BACKGROUND: Assessment of alkaline phosphatase (ALP) activity in cytologic smears is used as a phenotyping tool for hematopoietic and solid tissue neoplasms in dogs. Different procedures that vary in substrate-dye combinations, fixative, incubation times, and nuclear counterstains are available for detection of ALP activity. It is unknown if these procedures provide comparable results for phenotyping tumors in the same animal.

Preservation of fine cellular details of semi-adherent or suspension cells for imaging by immunofluorescence is challenging. This protocol enables staining of floating cells with minimal morphological distortions, as we demonstrate with the semi-adherent multiple myeloma cell line RPMI 8226. We describe steps to better preserve structural details by fixing, permeabilizing, and staining cells in solution, while minimizing the number of centrifugation steps and centrifugation g-force. BACKGROUND: Pleural mesothelioma (PM) is typically diagnosed late during the disease. Earlier detection can increase the chance of effective therapy. Recurrent pleural effusions are the earliest symptoms displaying an array of cytomorphological changes from reactive atypia to malignancy. OBJECTIVES: The cytomorphologic findings of bronchoalveolar lavage (BAL) in pediatrics were correlated with clinical symptoms.
METHODS: Patient demographics, clinical symptoms/history, cytomorphologic findings, and oil red O (ORO) staining of 100 pediatric patients who underwent BAL between 2014 and 2016 were reviewed at a large academic institution.
