A model of the cerebellum generates gait adaptations in a reflex-based neuromusculoskeletal model during split-belt walking.

BACKGROUND: During split-belt treadmill walking, neurotypical humans exhibit adaptations characterized by a gradual decrease in step length asymmetry (SLA) toward or beyond symmetry, whereas individuals with cerebellar damage do not show these motor adaptations. Neuromusculoskeletal simulations may help to better understand individual aspects of the underlying neural control, but are currently incapable of predicting adaptations to the continuous perturbations imposed by split-belt walking. METHODS: We extend a spinal reflex model with a biologically inspired model of the cerebellum, which en

Endotypes of Chronic Rhinosinusitis with Nasal Polyps: Pathology and Possible Therapeutic Implications.

Today, chronic rhinosinusitis (CRS) is a symptomatic disease diagnosed by nasal endoscopy and eventually computed tomography scan, and is treated by pharmacotherapy or, when unsuccessful, by sinus surgery. With the advent of biologics, the diagnostic approach needs to be adjusted to appreciate CRS endotypes, introducing biomarkers, and the therapeutic options will be extended by the application of biologics. Specifically, type 2 immune reactions moved into the focus, similar to asthma, involving innate and adaptive immunity pathways to establish an often severe, persistent disease. The role fo

Diagnosis and management of implant debris-associated inflammation.

Introduction: Total joint replacement is one of the most common, safe, and efficacious operations in all of surgery. However, one major long-standing and unresolved issue is the adverse biological reaction to byproducts of wear from the bearing surfaces and modular articulations. These inflammatory reactions are mediated by the innate and adaptive immune systems.Areas covered: We review the etiology and pathophysiology of implant debris-associated inflammation, the clinical presentation and detailed work-up of these cases, and the principles and outcomes of non-operative and operative manageme

Glutamine Metabolism Supports α cell Mass and Glucagon Secretion.

The liver-α cell axis is a finely tuned biological rheostat that regulates whole body amino acid availability. Pancreatic α cells secrete glucagon that regulates amino acid catabolism through gluconeogenesis and ureagenesis, yet the mechanisms linking amino acid levels to α cell growth and function are not fully understood. Here, we identify glutaminase, the enzyme that catalyzes glutamine catabolism, as a critical α cell regulator. Glutaminase is highly enriched in α cells across species. α cell expression of glutaminase is required for nutrient-dependent mTORC1 activation, suppression of AMP

Mitochondrial Dysfunction promotes Keloid progress through Metabolic Reprogramming: A Narrative Review.

Keloids are a refractory fibroproliferative disorder characterized by excessive extracellular matrix (ECM) deposition and invasive growth beyond the original wound boundaries, severely impairing patients' quality of life. Accumulating evidence highlights that metabolic reprogramming and mitochondrial dysfunction are important in keloid pathogenesis, particularly in regulating the biological behavior of keloid fibroblasts (KFBs). This narrative review synthesizes current research on the interplay between mitochondrial dysfunction and metabolic abnormalities in keloids. Mitochondrial structural

GenAI-Net: A generative AI framework for automated biomolecular network design.

Biomolecular networks underlie both natural biological processes and engineered cellular technologies, from intracellular regulation and ecological dynamics to biomanufacturing, smart therapeutics, and cell-based diagnostics. However, designing chemical reaction networks (CRNs) that implement a desired dynamical function remains a challenging task. Although candidate networks can be evaluated by simulation, the inverse problem of discovering networks from behavioral specifications remains difficult. It requires navigating vast spaces of topologies and kinetic parameters governed by nonlinear a

Application of electrospun piezoelectric nanofibers in wound healing.

Wound healing is essential for maintaining organismal homeostasis and barrier function, posing significant challenges particularly in the management of hard-to-heal wounds such as diabetic ulcers. This review systematically summarizes the functional properties of piezoelectric electrospun biomaterials and their biological mechanisms in promoting wound healing via electrical stimulation (ES). Piezoelectric nanofiber wound dressings fabricated by electrospinning technology offer dual advantages: on one hand, their unique fibrous architecture enables adaptation to irregular wound contours, facili

From oxygen detoxification to peroxide supply: Microbial NADH oxidases in redox physiology and H₂O₂-coupled biocatalysis.

NADH oxidases (NOXs) are flavin adenine dinucleotide (FAD)-dependent flavoproteins widely distributed in bacteria, archaea, and other microorganisms, where they couple NADH oxidation to oxygen reduction and contribute to redox homeostasis, oxygen adaptation, and peroxide metabolism. H₂O-forming NOX has traditionally been preferred for cofactor regeneration, whereas H₂O₂-forming NOX is increasingly recognized as a dual-function catalyst for NAD⁺ regeneration and in situ H₂O₂ production. This review summarizes the catalytic mechanisms, structural basis, and physiological functions of NOX, focusi

[Targeting tumor hypoxia in combined therapeutic strategies in breast and ovarian cancer].

AIM: Tumor hypoxia is a major driver of tumor progression and metastasis. Hypoxic adaptation is regulated by hypoxiainducible factor-1 (HIF-1), which controls the transcription of genes promoting malignant behavior. HIF-1 inhibitors have failed as monotherapies in clinical trials, highlighting the need for combination-based strategies. This study aimed to develop hypoxia-targeted combination therapies for breast and ovarian cancers. METHODS: Using established in vitro and in vivo hypoxia models, we evaluated acriflavine, a HIF-1 inhibitor, in combination with paclitaxel and an epithelial-mese

Quantitative RNA modification mapping by mass spectrometry with isobaric tags and nucleobase fragment analysis.

RNA modifications regulate RNA stability, translation, stress responses, and disease processes, yet their function remains poorly understood due to technical limitations in sequence analysis. Here, we present an RNA-specific isobaric tandem mass tagging (RMT) platform for omic-scale quantitative mapping of RNA modifications. The platform combines RNA-specific tags adapted from proteomics with an end-to-end workflow spanning sample preparation through data processing. Validation using synthetic oligonucleotides and total tRNA from Pseudomonas aeruginosa yielded reproducible quantification, with

3D-Printed Device for Efficient Packing of Semisolid Samples in 3.2-mm Rotors Used in Cryoprobe Systems.

We present a compact, 3D-printed device designed to facilitate the efficient packing of semisolid or highly viscous samples into 3.2-mm rotors compatible with cryogenic solid-state NMR probes. The tool enables sample loading by centrifugation under standard laboratory conditions, significantly improving packing reproducibility and minimizing sample loss. In contrast to previously reported designs for conventional rotors, this device is optimized for the expanded volume and geometrical constraints of 90-μL rotors used in the Bruker CPMAS cryoprobe. A complementary unloading tool is also describ

The kinases Hsl101 and Urk1 regulate metabolism of Cryptococcus neoformans.

Cryptococcus neoformans is a significant human fungal pathogen, particularly concerning for immunocompromised individuals. Key kinases, Hsl101 and Urk1, are critical for crossing the blood-brain barrier, although the specific mechanisms by which they do so remain undefined. In this study, we systematically investigate the impact of HSL101 or URK1 deletion on the proteomic and metabolomic profiles of C. neoformans. By generating a deletion mutant for HSL101, we observed profound alterations in the expression levels of 366 proteins and 421 metabolites, highlighting significant disruptions in key

Biomechanical analysis of axial-radial integrated functional gradient material implants in healthy and osteoporotic bones.

People with osteoporosis, common among middle-aged and elderly individuals, often need dental implants. Titanium implants, though generally safe, can cause problems due to their stiffness, especially in osteoporotic bone, leading to fractures. This study aims to identify gradient types that offer improved biological adaptation. This was achieved by comparing the mechanical properties of four new two-dimensional functional gradient materials (FGMs) implants to those of conventional and one-dimensional FGM implants in healthy and osteoporotic bone models. The new FGM implants, with reduced stiff

Stress-Induced Changes of the Skin: A Narrative Review.

Adaptation to physical and psychological stress is a fundamental biological process aimed at restoring homeostasis and maintaining health. While acute stress responses are developed for short-term survival, chronic stress has increasingly been recognized as an important trigger to a wide spectrum of systemic and dermatological disorders. Stress can exacerbate inflammatory and autoimmune dermatological conditions - such as acne, psoriasis, and atopic dermatitis. This narrative review highlights the function of the hypothalamic-pituitary-adrenal (HPA) axis, neurogenic inflammation, pigmentation

Enhanced harm detection following maternal separation: Transgenerational transmission and reversibility by inhaled amiloride.

Early-life adversities are associated with altered defensive responses. Here, we demonstrate that the repeated cross-fostering (RCF) paradigm of early maternal separation is associated with enhancements of distinct homeostatic reactions: hyperventilation in response to hypercapnia and nociceptive sensitivity, among the first generation of RCF-exposed animals, as well as among two successive generations of their normally reared offspring, through matrilineal transmission. Parallel enhancements of acid-sensing ion channel 1 (ASIC1), ASIC2, and ASIC3 messenger RNA transcripts were detected transg

Disseminated Cryptococcosis Revealing an HIV Infection: A Case Report.

Cryptococcosis is a common fungal infection regarded as a disease of immunocompromised patients with high mortality. Cryptococcosis is usually observed in the central nervous system and lungs. However, other organs may be involved such as skin, soft tissue, and bones. Disseminated cryptococcosis is defined as fungemia or the involvement of two distinct sites. Here, we report the case of a 31-year-old female patient with disseminated cryptococcosis with neuro-meningeal and pulmonary involvement revealing a human immunodeficiency virus (HIV) infection. Chest computed tomography scan showed a rig

Transgenesis in parasitic helminths: a brief history and prospects for the future.

Helminth infections impact the health of hundreds of millions of persons globally and also cause important economic losses in livestock farming. Methodological limitations as well as the low attention given to the study of helminths have impacted biological research and, thus, the procurement of accurate diagnosis and effective treatments. Understanding the biology of helminths using genomic and proteomic approaches could contribute to advances in understanding host-helminth interactions and lead to new vaccines, drugs and diagnostics. Despite the significant advances in genomics in the last d

Structural and evolutionary analysis unveil functional adaptations in the promiscuous behavior of serum albumins.

Promiscuous activities have been related to the capacity to catalyze reactions different from those a protein has evolved to sustain. In this work, we rethought the serum albumin's promiscuous behavior using evolutionary and structural analysis. We found that the cross aldol condensation of acetone and p-formylbenzonitrile is a promiscuous reaction conserved in humans serum albumin and in closely related albumins from other mammals. Evolutionary analysis indicates that the residues involved in this promiscuous reaction are evolving under positive selection, an evolutionary pattern indicating a

Anatomical and histological investigation of the pineal gland in the lake van fish (Alburnus tarichi (Güldenstädt, 1814)).

The pineal gland and melatonin secreted from the gland regulate the biological clock and adaptation to seasonal changes, glucose balance, nutrition and locomotor activities. In this study, the pineal gland of the Lake Van fish was examined anatomically and histologically. The melatonin level secreted from the pineal gland was determined in fish plasma sampled from both lakes and streams during reproduction migration. The pineal gland in the Lake Van fish, as in other teleost fish, is located in the head, under the translucent pineal window, which does not contain many pigment cells. The gland

Genome-wide scan reveals genomic regions and candidate genes underlying direct and maternal effects of preweaning calf mortality in Nellore cattle.

We conducted analysis to estimate genetic parameters and to identify genomic regions and candidate genes affecting direct and maternal effects of preweaning calf mortality (PWM) in Nellore cattle. Phenotypic records of 67,196 animals, and 8443 genotypes for 410,936 SNPs were used. Analysis were performed through the weighted single-step GBLUP approach and considering a threshold animal model via Bayesian Inference. Direct and maternal heritability estimates were of 0.2143 ± 0.0348 and 0.0137 ± 0.0066, respectively. The top 10 genomic regions accounted for 13.61 and 14.23% of the direct and mat
