Propionibacterium acnes bacteriophages display limited genetic diversity and broad killing activity against bacterial skin isolates.

UNLABELLED: Investigation of the human microbiome has revealed diverse and complex microbial communities at distinct anatomic sites. The microbiome of the human sebaceous follicle provides a tractable model in which to study its dominant bacterial inhabitant, Propionibacterium acnes, which is thought to contribute to the pathogenesis of the human disease acne. To explore the diversity of the bacteriophages that infect P. acnes, 11 P. acnes phages were isolated from the sebaceous follicles of donors with healthy skin or acne and their genomes were sequenced. Comparative genomic analysis of the

Integrative multi-omics analysis reveals gut-skin axis mechanisms and novel therapeutic target GALE in atopic dermatitis.

UNLABELLED: The gut-skin axis represents a critical but poorly understood pathway in atopic dermatitis (AD) pathogenesis. This study aimed to establish causal relationships between gut microbiota and AD risk while identifying key molecular bridges and therapeutic targets. We integrated multiple analytical approaches, including single-cell RNA sequencing analysis of skin biopsies from five AD patients and four healthy controls, intercellular communication network analysis, pseudotime trajectory inference, reverse drug prediction, molecular docking, and molecular dynamics simulations. Molecular and Genetic Markers for Malignant Melanoma: Implications for Prognosis and Therapy.

Despite therapeutic advancements, malignant melanoma remains a leading cause of skin cancer-related mortality, with incidence continuing to rise globally. Traditional prognostic tools offer important clinical guidance but fail to capture the biological heterogeneity of melanoma or reliably predict responses to emerging therapies. In this review, we summarize recent advances in prognostic and predictive molecular biomarkers reported over the past five years. We discuss immunohistochemical and tissue-based markers, circulating biomarkers, microRNAs, and gene expression profiles that enhance risk

Vulvar Lichen Sclerosus: A Literature Review with Consideration of Integrative Therapies.

Vulvar lichen sclerosus (VLS) is a chronic inflammatory skin condition characterized by vulvar pruritus, pain, dyspareunia, and architectural changes, including significant and permanent scarring and deformity of the vulva if left untreated. Untreated VLS significantly increases the risk of invasive squamous cell carcinoma, even in asymptomatic patients. However, there is an overall paucity of preclinical and clinical research on VLS. Although the disease is becoming more commonly recognized, it is often under- or misdiagnosed and its prevalence is likely underestimated. While the exact underl

Are antibiotics still relevant in acne? A review of the therapeutic conundrum.

Antibiotics have constituted the mainstay of acne therapy despite acne being classified as an inflammatory disorder. The indiscriminate usag … dies have highlighted the physiologic role of C. acnes in maintaining the normal homeostasis of the skin microbiome. The role of Malassezia in causation of acne has piqued interest in recent times. The efficacy of antibiotics in acne is attributed to their para-antibiotic, anti-inflammatory action rather than antimicrobial action. Thus, usage of low-dose antibiot …

Assessing microbial manipulation and environmental pollutants in the pathogenesis of psoriasis.

The cutaneous microbiome is increasingly recognized as a contributor to skin diseases like atopic dermatitis (AD) and psoriasis. Although traditionally AD and psoriasis have been viewed as having opposing immunologic findings, recent evidence suggests an overlap in ceramide-family lipid production in the protection against symptoms. We recently identified that specific environmental pollutants may drive dysbiosis through direct suppression of ceramide-family lipids produced by health-associated skin bacteria in atopic dermatitis (AD). We further demonstrated that one such bacteria, Roseomonas

The response of human skin commensal bacteria as a reflection of UV radiation: UV-B decreases porphyrin production.

Recent global radiation fears reflect the urgent need for a new modality that can simply determine if people are in a radiation risk of deve … d a different signature of protein oxidation/de-oxidation. We highlight that uncovering response of skin microbiome to radiation will facilitate the development of pre-symptomatic diagnosis of radiation risk in a battlefield exposure, nuclear accidents, terrorist attacks, or cancer imaging/therapy.

Obesity: A Modulator in Acne Management.

Acne vulgaris is an inflammatory and multifactorial skin disease involving the sebaceous gland and the skin microbiome. Different exposome factors, including hormonal and family factors, have been suggested to influence acne. Obesity is an increasingly observed condition worldwide, and is considered as a public health problem by the World Health Organization. Recently, a high body mass index has been identified as an acne severity risk factor in adolescents. A group of 5 dermatologists from different institutions and private practices from France involved in the research and clinical fields of…

Potential Mechanisms of the Sparing of Atopic Dermatitis in the Diaper Region: A Scoping Review.

Atopic dermatitis (AD) is a chronic, inflammatory skin condition commonly affecting infants with notable sparing of the diaper region. Though sources anecdotally attribute this sparing to the physical barrier formed by the diaper and the subsequent retention of moisture, urine, sweat and feces, no studies have formally investigated the factors contributing to this sparing phenomenon. We performed a scoping literature review to investigate the factors involved in sparing of AD in the diaper region, namely humidity, scratching, urine, sweat, feces, and microbiome composition. A total of 130 pape

An approach on the potential use of probiotics in the treatment of skin conditions: acne and atopic dermatitis.

Acne and Atopic Dermatitis (AD) are chronic inflammatory skin conditions with severe impact on a patient's life. Current treatments are related to adverse effects and do not represent a definitive cure. The present paper reviews the alterations in skin microbiome, specifically in acne and AD, and aims in searching for potential treatments based on benefic microorganisms, called probiotics. The review was made through bibliographic search of the main databases (Science Direct, PubMed, Scielo, Medline) between September 2015 and June 2016. Acne lesions create an environment that facilitates the …

Human demodicosis: revisit and a proposed classification.

Human Demodex mites (Demodex folliculorum and Demodex brevis) hold a high rank in the evolutionary and phylogenetic hierarchy of the skin microbiome, although in most people their presence is of no consequence. While human demodicosis is a skin disease sui generis, it can mimic many other inflammatory dermatoses, such as folliculitis, rosacea and perioral dermatitis, leading to unspecific and confusing descriptions in the literature. Here, we propose to classify human demodicosis into a primary form and a secondary form, which is associated mainly with immunosuppression. The clinical manifesta…

Antimicrobial peptides in the female reproductive tract: a critical component of the mucosal immune barrier with physiological and clinical implications.

BACKGROUND: At the interface of the external environment and the mucosal surface of the female reproductive tract (FRT) lies a first-line defense against pathogen invasion that includes antimicrobial peptides (AMP). Comprised of a unique class of multifunctional, amphipathic molecules, AMP employ a wide range of functions to limit microbial invasion and replication within host cells as well as independently modulate the immune system, dampen inflammation and maintain tissue homeostasis. The role of AMP in barrier defense at the level of the skin and gut has received much attention as of late.

Immunomodulating effects of the single bacterial strain therapy EDP1815 on innate and adaptive immune challenge responses - a randomized, placebo-controlled clinical trial.

The gut microbiome can modulate systemic inflammation and is therefore target for immunomodulation. Immunomodulating effects of EDP1815, a bacterial commensal strain of Prevotella histicola, were studied in healthy participants. Effects on adaptive immunity were evaluated by a neo-antigen challenge with keyhole limpet haemocyanin (KLH), while effects on innate immunity were evaluated by topical toll-like receptor 7 (TLR7) agonist imiquimod. Capsules with two enteric coating levels (EC1, EC2) were compared. Thirty-six healthy participants were included and received a daily dose of 8 × 1010 cell

INulin-type β-fructans supplementation to modulate gut microbiota and assessment of its effects on health status and GUT-SKIN axis parameters in patients with psoriasis (INGUTSKIN): the 8-week, randomised, double-blind, placebo-controlled clinical study protocol.

BACKGROUND: A strong, bidirectional association exists between gastrointestinal health and skin homeostasis in many chronic skin inflammations, including psoriasis (PS). It is postulated that, apart from genetic predisposition, the rise of local and systemic immune response in PS could be a consequence of intestinal dysbiosis associated with increased intestinal permeability. We hypothesised that restoring gut microbiome homeostasis and proper functioning of the intestinal barrier in PS patients would alleviate the inflammatory symptoms and severity of the skin lesions. The study aims to deter

Gut microbiota shapes the effects of saturated and polyunsaturated fatty acids on skin and hair follicle homeostasis.

High-fat diets are linked to obesity and skin and hair disorders, yet the role of specific lipid classes remains unclear. In this study, we show that cutaneous homeostasis is driven primarily by fatty acid (FA) composition rather than total fat content and is differentially regulated by the gut microbiota. Using omega-3 or omega-6/saturated FA-enriched diets under specific pathogen-free and germ-free conditions, we demonstrate that omega-6/saturated FA feeding promotes weight gain, dermal white adipose tissue expansion, sebaceous gland enlargement, and increased hair follicle proliferation. An Acid-Oxidising Solution Containing Hypochlorous Acid Reduces Staphylococcus aureus and Improves Bacterial Diversity in Epidermolysis Bullosa Wounds.

Epidermolysis bullosa (EB) is a group of rare genetic skin disorders characterised by skin fragility and chronic, painful wounds that are highly susceptible to bacterial infection, particularly by Staphylococcus aureus (SA). This study evaluated the efficacy of an acid-oxidising solution containing hypochlorous acid (HOCl) in reducing SA colonisation, promoting wound healing, and restoring a healthier microbiome in EB wounds. In a 12-week open-label pilot study, 15 EB patients applied the HOCl-based spray (APR-TD011) daily to chronic wounds for 8 weeks, with full-length 16S rRNA sequencing of

Genome of Malassezia arunalokei and Its Distribution on Facial Skin.

Malassezia is a fungal genus found on the skin of humans and warm-blooded animals, with 18 species reported to date. In this study, we sequenced and annotated the genome of Malassezia arunalokei, which is the most recently identified Malassezia species, and compared it with Malassezia restricta, the predominant isolate from human skin. Additionally, we reanalyzed previously reported mycobiome data sets with a species-level resolution to investigate M. arunalokei distribution within the mycobiota of human facial skin. We discovered that the M. arunalokei genome is 7.24 Mbp in size and encodes 4

Beyond commensalism: genomic insights into micrococcin P1-producing Staphylococcus chromogenes.

UNLABELLED: Staphylococcus chromogenes (S. chromogenes) is a predominant non-aureus staphylococcal species colonizing the teat skin and mammary gland of dairy ruminants. Although often linked to mild or subclinical mastitis, specific strains may also play protective roles against major udder pathogens. In this study, we characterized two S. chromogenes isolates (4S77 and 4S90) that displayed antimicrobial activity against Gram-positive bacteria. Complete genome sequencing revealed a conserved, plasmid-encoded biosynthetic gene cluster for the thiopeptide bacteriocin micrococcin P1 (MP1). All g
