The recent development of analytical models of trypanosomiasis has increased the general applicability of models to the strategic control of the disease. An analysis of detailed data on tsetse abundance and infection rates and trypanosome prevalence in village-based trypanotolerant cattle over 4 years in The Gambia showed that seasonal patterns of abundance in Glossina morsitans-infested areas were consistent, and that the rates of trypanosome infection remained relatively unchanging. However, there were two distinct seasonal trypanosome prevalence patterns in cattle, with peaks occurring eith…

A detailed study of the combined effect of suramin, MK 436 (5-nitroimidazole) and Cymelarsan (melaminylthioarsenate) was investigated in the murine CNS-trypanosomiasis model. Using a combination of Cymelarsan and MK 436 without preliminary suramin, complete cures were obtained with a 2 day treatment of 15.0 mg/kg MK 436 and 5.0 mg/kg Cymelarsan. If the MK 436 was reduced to 10 mg/kg then 4 consecutive daily treatments were required. If the Cymelarsan was reduced to 2.5 mg/kg either a 3 day treatment in combination with 15.0 mg/kg or a 4 day treatment with 10 mg/kg MK 436 was necessary. Utilizi…

The inhabitants of three Liberian villages in a known gambiense sleeping sickness area with a low prevalence were examined comparatively with the haematocrit centrifugation technique (HCT), the miniature anion exchange centrifugation technique (m-AECT), the indirect fluorescent antibody test (IFAT) and the Testryp CATT (Card Agglutination Test for Trypanosomiasis). Among 418 persons examined was one individual from whom trypanosomes could be isolated with the HCT and m-AECT. This positive case and another 21 individuals had positive titres of 1:40 and more in the IFAT. The same number, but not…

Camels from herds in four different areas of Kenya were sampled at regular intervals over periods of 9-15 months and examined for the presence of trypanosomiasis. Five hundred and eight camels divided into three age categories were studied. Trypanosomiasis caused by T. evansi, T. brucei and T. congolense was shown to be present in varying degrees in all the herds. Camels were divided into 5 types according to mortality, the presence or absence of circulating trypanosomes and antibody, and the presence or absence of emaciation and anaemia. Herd profiles were constructed according to the percent…

Trypanosoma congolense releases a number of hemolytic free fatty acids on autolysis of which the most potent is linoleic acid. These fatty a … vivo also confirm that this mechanism of erythrocyte destruction is probably unimportant in bovine trypanosomiasis.

An enzyme-linked immunosorbent assay (ELISA) for the serodiagnosis of human African trypanosomiasis (sleeping sickness) is described. A crude extract of a Trypanosoma brucei suspension which was purified from all blood components was used as antigen. In rabbits experimentally infected with T. brucei or T. rhodesiense both homologous (anti-T. brucei) and heterologous (anti-T. rhodesiense) Trypanosoma antibodies could be detected with ELISA using T. brucei as antigen. The sensitivity of ELISA was comparable with that of the immunofluorescence (IF) technique. Sera of patients with sleeping sickne…

The pathological features of 14 cases of human African trypanosomiasis (HAT) occurring in Uganda over an 8 year period are described. Three cases were clinically proven and in all 14 cases a chronic meningoencephalitis was found. In 2 cases there was histological evidence of ganglion radiculitis and in one of these chronic choroiditis and peripheral neuritis associated with chronic myositis were present. The cardiac lesions consisted of a chronic pancarditis of varying degree in 8 cases and in 3 a generalized valvulitis was observed. In 2 cases, specially investigated, generalized lesions of t…

It was postulated that the anemia commonly seen in mammalian trypanosomiasis, and specifically in Trypanosoma congolense-infected calves, was of immunological origin. Specifically, we postulated that trypanosome antigen-antibody-complement complexes, deposited on the surface of erythrocytes of infected calves, resulted in their immune elimination leading to clinical anemia. This hypothesis was tested experimentally. Immunoglobulins bound to the erythrocytes of 13 infected calves were detected by a direct antiglobulin test from 7 to 10 days post infection. The reaction was strongest between 3 a…

Strategies to detect Human African Trypanosomiasis (HAT) cases rely on serological screening of populations exposed to trypanosomes. In Guinea, mass medical screening surveys performed with the Card Agglutination Test for Trypanosomiasis have been progressively replaced by door-to-door approaches using Rapid Diagnostic Tests (RDTs) since 2016. However, RDTs availability represents a major concern and medical teams must often adapt, even in the absence of prior RDT performance evaluation. For the last 5 years, the Guinean HAT National Control Program had to combine three different RDTs accordin…

OBJECTIVES: A critical step before treatment of human African trypanosomiasis (HAT) is the correct staging of the disease. As late stage is established when trypanosomes cross the blood-brain barrier and invade the central nervous system, we hypothesized that matrix metalloproteinases and cell adhesion molecules could indicate, alone or in combination, the disease progression from the first to the second stage of HAT.
METHODS: We measured the levels of MMP-2, MMP-9, ICAM-1, VCAM-1 and E-selectin in the cerebrospinal fluid (CSF) of 63 Trypanosoma brucei gambiense-infected patients (15 stage 1 …

African trypanosomiasis (AT), also known as sleeping sickness in humans and Nagana in animals, is a disease caused by the protozoan parasite Trypanosoma brucei. AT is an extremely debilitating disease in human, cattle, and wild animals, and the treatment is difficult with frequent relapses. This work shows that BALB-c mice immunized intramuscularly with a single dose (100 microg) of a plasmid DNA encoding the 5'-terminal region of the trans-sialidase (nTSA) gene of T. brucei brucei are able to produce IgG antibodies that bind to the bloodstream form of T. brucei-protein extract and recognize t…

The serological and parasitological tests used for Trypanosoma brucei gambiense human African trypanosomiasis (HAT) diagnosis have low specificity and sensitivity, respectively, and in the field, control program teams are faced with subjects with positive serology but negative parasitology who remain untreated. The aim of this work was to explore, using PCR tool, the significance of these aparasitemic serological suspects. Since discordant PCR results have been observed earlier with different extraction methods, two DNA extraction methods were compared (the Chelex 100 resin and the DNeasy Tiss…

We have previously demonstrated that human African trypanosomiasis (sleeping sickness) at the stage of meningoencephalitis results in a major disruption of the circadian rhythmicity of sleep and wakefulness that is proportional to the severity of the disease. This paper examines the corresponding 24-hourly secretion in cortisol and prolactin and compares it with the hourly distribution of sleep composition in infected patients and healthy African subjects. The secretion of cortisol in humans follows a circadian rhythm relatively independent of the sleep-wake cycle, whereas that of prolactin ex…

Alpha-difluoromethylornithine (DFMO, eflornithine) is a specific irreversible inhibitor of ornithine decarboxylase, shown to be curative in various Trypanosoma species infections of animals. In the present open study, the efficiency of DFMO was assessed in 7 patients (4 Africans, 3 Europeans) with Trypanosoma brucei gambiense (Tbg) infection, 4 in the advanced stage and 3 in the early phase of the disease. Treatment with DFMO at initial dosages ranging 300-500 mg/kg/day administered IV (except 1 case) for 10-15 days, followed by 200-300 mg/kg/day per os for 28-69 days was associated with clear

Trypomastigotes attached to the wall of the hypopharynx in tsetse flies infected with Trypanosoma vivax are believed to represent the true metacyclic stage of this trypanosome. Electron microscopy demonstrates that attachment is mediated by hemidesmosome-like junctions along the flagellar membrane and that none of the trypomastigotes, either attached or free in the hypopharynx lumen, possesses a surface coat comparable with that on the metacyclics of T. brucei and T. congolense and on the bloodstream stages of all salivarian trypanosomes. As the variable antigen of bloodstream and metacyclic T
