Optimization of In-Situ Exosome Enrichment Methodology On-a-Chip to Mimic Tumor Microenvironment Induces Cancer Stemness in Glioblastoma Tumor Model.

Understanding cancer etiology requires replicating the tumor microenvironment (TME), which significantly differs from standard in vitro cultures due to nutrient limitations, acidic pH, and oxidative stress. To address this, a microfluidic bioreactor (µBR) with an expanded culture surface was designed to optimize exosome enrichment and glioblastoma cell behavior. Using response surface methodology (RSM), key parameters-including medium exchange volume and interval time-were optimized, leading to about a six-fold increase in exosome concentration without artificial inducers. Characterization tec…

Tumor-Infiltrating Lymphocytes and Tumor-Stroma Ratio on Early-Stage Cervix Carcinoma: Prognostic Value of Two Distinct Morphological Patterns of Microenvironment.

Background Tumor progression is influenced by the complex network of different cellular elements that make up its microenvironment. Tumor-infiltrating lymphocytes (TILs) and stroma characteristics reflect two faces of the intricate mechanisms involved in the tumor-host interaction and can be easily evaluated by routine histological examination. Their prognostic value could be demonstrated in different tumor tumor types, but they are poorly explored in cervical cancer. Methodology In this retrospective study, we analyzed the association of TILs, tumor-stroma ratio (TSR), and pattern of stromal

Tumor Microenvironment-Activated Hydrogel Platform with Programmed Release Property Evokes a Cascade-Amplified Immune Response against Tumor Growth, Metastasis and Recurrence.

In situ tumor vaccines (ITV) have been recognized as a promising antitumor strategy since they contain the entire tumor-specific antigens, avoiding tumor cells from evading immune surveillance due to antigen loss. However, the therapeutic benefits of ITV are limited by obstacles such as insufficient antigen loading, inadequate immune system activation, and immunosuppressive tumor microenvironments (TME). Herein, a tumor microenvironment-activated hydrogel platform (TED-Gel) with programmed drug release property is constructed for cascaded amplification of the anti-tumor immune response elicite…

The construction of CD8+ T cell-associated molecular subtypes in esophageal squamous cell carcinoma reveals tumor heterogeneity, tumor microenvironment, and immunotherapy.

BACKGROUND: Esophageal squamous cell carcinoma (ESCC) is a cancer that is common worldwide. Its morbidity and mortality rates remain high, seriously threatening human health. In the tumor microenvironment (TME), CD8+ T cells undergo a series of dynamic changes that have important implications for tumor progression and the efficacy of immunotherapy. Our research aims to construct novel molecular subtypes through the gene expression profiling of CD8+ T cells during differentiation and to predict the prognostic and therapeutic effects in ESCC patients. METHODS: In the single-cell sequencing (scR…

Single-cell and spatial transcriptome analysis of breast cancer tumor-associated fibroblast heterogeneity and its mediated remodeling of the tumor microenvironment.

Breast cancer (BC) progression is driven by a complex tumor microenvironment (TME), where heterogeneous cancer-associated fibroblasts (CAFs) play pivotal roles in metastasis and therapy resistance. To decipher CAF diversity and spatial interactions, this study integrated single-cell RNA sequencing data from 24 BC samples with spatial transcriptomics. Intercellular networks were mapped using CellChat, spatial single-cell maps were reconstructed with Seurat, CAF developmental origins were traced through Monocle2-based trajectory analysis, and key spatial interactions were validated via immunohis…

The Tumor Microenvironment Affects Circulating Tumor Cells Metastasis and the Efficacy of Immune Checkpoint Blockade in Non-small Cell Lung Cancer.

Lung cancer is one of the most lethal malignancies, with non-small cell lung cancer (NSCLC) being the most common histologic subtype. Metastasis leads to poor prognosis for patients with cancer. Tumor cells leave the tumor lesions, invade the surrounding stroma, and enter the bloodstream as circulating tumor cells (CTCs). The development of CTCs is the beginning of metastasis. The internal environment in which tumor cells grow and survive is called the tumor microenvironment (TME). It includes tumor cells, fibroblasts, immune cells, and the extracellular matrix. The TME is complex and dynamic.…

Atlas of Tumor and Tumor Microenvironment Cells of Lymphovascular Space Invasion (LVSI) in High-Grade Serous Endometrial Adenocarcinoma: A Case Study.

Lymphovascular invasion (LVSI) is defined as the presence of tumor cells within a definite endothelial-lined space (lymphatics or blood vessels) in the organ surrounding invasive carcinoma. The presence of LVI is associated with an increased risk of lymph nodes and distant metastases. Lymphovascular invasion is described as cancer within blood or lymph vessels and is an independent risk factor for metastasis, recurrence, and mortality. This study aims to present the marker-based immunohistological characterization of cells around LVSI in a high-grade adenocarcinoma of the endometrium to build

Prehabilitation as a Biologically Active Intervention Is Associated with the Remodeling of the Pancreatic Tumor-Immune Microenvironment.

Pancreatic ductal adenocarcinoma (PDAC) is highly lethal, and many patients cannot undergo curative surgery due to frailty. Multimodal prehabilitation: combining exercise, nutrition, and psychological support improves functional readiness, but its biological impact on the PDAC tumor microenvironment (TME) is unclear. In this exploratory pilot study, we profiled resected PDAC tissues from prehabilitation-treated patients and matched controls using NanoString GeoMx Digital Spatial Profiling across immune, tumor, and stromal compartments (n = 4). Transcriptomic signatures were analyzed via differ…

Mechanisms and clinical challenges of ICB-mediated remodeling of the tumor microenvironment in nasopharyngeal carcinoma.

Nasopharyngeal carcinoma (NPC) is an Epstein-Barr virus (EBV)-driven malignancy characterized by a profoundly immunosuppressive tumor microenvironment (TME) that severely limits the efficacy of immune checkpoint blockade (ICB). In the modern comprehensive treatment era, the 5‑year overall survival rate for non‑metastatic NPC has reached 80-90%, and the prognosis of newly diagnosed non‑metastatic patients is substantially better than that of more prevalent cancers such as lung and colorectal cancer. Although ICB has improved survival in recurrent/metastatic NPC, clinical benefits are restricted…

CDK8 remodels the tumor microenvironment and promotes resistance to KRASG12D inhibitors and daraxonrasib in PDAC.

Mutations in KRAS are a dominant driver of pancreatic ductal adenocarcinoma (PDAC), with about 50% of patients presenting with KRASG12D mutations. Small molecule inhibitors targeting KRASG12D suppress PDAC; however, the contribution of the tumor microenvironment (TME) to the sustained efficacy of KRASG12D inhibition and mechanisms of resistance to KRASG12D suppression remain to be elucidated. Here, integrated spatial transcriptomics, single-cell RNA sequencing, and CODEX-based spatial proteomics analyses of PDAC mouse models uncover that while KRASG12D inhibition by MRTX1133 initially increase…

The duality of testosterone in glioblastoma: From targeted therapy to modulation of the tumor microenvironment and neuroprotection.

Glioblastoma (GB) remains one of the most aggressive and lethal human tumors, displaying a pronounced predominance among men that suggests a potential involvement of sex hormones, particularly testosterone (TST), in its pathogenesis. The present review summarizes and integrates current evidence regarding the dual role of testosterone in GB biology. On the one hand, testosterone promotes oncogenic progression by stimulating the proliferation, survival, and invasion of cancer stem cells (CSCs) through the activation of androgen receptors (ARs) and subsequent conversion to estradiol, which trigge

Composition and Functional State of T and NK Cells in the Extramedullary Myeloma Tumor Microenvironment.

UNLABELLED: Extramedullary multiple myeloma (EMM) is a high-risk feature of multiple myeloma associated with increased resistance to treatments, including modern immunotherapies, and shorter survival. The composition and functional state of immune cells within the EMM tumor microenvironment (TME) remain poorly understood. Using single-cell RNA sequencing, flow cytometry, and spatial transcriptomics, we revealed significant differences in the EMM TME compared with multiple myeloma bone marrow (BM). T and NK cells were verified as the most abundant immune subsets in the EMM TME. Compared with th…
