Bone Tissue Engineering by Using Calcium Phosphate Glass Scaffolds and the Avidin-Biotin Binding System.

Highly porous and interconnected scaffolds were fabricated using calcium phosphate glass (CPG) for bone tissue engineering. An avidin-biotin binding system was used to improve osteoblast-like cell adhesion to the scaffold. The scaffolds had open macro- and micro-scale pores, and continuous struts without cracks or defects. Scaffolds prepared using a mixture (amorphous and crystalline CPG) were stronger than amorphous group and crystalline group. Cell adhesion assays showed that more cells adhered, with increasing cell seeding efficiency to the avidin-adsorbed scaffolds, and that cell attachmen…

Fabrication and characterization of biomimetic collagen-apatite scaffolds with tunable structures for bone tissue engineering.

The objective of the current study is to prepare a biomimetic collagen-apatite scaffold for improved bone repair and regeneration. A novel bottom-up approach has been developed, which combines a biomimetic self-assembly method with a controllable freeze-casting technology. In this study, the mineralized collagen fibers were generated using a simple one-step co-precipitation method which involved collagen self-assembly and in situ apatite precipitation in a collagen-containing modified simulated body fluid (m-SBF). The precipitates were then subjected to controllable freeze casting, forming sca

Gap junction intercellular communication: a review of a potential platform to modulate craniofacial tissue engineering.

Defects in craniofacial tissues, resulting from trauma, congenital abnormalities, oncologic resection or progressive deforming diseases, may result in aesthetic deformity, pain and reduced function. Restoring the structure, function and aesthetics of craniofacial tissues represents a substantial clinical problem in need of new solutions. More biologically-interactive biomaterials could potentially improve the treatment of craniofacial defects, and an understanding of developmental processes may help identify strategies and materials that can be used in tissue engineering. One such strategy tha…

Hepatic tissue engineering: from transplantation to customized cell-based liver directed therapies from the laboratory.

Today, liver transplantation is still the only curative treatment for liver failure due to end-stages liver diseases. Donor organ shortage, high cost and the need of immunosuppressive medications are still the major limitations in the field of liver transplantation. Thus, alternative innovative cell-based liver directed therapies, e.g. liver tissue engineering, are under investigation with the aim, that in future an artificial liver tissue could be created and be used for the replacement of the liver function in patients. Using cells instead of organs in this setting should permit (i) expansio…

A tissue engineering approach to bone repair in large animal models and in clinical practice.

The repair of large segmental bone defects due to trauma, inflammation and tumor surgery remains a major clinical problem. Animal models were developed to test bone repair by tissue engineering approaches, mimicking real clinical situations. Studies differed with regard to animals (dog, sheep, goat), treated bone (femur, tibia, mandible), chemistry and structure of the scaffolds. Still, an advantage in the bone formation and in the healing of the segmental defect was always observed when scaffolds were seeded with bone marrow derived stromal cells (BMSCs). In the year 1998 was performed the fi…

Cellular integration of thyrocytes and thyroid folliculogenesis: a perspective for thyroid tissue regeneration and engineering.

Thyroid gland is composed of many spheroid structures called thyroid follicles, in which thyrocytes are integrated in their specific structural and functional polarization. In conventional monolayer and floating cultures, the cells cannot reorganize follicle structures with normal polarity. By contrast, in a 3-D collagen gel culture thyrocytes easily and stably reconstruct follicles with physiological polarity. Integration of thyrocyte growth and differentiation appears to result in eventual thyroid folliculogenesis. 3-D collagen gel culture and subacute thyroiditis, a specific thyroid disorde

Biomimetic electrospun nanofibrous composites for bone tissue engineering: a review of materials, fabrication, and applications.

A flexible and promising method for creating polymeric nanofibrous composites for BTE (bone tissue engineering) is electrospinning. The optimal composites should resemble bone's natural ECM (extracellular matrix), possess suitable surface chemistry, exhibit strong mechanical strength, and demonstrate bioactivity and biocompatibility. Selecting the appropriate material for composite fabrication is a critical initial step in developing a BTE (tissue engineered) product. BTE is a multidisciplinary area that applies engineering concepts to bone-related biological processes. Key considerations in t…

Alginate-Methacrylate-RGD MicroSpheres (Alg-MR-MS) for cell expansion and delivery in tissue engineering.

Clinically relevant cell numbers are essential for many tissue engineering applications, requiring scalable expansion platforms. Microcarrier-based bioreactor systems address this demand; however, conventional non-dissolvable carriers require harsh enzymatic or mechanical detachment, reducing cell viability and integrity. Although dissolvable microcarriers improve recovery, most are designed only for harvesting and lack functionality for downstream tissue engineering. Here, we present a sequential dual-crosslinkable alginatemethacrylate-RGD microsphere system (Alg-MR-MS) that integrates scalab…

Natural Polymer-Based Scaffolds for Bone Tissue Engineering: A Review of Materials, Function, and Translation.

Scaffold-based bone tissue engineering (BTE) represents a pivotal strategy in regenerative medicine, offering substantial promise for repairing damaged bone. These scaffolds provide essential structural support and promote new bone formation by mimicking the native extracellular matrix (ECM). The rational design of an ideal scaffold requires careful balancing of biomaterial properties with biomechanical performance. Natural polymers have gained considerable attention in this context due to their inherent bioactivity, biocompatibility, and tunable mechanical properties. Their key advantage lies…

Can tissue engineering applications replace cardiovascular medical devices? Searching for the key to clinical translation.

Cardiovascular diseases are the number one killer worldwide. Malfunction of the conduction system, heart valves, the myocardial muscle or the vessels irrigating the tissue have been traditionally addressed with medical devices such as pacemakers, left ventricular assist devices, heart valves and vascular grafts. These positively impact critical outcomes such as functionality and mortality. However, by their non-biological nature, they cannot operate with the same efficiency as their original natural counterpart. Conversely, tissue engineering is an interdisciplinary field aimed at fabricating …

3D Printable and Electrically Conductive Hyaluronic Acid Hydrogel for Cardiac and Vascular Tissue Engineering Applications.

Cardiovascular disease (CVD) is the leading cause of death worldwide. After acute myocardial infarction (AMI), the adult human heart has very limited regenerative capacities, and the electrical propagation of the myocardium is severely disrupted, which makes the patient more susceptible to heart failure and death. Current pharmaceutical treatments do not address the loss of cardiomyocytes (CMs) or the disruption of electrical propagation in the heart. Tissue engineering provides a potential solution with electrically conductive hydrogels that are biocompatible to promote CM viability, lineage-…

Tissue engineering driven regeneration in oral squamous cell carcinoma: from biomaterials to precision gene editing.

Oral cancer is a significant global health challenge, ranking as the sixth most prevalent cancer worldwide, with approximately 377,000 new cases diagnosed annually. The high morbidity and mortality rates are largely attributed to tobacco and alcohol use. While conventional treatments such as surgery, radiation, and chemotherapy have improved survival rates, they often lead to unfavourable aesthetic and functional outcomes. Tissue engineering offers a promising alternative, providing regenerative solutions aimed at restoring both oral function and appearance. By integrating biomaterials, biolog…

Evaluation of Colombian silk fibroin hydrogels functionalized with recombinant LSECtin for intervertebral disc tissue engineering.

This study evaluated the potential of Colombian silk fibroin hydrogels functionalized with recombinant LSECtin (rLSECtin) for intervertebral disc tissue engineering. In silico analysis indicated that rLSECtin may interact with glycans similar to those found in human adipose-derived stem cells (ADSCs) through residues Asn105, Asp106, Glu93, Asn87, and Glu85, which coordinate calcium to form hydrogen bonds with GlcNAc hydroxyl groups. rLSECtin was successfully produced, purified, and incorporated into silk fibroin hydrogels. Thermal analysis showed that the hydrogels were resistant to sterilizat…
