An experimental model was designed to study the role of both diethylstilbesterol (DES) and phenobarbitone (PB) singly or in combination, in diethylnitrosamine (DEN) induced hepatic neoplasia. Rat submaxillary glands were exposed to excretory duct ligation, high-energy electron irradiation, or a combination of both. Ligation was done on the left excretory ducts, while 24 Gy irradiation was given within 8 days in three divided doses of 8 Gy. Surgically induced chronic gastric ulcers were created in rats. Cimetidine was administered for 1 year and the histological features of the gastric mucosa were studied to determine if cimetidine induced greater than normal proliferative or metaplastic changes or atypia. We examined the pathophysiological roles of the urea-urease-ammonia system in gastric ulcer disease using rats. Exposure of the stomach to ammonia (0.01-1.0%) decreased the transmucosal potential difference (PD) and histological injury in a concentration-dependent manner. Exposure of the stomach for 20 min to urea (0.025-0.2%) together with urease (100 IU) produced a decrease in PD and microscopic injury similarly, and the lesion was closely associated with the amount of ammonia produced. Aneurysms were produced on the common carotid artery of rats by milliwatt CO2 laser welding of an adventitia patch over a hole. Sixty-nine animals were operated on.
