Acute toxicity of diphacinone in Northern bobwhite: effects on survival and blood clotting.

The anticoagulant rodenticide diphacinone was slightly toxic (acute oral LD50 2014 mg/kg) to Northern bobwhite (Colinus virginianus) in a 14-day acute toxicity trial. Precise and sensitive assays of blood clotting (prothrombin time, Russell's Viper venom time, and thrombin clotting time) were adapted for use in quail, and this combination of assays is recommended to measure the effects of anticoagulant rodenticides. A single oral sublethal dose of diphacinone (434 mg/kg body weight) prolonged clotting time at 48 h post-dose compared to controls. At 783 mg/kg (approximate LD02), clotting time w

Permeability, cytotoxicity, and genotoxicity of chromium (V) and chromium (VI) complexes in V79 Chinese hamster lung cells.

The genotoxicity of Cr(V) complexes in mammalian cells (V79 Chinese hamster lung cells) has been studied for the first time using the in vitro micronucleus assay. Two complexes were investigated, [CrO(ehba)2]-, which undergoes ligand-exchange and disproportionation reactions in the cell growth medium, and [CrO(mampa)]-, which is chemically inert in the medium for the duration of the exposure period. Results of in vitro micronucleus assays show that both complexes are genotoxic and exhibit similar potencies to that of [Cr2O7]2-. The permeabilities of the Cr(V) complexes were also investigated f

Integrating new approach methodologies and artificial intelligence to advance central nervous system toxicity prediction: lessons from preclinical and clinical case studies.

Central nervous system (CNS) toxicities remain a major cause of drug attrition and represent a persistent challenge in predicting neurological risk during drug development. Limitations in the predictive resolution and translational relevance of conventional nonclinical paradigms contribute to uncertainty in identifying and interpreting neurotoxicity signals. This manuscript examines key challenges in CNS safety assessment and highlights emerging strategies to improve early detection and prediction of neurological risk. Through a series of case studies, we demonstrate practical approaches for i

Environmental and sustainable valorization of spent adsorbent: safety and acute toxicity evaluation in rats via probit analysis.

Valorization of adsorbent spent is a modern trend to maximize the utilization of absorption waste. Analytical methods, inclusive Fourier Transform Infra Red (FTIR) spectroscopy, Scanning Electron Microscope (SEM), and X-Ray Diffraction (XRD), were applied to evaluate the adsorbent spent (Zn-Co-Fe/LDH @ heavy metal). We examined the mechanistic toxicological effects of heavy metals within an animal model (rats) via probit analysis to evaluate the safety and acute toxicity of the spent adsorbent. The acute toxicity was evaluated and documented for 24 h, and it persisted for 14 days. A diverse mu

Prioritizing chemicals for developmental neurotoxicity by integrating data from a new approach methods (NAMs) battery covering key cellular events in neurodevelopment.

Thousands of chemicals have not been assessed for developmental neurotoxicity (DNT) despite potential human exposure, prompting efforts to screen chemicals for possible DNT hazard using in vitro approaches. Ninety-five chemicals were screened in in a battery including in vitro assays covering the key neurodevelopmental processes (KNDPs) of proliferation, apoptosis, cell migration, neuronal differentiation, neurite outgrowth, neuronal maturation and synaptogenesis, oligodendrocyte differentiation, and network formation using human and rat (primary cortical) cells. An in vivo assay of zebrafish

Single and combined chronic toxicity of cadmium and titanium dioxide nanoparticles in Aquarana catesbeiana (Anura: Ranidae) tadpoles.

The rising production and improper disposal of titanium dioxide nanoparticles (TiO2 NPs) into aquatic systems present considerable environmental challenges, especially when these particles interact with other contaminants such as cadmium (Cd). Thus, the current study aimed to evaluate the potential toxic effects on the gills, chondrocranium, body growth, and mortality of Aquarana catesbeiana tadpoles. The tadpoles were exposed to environmentally relevant concentrations of TiO2 NPs (10 µg L-1), and CdCl2 (10 µg L-1), both individually and in combination, for 30 days (chronic exposure), along wi

Development of an Interpretable Machine Learning Model for Neurotoxicity Prediction of Environmentally Related Compounds.

The rising prevalence of nervous system disorders has become a significant global health challenge, with environmental pollutants identified as key contributors. However, the large number of environmental related compounds, combined with the low efficiency of traditional methods, has resulted in substantial gaps in neurotoxicity data. In this study, we developed a robust and interpretable neurotoxicity prediction model using a high-quality data set. To identify the best predictive model, three molecular representation methods (molecular fingerprints, molecular descriptors, and molecular graphs

Treatment Approach of Oral Acute Toxicity in Domestic Pigeons by Spoiled Drinking Water Exposure to Insecticide Liquid (Phenylpyrazole), Fipronil.

Fipronil, a broad-spectrum phenylpyrazole insecticide, is widely used in agriculture and pest control but poses toxicity risks to non-target avian species through environmental contamination, particularly via drinking water. Despite known neurotoxic effects in birds, data on domestic pigeons (Columba livia) and effective clinical interventions remain limited. This study aimed to characterize the acute oral toxicity profile of Fipronil-contaminated drinking water in domestic pigeons and evaluate treatment strategies to mitigate intoxication symptoms. Using the OECD Avian Acute Oral Toxicity Tes

Benchmarking the sub-lethal chronic aquatic toxicity of an emerging biosurfactant (Sophorolipid) to a traditional amine oxide surfactant in a freshwater fish-pathogen system.

Surfactants, a broad category of amphiphilic compounds, are one of the largest categories of emerging contaminants within freshwaters, many of which have known aquatic toxicity profiles. This, together with a global push towards a greener economy that prioritises less carbon intensive methods of chemical production, has led to a rise in biosurfactants entering the market. However, their toxicity requires further assessment and benchmarking against traditional synthetic surfactants. This study benchmarks the sub-lethal chronic (31-day exposure) aquatic toxicity of an emerging biosurfactant, sop

Subchronic Exposure of Tri-Ortho-Cresyl Phosphate Reduces T-Cell Population in Mice.

Tri-ortho-cresyl phosphate (TOCP) is a widely used organophosphorus compound in industry and agriculture, but its immunotoxicity has not been thoroughly investigated. Therefore, this study aimed to assess the immunotoxicity of TOCP in mice by examining changes in immune cell populations and histopathological alterations in the spleen. Male ICR mice were administered TOCP via daily oral gavage at doses of 200 and 400 mg/kg body weight (BW)/d for 28 days to evaluate its effects on immune cell populations in blood and spleen. TOCP treatment at 400 mg/kg BW/d reduced white blood cell counts by 18%

In silico and in vivo toxicity assessment of cysteamine-modified nanoparticles: implications for pharmacotherapy application.

BACKGROUND: Given the increasing therapeutic potential of cysteamine (CYST) at appropriate doses and expert concerns regarding the toxicity of nanoparticles, this study aimed to assess the toxicity profile of both CYST and silver nanoparticles conjugated with cysteamine (CYST-AgNPs). METHODS: For the acute study, a 300 mg/kg starting dose of CYST (i.p administration) produced a toxic response in some mice (n = 3/group), and a 300 mg/kg beginning dose of CYST-AgNPs produced delayed mild toxicity. Lower doses of CYST and CYST-AgNPs (50, 100, and 200 mg/kg; n = 3/group) were administered (i.p) f

TBBPA-BDBPE as a safer alternative? Comparative assessment of systemic toxicity with TBBPA in zebrafish.

Tetrabromobisphenol A (TBBPA) and its derivative, TBBPA-bis (2,3-dibromopropyl ether) (TBBPA-BDBPE), are widely used brominated flame retardants (BFRs) that are frequently detected in environmental and human biological samples. Despite their widespread application, the potential toxicity of TBBPA-BDBPE is still not thoroughly assessed. In this study, we employed zebrafish as a model organism to systematically evaluate and compare the developmental toxicity of TBBPA and TBBPA-BDBPE. Our results demonstrated that TBBPA induced significant developmental toxicity at 1000 μg/L, as evidenced by redu

Acute contact toxicity of insecticides for the chemical control of the invasive yellow-legged hornet Vespa velutina nigrithorax (Hymenoptera: Vespidae).

The yellow-legged hornet, Vespa velutina subs. nigrithorax Buysson, 1905, originally from Southeast Asia, has become an invasive species in Europe since its introduction in France around 2004. Its rapid proliferation and voracious predatory behavior pose a significant threat to native insects, particularly honeybees and other pollinators, impacting agricultural production, biodiversity, and human safety. Eradication in Europe seems now impossible, and the control efforts are hindered by the lack of standardized application protocols, including for insecticide use, leading to potential indiscri

Assessment of ocular irritation potential of moisturizing and sunscreen cosmetic creams marketed in Algeria using the HET-CAM test.

Cosmetic products applied around the eyes, such as moisturizing and sunscreen creams, can come into contact with the conjunctiva, potentially causing irritation due to the thinness of the ocular epithelium. These products, widely used by the general population, remain in contact with the skin for extended periods. In this study, 41 cosmetic creams marketed in Algeria (moisturizers and sunscreens) were evaluated using the Hen's Egg Test on the Chorioallantoic Membrane (HET-CAM), an alternative to the Draize test. The results showed that 36.6 % of the tested creams induced irritation, with varyi

A quantitative weight-of-evidence method for confidence assessment of adverse outcome pathway networks: A case study on chemical-induced liver steatosis.

The field of chemical toxicity testing is undergoing a transition to overcome the limitations of in vivo experiments. This evolution involves implementing innovative non-animal approaches to improve predictability and provide a more precise understanding of toxicity mechanisms. Adverse outcome pathway (AOP) networks are pivotal in organizing existing mechanistic knowledge related to toxicological processes. However, these AOP networks are dynamic and require regular updates to incorporate the latest data. Regulatory challenges also persist due to concerns about the reliability of the informati

Development of a zebrafish model for toxicity evaluation of adulterated Apis mellifera honey.

Since ancient times, honey has been used for medical purposes and the treatment of various disorders. As a high-quality food product, the honey industry is prone to fraud and adulteration. Moreover, limited experimental studies have investigated the impact of adulterated honey consumption using zebrafish as the animal model. The aims of this study were: (1) to calculate the lethal concentration (LC50) of acid-adulterated Apis mellifera honey on embryos, (2) to investigate the effect of pure and acid-adulterated A. mellifera honey on hatching rate (%) and heart rate of zebrafish (embryos and la

Evaluation of temperature- and ethanol-related developmental degree variations by a new scoring system (FETAX-score) applicable to Frog Embryo Teratogenicity Assay: Xenopus.

The aim of the present work is to propose a new quantitative assessment method (FETAX-score) for determining the degree of Xenopus laevis embryo development intended for use in embryotoxicity studies. Inspired by a similar scoring system used to evaluate developmental delays (young-for-age phenotypes) in rat embryos cultured in vitro, the FETAX-score was established by considering seven morphological features (head, naris, mouth, lower jaw, tentacles, intestine, anus) that are easily evaluable in tadpoles during the late stages of development at the conclusion of the test. Given that X. laevis
