ARNIs), beta-blockers (BBs), mineralocorticoid receptor antagonists (MRAs), and sodium-glucose cotransporter-2 inhibitors (SGLT2i). Torsemide, furosemide, and bumetanide are common loop diuretics used to control fluid overload in HF and provide symptomatic relief. Furthermore, loop diuretics are frequent

Effects of the external ions and metabolic poisoning on the constriction of the squid giant axon after axotomy.

After transecting the squid giant axon in the presence of an artificial external medium, which was composed of the ions normally present in squid blood, the cut ends of the axon constrict. This constriction could be completely blocked by cutting the axon in the presence of an artificial internal medium composed of the ions normally present inside the axon. By interchanging the ions in the internal medium with those in the external medium, it was determined that constriction was stimulated by the high concentrations of calcium, chloride, and magnesium ions present in the external medium and inh

Antispasmodic Effect of Asperidine B, a Pyrrolidine Derivative, through Inhibition of L-Type Ca2+ Channel in Rat Ileal Smooth Muscle.

Antispasmodic agents are used for modulating gastrointestinal motility. Several compounds isolated from terrestrial plants have antispasmodic properties. This study aimed to explore the inhibitory effect of the pyrrolidine derivative, asperidine B, isolated from the soil-derived fungus Aspergillus sclerotiorum PSU-RSPG178, on spasmodic activity. Isolated rat ileum was set up in an organ bath. The contractile responses of asperidine B (0.3 to 30 µM) on potassium chloride and acetylcholine-induced contractions were recorded. To investigate its antispasmodic mechanism, CaCl2, acetylcholine, Nω-ni…

Human CLC-K Channels Require Palmitoylation of Their Accessory Subunit Barttin to Be Functional.

CLC-K/barttin chloride channels are essential for NaCl re-absorption in Henle's loop and for potassium secretion by the stria vascularis in the inner ear. Here, we studied the posttranslational modification of such channels by palmitoylation of their accessory subunit barttin. We found that barttin is palmitoylated in vivo and in vitro and identified two conserved cysteine residues at positions 54 and 56 as palmitoylation sites. Point mutations at these two residues reduce the macroscopic current amplitudes in cells expressing CLC-K/barttin channels proportionally to the relative reduction in

A mathematical model of the pancreatic duct cell generating high bicarbonate concentrations in pancreatic juice.

OBJECTIVE: To develop a simple, physiologically based mathematical model of pancreatic duct cell secretion using experimentally derived parameters that generates pancreatic fluid bicarbonate concentrations of >140 mM after CFTR activation. METHODS: A new mathematical model was developed simulating a duct cell within a proximal pancreatic duct and included a sodium-2-bicarbonate cotransporter (NBC) and sodium-potassium pump (NaK pump) on a chloride-impermeable basolateral membrane, CFTR on the luminal membrane with 0.2 to 1 bicarbonate to chloride permeability ratio. Chloride-bicarbonate antip

Influence of trifluoperazine and verapamil on the isolated perfused rat kidney.

1. Isolated rat filtering kidneys were perfused with Ringer bicarbonate solutions containing either trifluoperazine (TFP, 50 microM) or verapamil (VER, 100 microM) to prevent tissue dysfunctions observed during perfusion. 2. Water, sodium and chloride kidney contents diminished under both treatments as compared with control preparations, and potassium content increased. 3. When albumin concentration was increased (10 g%) in the perfusion medium (nonfiltering kidney preparation) these effects of TFP or VER were not observed. 4. Lipid peroxidation and LDH release diminished significantly under 5

[Results of therapy of severe metabolic alkalosis using hydrochloric acid infusions].

Severe metabolic alkalosis is often treated by infusion of arginine-HCl. Since we know that this substance leads to a supplemental increase of intracellular pH and fails to reduce intracellular bicarbonate concentration we use HCl for correction of this disturbance of acid-base equilibrium. 18 intensive-care patients with severe metabolic alkalosis were treated with an infusion of 0.2 m HCl. While base excess and sodium decreased significantly, chloride increased slightly. Arterial plasma pH, potassium, Hb, Hk, pCO2, pO2, and SO2 remained unchanged. Instructions for preparing different HCl sol

Temperature dependence of muscle function in malignant hyperpyrexia-susceptible swine.

Contractures induced by halothane, caffeine, suxamethonium chloride and potassium in malignant hyperpyrexia-susceptible (MHS) and control swine skeletal muscle preparations were temperature-dependent, but differed in their temperature response profiles. Increasing the temperatures from 20 degrees C to 37 degrees C increased drug-induced contractures in MHS muscle. At 5 degrees C drug-induced contractures were not inhibited by dantrolene sodium, deuterium oxide or glycerination in either MHS or control muscle. These observations give further support to the view that the abnormality in MHS skele

Effects of desensitizing and/or anti-erosive toothpastes combined with mouthrinses on enamel structure and dental pulp fibroblasts viability.

This study evaluated the chemical profile of toothpastes (TPs) and mouthrinses (MRs) and their effects on tooth enamel ultrastructure, and the viability of human dental pulp fibroblasts (hDPF). Four TPs and MRs containing different remineralizing agents (arginine, potassium nitrate, pro arginine, and stannous chloride) were analyzed for pH, titratable acidity (TA), and ion concentrations (Ca+2, K+, Na+2). Enamel ultrastructure was evaluated using Fourier-transform infrared (FTIR) spectroscopy and scanning electron microscopy (SEM). Cell viability was assessed after treatments exposure. TPs and
