Implication of the thalamus in sleep alterations observed in Alzheimer's disease.

Alzheimer's disease (AD), the most common neurodegenerative disorder, is frequently accompanied by sleep disturbances. Alterations of sleep, in both quality and quantity, contribute to the progression of AD neuropathology. The thalamus, through its diverse nuclei, plays a central role in sleep regulation and sleep-dependent memory consolidation, making it a relevant structure to investigate sleep disturbances associated with AD. Structural studies reveal that AD-related changes emerge early within the thalamus: Higher-order nuclei show atrophy, degeneration, and tau and Aβ accumulation. AD is

Impact of DSP4-induced locus coeruleus dysfunction on sleep: Systematic review and methodology.

Sleep and brain health are closely coupled, yet the mechanisms underlying this relationship remain incompletely understood. The locus coeruleus (LC) is a key modulator of sleep architecture, and LC dysfunction (LCd) is implicated in a wide range of neurodegenerative and psychiatric disorders, including Alzheimer's disease, Parkinson's disease, depression, and post-traumatic stress disorder. The isolated effects of LCd on sleep and brain health cannot be studied in humans, but they can be modeled in animals using the noradrenergic neurotoxin DSP4, which produces chronic and selective LC degener

Sleep, anxiety, and depression: The bidirectional brain-behavior axis.

Sleep, anxiety, and depression are interconnected processes that greatly influence mental health throughout life. Recent studies indicate that sleep issues are not just symptoms of mental illness but can also cause emotional and cognitive problems. Anxiety and depression can disrupt sleep patterns, and ongoing sleep difficulties can raise the likelihood of mood disorders. This reciprocal relationship involves neural pathways, neurochemicals, hormones, immune responses, and social factors. This chapter explores how sleep, anxiety, and depression interact from various scientific and clinical vie

Sleep and Neurodegeneration.

OBJECTIVE: This article discusses the relationship between sleep and neurodegenerative disorders. LATEST DEVELOPMENTS: Sleep disorders disrupt neurotransmitters, facilitating the progression of neurodegenerative changes. Sleep impairment promotes amyloid-β aggregation, propagating memory impairment in neurodegenerative disorders. The use of biomarkers for early diagnosis of neurodegenerative disorders may include sleep features. Sleep apnea is associated with an increased risk of Parkinson disease. ESSENTIAL POINTS: Sleep disorders, such as obstructive sleep apnea, are five times more likely

The Effectiveness of Melatonin for Sleep Disturbances in Parkinson' Disease: Systematic Review and Meta-Analysis.

Sleep disturbances in Parkinson's disease (PD) reduce quality of life. Melatonin, a sleep-wake hormone, shows treatment potential. However, its efficacy in PD remains unclear due to inconclusive findings across studies, warranting a systematic review to synthesise the available evidence. We searched PubMed, Cochrane, Web of Science, and Scopus for RCTs up to May 2025. Study quality was evaluated using the Cochrane Rob-2 tool. Statistical analysis was conducted using Review Manager version 5.4.1 with outcomes expressed as mean differences (MD) with 95% confidence intervals (CI). Out of eight st

Sleep in patients with traumatic brain injury: a systematic review and meta-analysis of actigraphy studies.

STUDY OBJECTIVES: Sleep disturbances are common in patients with traumatic brain injury. Actigraphy can provide an objective and naturalistic assessment of sleep patterns in patients with traumatic brain injury. Yet, the evidence of objective sleep-wake abnormalities using actigraphy in this clinical population remained unclear. To explore whether there might be differences in actigraphy-based sleep measures between patients with traumatic brain injury and controls. METHODS: An electronic systematic literature search was conducted using the following databases: PubMed, EMBASE, and PsycINFO, f

Targeting Sleep Physiology to Modulate Glymphatic Brain Clearance.

Sleep has been postulated to play an important role in the removal of potentially neurotoxic molecules, such as amyloid-β, from the brain via the glymphatic system. Disturbed sleep, on the other hand, may contribute to the accumulation of neurotoxins in brain tissue, eventually leading to neuronal death. A bidirectional relationship has been proposed between impaired sleep and neurodegenerative processes, which start years before the onset of clinical symptoms associated with conditions like Alzheimer's and Parkinson's diseases. Given the heavy burden these conditions place on society, it is i

Bidirectional relationship between sleep and depression.

Patients with depression almost inevitably exhibit abnormalities in sleep, such as shortened latency to enter rapid eye movement (REM) sleep and decrease in electroencephalogram delta power during non-REM sleep. Insufficient sleep can be stressful, and the accumulation of stress leads to the deterioration of mental health and contributes to the development of psychiatric disorders. Thus, it is likely that depression and sleep are bidirectionally related, i.e. development of depression contributes to sleep disturbances and vice versa. However, the relation between depression and sleep seems com

The relationship between the HDRS insomnia items and polysomnographic (PSG) measures in individuals with treatment-resistant depression.

The Hamilton Depression Rating Scale (HDRS), which includes several insomnia-related items, is potentially valuable in evaluating both depressive and sleep symptoms. However, the HDRS insomnia items have not been fully assessed by objective measures. This study compared the three HDRS insomnia items (Early, Middle, and Late) with the corresponding objective polysomnography (PSG) measures of Sleep Latency (SL), middle wakefulness, and late wakefulness. The study used HDRS and PSG data from 130 baseline nights, drawn from 80 participants enrolled in clinical trials for treatment-resistant depres

Hildegard of Bingen (c. 1098-1179) on sleep and dreams in her Causae et curae and Physica: a historical perspective.

Saint Hildegard of Bingen (c. 1098-1179) was a 12th century Benedictine abbess, a visionary, a composer, a poet, a healer, and one of few medieval women who produced treatises on medicine. In her medical writings, Causae et curae and Physica, the abbess described, among other topics, physical functions and mechanisms of sleep, dreams and waking, emphasizing the importance of sleep for the human body. She regarded sleep as both a passive and an active process. Hildegard warned her readers about sleeping too little or too much, evaluated possible causes of insomnia and nightmares, and discussed

Chronotherapy.

The objective of chronotherapy is to optimize medical treatments taking into account the body's circadian rhythms. Chronotherapy is referred to and practiced in two different ways: (1) to alter the sleep-wake rhythms of patients to improve the sequels of several pathologies; (2) to take into account the circadian rhythms of patients to improve therapeutics. Even minor dysfunction of the biological clock can greatly affect sleep/wake physiology causing excessive diurnal somnolence, increase in sleep onset latency, phase delays or advances in sleep onset, frequent night awakenings, reduced sleep
