Expression of cytoskeletal and matrix genes following exposure to ionizing radiation: dose-rate effects and protein synthesis requirements.

Experiments were designed to examine the effects of radiation dose-rate and cycloheximide on expression of cytoskeletal and matrix elements in Syrian hamster embryo cells. Results here demonstrated little effect of dose-rate for fission-spectrum neutrons when comparing expression of alpha-tubulin or fibronectin genes. Effects of cycloheximide, however, revealed several findings: (a) Cycloheximide repressed accumulation of alpha-tubulin following exposure to high dose-rate neutrons or gamma-rays. (b) Cycloheximide did not affect accumulation of mRNA for actin genes. (c) Cycloheximide abrogated

Late effects of intraoperative radiation therapy on retroperitoneal tissues, intestine, and bile duct in a large animal model.

PURPOSE: The late histopathological effects of intraoperative radiotherapy (IORT) on retroperitoneal tissues, intestine, and bile duct were investigated in dogs. METHODS AND MATERIALS: Fourteen adult foxhounds were subjected to laparotomy and varying doses (0-45 Gy) of IORT (11 MeV electrons) delivered to retroperitoneal tissues including the great vessels and ureters, to a loop of defunctionalized small bowel, or to the extrahepatic bile duct. One control animal received an aortic transection and reanastomosis at the time of laparotomy; another control received laparotomy alone. This paper d

Effects of postoperative mediastinal radiation on completely resected stage II and stage III epidermoid cancer of the lung.

We randomly assigned 230 patients with resected Stage II or III epidermoid (squamous-cell) lung cancer to receive postoperative adjuvant radiotherapy or no adjuvant treatment. Careful intraoperative staging had been performed in all patients. Before randomization, patients were stratified according to stage, weight loss, age, and institution. Prognostic variables, such as stage, weight loss, age, nodal-disease status, and tumor status, were equally distributed between the two groups. The mean time from randomization to analysis was 3.5 years among the 210 eligible patients. There was no eviden

Fibroblasts from patients with inherited predisposition to retinoblastoma exhibit normal sensitivity to the mutagenic effects of ionizing radiation.

Retinoblastoma (RB) is a cancer of the retina which characteristically occurs in early childhood. Bilateral RB is an inherited form of this disease. Such patients are at greatly increased risk of subsequently developing second tumors in mesenchymal tissue, especially in areas exposed to ionizing radiation therapy. Fibroblasts from bilateral RB patients have been reported to be more sensitive than normal fibroblasts to the cytotoxic effects of ionizing radiation. Because xeroderma pigmentosum patients have a hereditary predisposition to UV-induced cancer and the cells of such patients are abnor

Study of ultraviolet radiation and genotoxic effects of natural sunlight in relation to skin cancer in Saudi Arabia.

Sunlight-related biological effects such as skin cancer, sunburning, and synthesis of vitamin D in the body have been found to be very low in people in the midregion of Saudi Arabia. The present studies were undertaken to measure the sunburning-carcinogenic ultraviolet light (UV) radiation (UV-B) in natural sunlight in the city of Riyadh (25 degrees north latitude). The average noontime incidence of UV-B, measured with a sunburn UV-meter, was found to be between 1.9 and 3.4 sun units/hr in the months of March to May. Concomitantly with the UV measurements, a bioassay using a wild-type and an e

Long term effects of radiation of T and B lymphocytes in peripheral blood of patients with Hodgkin's disease.

Total lymphocyte counts, and the percentage of T and B lymphocytes and monocytes in untreated patients with Hodgkin's disease were not significantly different from those observed in normal donors. At the completion of radiotherapy, the mean total lymphocyte count of 503/mm3 was 4 SD below the mean for normal controls. Although a group of 26 patients in continuous complete remission from 12 to 111 mo after radiation treatment regained normal total numbers of lymphocytes and monocytes, they exhibited a striking T lymphocytopenia and B lymphocytosis. Concomitantly, there was a significant increas

Dose-effect analysis of late dental adverse effects following radiation therapy in survivors of paediatric head and neck rhabdomyosarcoma.

INTRODUCTION: Paediatric head and neck rhabdomyosarcomas (HNRMS) often require radiotherapy, causing dental complications during critical development. Limited literature suggests dose-dependent risks, but clear thresholds for late dental adverse effects are lacking. This study aims to characterize dose effects on developing dentition in paediatric HNRMS patients to identify dose-response relationships informing evidence-based treatment planning. MATERIALS AND METHODS: Orthopantomograms from survivors of HNRMS were evaluated using the validated DENTALE scoring system. Radiation doses from brac

The double jeopardy effect of socioeconomic barriers on guideline-concordant postoperative radiation in oropharyngeal cancer.

Receipt of timely adjuvant radiation after primary surgery for head and neck cancer is a well-established quality benchmark. The social and structural drivers of suboptimal care, however, remain incompletely characterized. Very little work has been done on understanding the combined and interacting effects of social determinants on postoperative radiation therapy (PORT) receipt and timing. Upon examining PORT delivery among intermediate- and high-risk oropharyngeal squamous cell carcinoma patients, we identified that patients who are both uninsured and live far from a treatment center are at t

Determinants of real-world sunscreen effectiveness: from ultraviolet radiation photobiology to human behaviour.

Sunscreen is often presented as a straightforward intervention for reducing sun-induced skin damage. In practice, however, its effectiveness is determined by a broad network of interacting factors rather than labelled Sun Protection Factor (SPF) alone. Real-world protection depends not only on filter chemistry and test performance, but also on wavelength coverage, formulation, durability, regulatory standards, user behaviour and public understanding. At the same time, sunscreen has become an increasingly contested topic, shaped by debates regarding systemic absorption, environmental impact, mi

Radiation therapy in combination with immune checkpoint inhibitors in metastatic lung cancer: Effect of fractionation.

Immunotherapy with checkpoint inhibitors has improved the outcomes of patients with metastatic lung cancer in recent years. Despite improved prognosis, not all patients respond to treatment. Therapeutic interventions to build on the success of immune checkpoint inhibitors are needed. A retrospective review of patient records for patients who had received immune checkpoint inhibitors in a single cancer center over 4 years was undertaken. Demographic and disease characteristics of patients with metastatic non-small cell lung cancer were recorded. Data on other treatments including chemotherapy a

Protective Effect of Vinegar Powder on 60Coγ Radiation-Injured Mice and Its Potential Mechanism.

BACKGROUND: Ionizing radiation induces significant oxidative stress, immune dysregulation, and cellular injury. Conventional chemical radioprotectants usually exhibit considerable toxicity and undesirable side effects, thereby restricting their applicability for prolonged administration. OBJECTIVES: This study aimed to assess the radioprotective efficacy of vinegar powder (VP), a natural and nontoxic agent with potential for long-term administration. METHODS: Male Kunming mice were randomly allocated into 6 groups and orally administered saline, leucogen (12 mg/kg), and VP at doses of 1.35,

Radiation Therapy Combined With 5-Aminolevulinic Acid: Effect on Primary Human Meningioma Cells.

AIMS: Radiation therapy (RT) is a standard treatment for certain meningiomas; however, it does not consistently achieve favorable favourable outcomes. Therefore, discovering new agents that can enhance the radiosensitivity of meningiomas is of considerable interest. Evidence suggests that 5-aminolevulinic acid (5-ALA) acts both as a photosensitiszer and a radiosensitiszer. This study aimed to evaluate the efficacy of combining 5-ALA with RT on primary meningioma cells, which were obtained from eight patients during surgery. METHODS: Primary meningioma cells were characterized by immunocytoche

Effect and Mechanism of Tricholoma matsutake Extract on UVA and UVB Radiation-Induced Skin Aging.

Ultraviolet (UV) radiation often causes skin aging, inflammation, cancer and other related skin diseases. In this study, the main components of Tricholoma matsutake extract (TME) were identified using UPLC-Q-TOF-MS, and their anti-photoaging effects were assessed through UV-induced cell and animal models. The key components identified were D-mannitol (27.41%), DL-malic acid (14%), alginate (12.5%), isoleucine (4.82%), and phenylalanine (4.31%), all of which played roles in anti-aging and UV protection. TME (50-100 mg/ml) significantly alleviated UVA/UVB-induced erythema and wrinkles in mice. P

Penetration depth and effective sample size characterization of UV/Vis radiation into pharmaceutical tablets.

The pharmaceutical industry is moving from off-line to real-time release testing (RTRT) to enhance quality while reducing costs. UV/Vis spectroscopy has emerged as a promising tool for RTRT given its simplicity, sensitivity and cost-effectiveness. Nevertheless, the effective sample size must be characterized in relation to the penetration depth to justify its representativeness and suitability for RTRT. In this study, bilayer tablets were produced using a hydraulic tablet press. The lower layer contained titanium dioxide and microcrystalline cellulose (MCC), while the upper layer consisted of

Reference radiation selection is confirmed as a significant source of relative biological effectiveness variation for neutrons.

PURPOSE: To confirm that the selection of a reference radiation affects the magnitude and range of the relative biological effectiveness (RBE) evaluations of neutron test radiation. Particular attention was paid to the published thermal neutron RBE dataset that is highly variable, with values ranging from 5.4-51.1. MATERIALS AND METHODS: This involved a dual approach of 1) reaffirming dicentric chromosome assay (DCA) dose-response curve differences for 60Co, 137Cs, and 250 kVp X-rays, and 2) recalculating maximum RBE at minimal doses (RBEM) for our previously reported neutron data, accompanie

Unlocking the Potential of Ultra-High Dose Fractionated Radiation for Effective Treatment of Glioblastoma in Mice.

Background: Current radiotherapy regimens for glioblastoma (GBM) have limited efficacy and fails to eradicate tumors. Regenerative medicine brings hope for repairing damaged tissue, opening opportunities for elevating the maximum acceptable radiation dose. In this study, we explored the effect of ultra-high dose fractionated radiation on tumor responses and brain injury in immunocompetent mice which can better mimic the tumor-host interactions observed in patients. We also evaluated the role of the hypoxia-inducible factor-1 alpha under radiation as potential target for combating radiation-ind

Effect of binary porogen mixtures on polymer monoliths prepared by gamma-radiation initiated polymerization.

Gamma-radiation initiated polymerization is an easy-to-use and simple technique for the preparation of porous polymer monoliths widely used as active materials in separation columns. Herein, binary porogen mixtures of ethyl acetate/acetone/acetonitrile with methanol were used to synthesize diethylene glycol dimethacrylate monoliths with this technique. Systematic studies have been performed to determine the effect of the composition of the solvent mixture on the morphology and the reaction kinetics, including the growth mechanism and the growth rate. The irradiation dose-dependent conversion p
