Large-scale meta- and cross-trait analyses uncover shared genetic risk factors for IBS and psychiatric disorders.

INTRODUCTION: Irritable bowel syndrome (IBS) is a common gut-brain axis disorder characterized by abdominal pain and altered bowel habits, and it shows high comorbidity with psychiatric disorders. However, the shared genetic mechanisms underlying these associations remain incompletely understood. METHODS: We performed a large-scale meta-analysis of IBS in individuals of European ancestry by integrating genome-wide association study (GWAS) summary statistics from the UK Biobank, Bellygenes, and the Million Veteran Program (MVP), thereby increasing statistical power to detect novel IBS loci. We

Genetic architectures of brain-related traits are shaped by strong selective constraints.

Genome-wide association studies (GWAS) have identified hundreds of significant loci for psychiatric disorders, yet the strength of these associations remains modest compared to other human complex traits with similar numbers of hits. Whether this pattern reflects statistical artifacts or real biological differences - and, if the latter, what underlies it - remains unclear. In addition to psychiatric disorders, we find that other traits with functional enrichment in the central nervous system (CNS), whether binary or quantitative, also share similar genetic architectures, characterized by GWAS

Computational and Experimental Biology Reveals Dihydroartemisinin's Efficacy Against Steroid-Induced Osteonecrosis of the Femoral Head Adjusting Ferroptosis via CCL17-PRDX6.

OBJECTIVE: According to existing research findings, dihydroartemisinin effectively regulates bone metabolism balance, while ferroptosis is c … finngen_R11_OSTEON_DRUGS in the Finnish database as outcome. The results showed that there were two quantitative trait loci that had a causal relationship with ferroptosis targets. There were 110 protein quantitative trait loci causally associated with plasma proteins from the UK Biobank, and none of these loci had an inverse causal relationship with SONFH. Through media …

Enrichment of regulatory expression quantitative trait loci among the CC variants was assessed in hemopoietic and intestinal cells. RESULTS: Three HLA alleles (HLA-B∗08:01, HLA-DRB1∗03:01, and HLA-DQB1∗02:01), related to the ancestral haplotype 8.1, were significantly associated with increased CC risk. We a …

BACKGROUND/OBJECTIVES: Rheumatoid arthritis (RA) is a chronic autoimmune disease that causes progressive joint damage and systemic complications. Despite multiple treatment options, many patients fail to achieve sustained remission. Our study aimed to integrate bioinformatics and single-cell RNA-seq analyses to identify potential biomarkers and therapeutic targets and explore bioactive compounds from traditional Chinese medicine (TCM). METHODS: We integrated gene expression quantitative trait loci (eQTL), protein quantitative trait loci (pQTL), and genome-wide association study (GWAS) data fo…

Identifying Potential Drug Targets in Coronary Atherosclerosis: Insights from the Druggable Genome and Mendelian Randomization.

PURPOSE: This study aims to identify therapeutic targets for coronary atherosclerosis (CA) using publicly available datasets while exploring its pathophysiologic mechanisms, mediators and potential side effects. METHODS: We conducted a two-sample Mendelian randomization (MR) and single-cell MR analyses integrating identified druggable genes to evaluate the causal relationship between expression quantitative trait loci (eQTL) and CA in both peripheral and central tissues. Druggable Genome-Wide Mendelian Randomization Identifies Distinct Therapeutic Targets for Low Back Pain, Intervertebral Disk Degeneration, and Sciatica.

OBJECTIVE: There are no effective drugs for the treatment of low back pain (LBP), intervertebral disk degeneration (IVDD), or sciatica. We aimed to identify potential therapeutic targets through druggable genome-wide Mendelian randomization (MR) analysis. METHODS: This study utilized large-scale expression quantitative trait loci (eQTLs) and protein quantitative trait loci (pQTLs), integrating existing druggable genome data. Conducted two-sample MR analysis to estimate the causal relationships between druggable genes with LBP, IVDD, and sciatica. Furthermore, we employed Bayesian colocalizati…

Investigation of chromosome 2q in osteoarthritis of the hand: no significant linkage in a Tasmanian population.

BACKGROUND: Previous studies have suggested a strong genetic component to osteoarthritis (OA), especially that of the hand, and three linkage studies have suggested the existence of susceptibility loci in disparate regions of chromosome 2q. OBJECTIVE: To examine for linkage to 2q in a Tasmanian population of women and men with familial hand OA. METHODS: Hand OA (distal interphalangeal, carpometacarpal, and Heberden's nodes) was assessed by a combination of hand photographs and radiographs. A non-parametric linkage (NPL) analysis was performed on chromosome 2q of 69 members in 22 families wit

sc-eQTL unveil immunogenetic architecture of polycystic ovary syndrome.

Polycystic ovary syndrome (PCOS) is a complex endocrine disorder characterized by metabolic, inflammatory, and reproductive dysfunction. We integrated genome-wide association study (GWAS) data, and single-cell expression quantitative trait loci (sc-eQTL) across various immune cells to elucidate the genetic and immune mechanisms underlying PCOS at the cellular level. Finally, we identified 19 risk loci for PCOS, prioritizing 16 candidate causal genes, approximately 30% of these genes are immune-related. Pathway analysis revealed 15 PCOS-associated signaling pathways, spanning hormonal regulatio…

LocusFocus: Web-based colocalization for the annotation and functional follow-up of GWAS.

Genome-wide association studies (GWAS) have primarily identified trait-associated loci in the non-coding genome. Colocalization analyses of SNP associations from GWAS with expression quantitative trait loci (eQTL) evidence enable the generation of hypotheses about responsible mechanism, genes and tissues of origin to guide functional characterization. Here, we present a web-based colocalization browsing and testing tool named LocusFocus (https://locusfocus.research.sickkids.ca). LocusFocus formally tests colocalization using our established Simple Sum method to identify the most relevant genes…

Genome-wide analysis of 102,084 migraine cases identifies 123 risk loci and subtype-specific risk alleles.

Migraine affects over a billion individuals worldwide but its genetic underpinning remains largely unknown. Here, we performed a genome-wide association study of 102,084 migraine cases and 771,257 controls and identified 123 loci, of which 86 are previously unknown. These loci provide an opportunity to evaluate shared and distinct genetic components in the two main migraine subtypes: migraine with aura and migraine without aura. Stratification of the risk loci using 29,679 cases with subtype information indicated three risk variants that seem specific for migraine with aura (in HMOX2, CACNA1A

Quantitative trait locus mapping for age-related cataract severity and synechia prevalence using four-way cross mice.

PURPOSE: The goal of this study was to map mouse quantitative trait loci (QTL) that influence the development of murine age-related cataract and synechia, by using a genetically heterogeneous mouse population bred by a four-way cross. METHODS: The test population consisted initially of 510 mice bred as the progeny of (BALB/cJ x C57BL/6J)F1 females and (C3H/HeJ x DBA/2J)F1 males. Each mouse was examined by slit lamp at 18 and 24 months of age and scored for degree of lens opacity on a 0 to 4+ scale, and the presence or absence of additional anterior chamber disease was noted. The presence of s…
