Mucosal delivery (in ovo and oral) of chitosan nanoparticle-based subunit vaccine enhances resistance against Campylobacter jejuni colonization in broiler chickens.

Campylobacter jejuni infection in poultry presents a significant public health concern due to the consumption of contaminated chicken meat and related products. Vaccination has been proposed as a potential mitigation approach to control infections in chickens and reduce the risk of zoonotic transmission. Yet, no commercial vaccines are currently available for controlling this microorganism in chickens. This study aimed to investigate the efficacy of mucosal delivery (in ovo and oral) of multi-antigen chitosan nanoparticle (CNP)-encapsulated vaccines, including C. jejuni lysate and outer membra

Distal and local mucosal immunization with a BoHV-4-based vector delivering CpHV-1 gD confers protection against intravaginal CpHV-1 challenge in goats.

Effective vaccines against sexually transmitted pathogens must elicit protective immunity at mucosal surfaces, particularly within the female reproductive tract. Caprine herpesvirus 1 (CpHV-1) causes genital disease, reproductive failure, and neonatal mortality in goats and provides a biologically relevant large-animal model for human genital herpesvirus infection. Here, we evaluated a mucosal vaccination strategy based on a bovine herpesvirus 4 (BoHV-4) vector expressing the CpHV-1 immunodominant glycoprotein D (BoHV4-A-gD(cp)gD(106)ΔTK). Goats were immunized via the intravaginal or intranasa

Nasal RNA-scaffold-protein vaccine protects mice from human H5N1 clade 2.3.4.4b virus lethal infection and safeguards against vaccine-unmatched viruses.

BACKGROUND: Recent cases of H5N1 avian influenza spilling over into human populations have raised concerns once again about the unpredictable emergence of zoonotic viral diseases. This phenomenon highlights the persistent threat posed by viruses that jump from animals to humans, underscoring the need for preparedness for antigens that humans have not encountered. Nasal vaccines, which can effectively block transmission at the site of viral entry, represent a promising strategy and should be prioritised for stockpiling to halt zoonotic spillover. METHODS: Here, we immunised a preclinical anima

The Potential Contribution of Dental Foci and Oral Mucositis to Febrile Neutropenia in Patients Treated With Myelosuppressive Chemotherapy for Solid Tumors and Lymphoma.

INTRODUCTION: Febrile neutropenia (FN) is a potential life-threatening complication of myelosuppressive chemotherapy, particularly when induced by infection. There is evidence that FN can originate from the oral cavity, but its contribution to FN is largely understudied in patients treated for solid tumors. The aim of this study was to assess the prevalence of FN in these patients and to evaluate its relation with dental foci and oral mucositis. MATERIAL AND METHODS: A prospective longitudinal observational study was conducted. Patients diagnosed with solid tumors and lymphoma scheduled to be

Quercetin Improves Mitochondrial Function and Inflammation in H2O2-Induced Oxidative Stress Damage in the Gastric Mucosal Epithelial Cell by Regulating the PI3K/AKT Signaling Pathway.

Functional dyspepsia (FD) is one of the most common functional gastrointestinal disorders, the therapeutic strategy of which it is limited due to its complex pathogenesis. Oxidative stress-induced damage in gastric mucosal epithelial cells is related to the pathogenesis and development of FD. Quercetin (Que) is one of the active ingredients of Zhishi that showed antioxidant, antiapoptotic, and anti-inflammatory effects. The aim of this study is to investigate the effect of Que on oxidative stress-induced gastric mucosal epithelial cells damage and its underlying molecular mechanism. The gastri

Synergism of Helicobacter pylori infection and stress on the augmentation of gastric mucosal damage and its prevention with alpha-tocopherol.

Despite evidence that Helicobacter pylori (H. pylori) infection is closely associated with stress in gastric ulcer patients, the underlying mechanism why ulcer recurrence after stress is augmented especially in patients with H. pylori remains unknown. In this study, we found that oxidative stress played a critical role in the augmented mucosal damage provoked by water immersion restraint stress (WIRS) in H. pylori infection and that an antioxidant, alpha-tocopherol, could ameliorate the aggravation of stress-associated gastric mucosal damage. Two hundred forty SD rats were divided into two gro

Role of anti-Trichinella spiralis antibodies and interferon-gamma in the protection against the enteral phase of experimental trichinosis.

Expulsion of adult Trichinella spiralis is the result of a complex immunologically mediated response. Experiments in mice demonstrated that IFN-gamma, released by mesenteric lymph nodes (MLN) during infection play a major role. However, the role of mucosal and serum antibody responses is thought to be limited. Working on the intestinal phase, this study investigated, the role played by antibodies, transferred from vaccinated infected animals, in the immune response in comparison to that obtained by IFN-gamma administration. Transfer of antibodies gave 82.7% protection, while IFN-gamma in a dos

Polymeric Hydrogels as Technology Platform for Drug Delivery Applications.

Hydrogels have become key players in the field of drug delivery owing to their great versatility in terms of composition and adjustability to various administration routes, from parenteral (e.g., intravenous) to non-parenteral (e.g., oral, topical) ones. In addition, based on the envisioned application, the design of bioadhesive or mucoadhesive hydrogels with prolonged residence time in the administration site may be beneficial. For example, hydrogels are used as wound dressings and patches for local and systemic therapy. In a similar way, they can be applied in the vaginal tract for local tre

Polymer-based nanocarriers for vaginal drug delivery.

The vaginal delivery of various drugs is well described and its relevance established in current medical practice. Alongside recent advances and achievements in the fields of pharmaceutical nanotechnology and nanomedicine, there is an increasing interest in the potential use of different nanocarriers for the delivery of old and new pharmacologically active molecules with either therapeutic or prophylactic purposes. Nanosystems of polymeric nature in particular have been investigated over the last years and their interactions with mucosal fluids and tissues, as well as genital tract biodistribu

[Effectiveness of and tolerability to oral desmopressin in the treatment of central diabetes insipidus].

Intranasal desmopressin represents the treatment of choice in Central Diabetes Insipidus. Nevertheless, this route of administration bears some practical disadvantage, linked to either difficult delivering technique, or the status of nasal mucose. The antidiuretic effectiveness of oral desmopressin has been recently demonstrated, both in experimental animals and in man. In our study we compared oral vs. intranasal desmopressin efficacy in 13 patients affected by Central Diabetes Insipidus. The results show that the peroral administration of Desmopressin at a mean dose of 500-600 micrograms/die

The role of neostigmine in development of acute gastric mucosal lesion in dogs with live bacterial flora-induced shock.

The factors that frequently cause the development of acute gastric mucosal lesions (AGML) were studied in 37 mongrel dogs. Bacterial shock was induced by bolus injection of E. coli live bacterial flora (10(10) cells/kg body weight) under parietal cell-stimulated condition by intravenous administration of 0.008 mg/kg per hour of pentagastrin. In addition, 0.01 mg/kg per hour of neostigmine sulfate was administered intravenously during the experiment. Frequent development of AGML was observed in the neostigmine-administered canines without significant changes in the gastric corporal and antral t

Dysbiosis-Associated Change in Host Metabolism Generates Lactate to Support Salmonella Growth.

During Salmonella-induced gastroenteritis, mucosal inflammation creates a niche that favors the expansion of the pathogen population over the microbiota. Here, we show that Salmonella Typhimurium infection was accompanied by dysbiosis, decreased butyrate levels, and substantially elevated lactate levels in the gut lumen. Administration of a lactate dehydrogenase inhibitor blunted lactate production in germ-free mice, suggesting that lactate was predominantly of host origin. Depletion of butyrate-producing Clostridia, either through oral antibiotic treatment or as part of the pathogen-induced d

A Case of Refractory Gastric Cardia Ulcer in Which Marked Elevation of the Surrounding Mucosa was Observed During the Clinical Course.

A 55-year-old man visited our department because of epigastric pain. Upper gastrointestinal endoscopy revealed a small, undermined ulcer in the gastric cardia. He had no history of taking NSAIDs, and was positive for Helicobacter pylori (Hp) infection. After Hp eradication therapy followed by 8 weeks of proton pump inhibitor (PPI) administration, re-endoscopy showed that the ulcer had slightly shrunk without scarring, and the surrounding mucosa was markedly elevated, like in a submucosal tumor. Endoscopic ultrasonography, performed at the same time, showed thickening of the submucosal and musc

Mucosal T cells expressing high levels of IL-7 receptor are potential targets for treatment of chronic colitis.

The IL-7/IL-7R-dependent signaling pathway plays a crucial role in regulating the immune response in intestinal mucosa. Here we demonstrate the pivotal role of this pathway in the development and treatment of chronic colitis. T cells expressing high levels of IL-7R were substantially infiltrated in the chronic inflamed mucosa of TCR alpha-chain knockout mice and IL-7 transgenic mice. Transfer of mucosal T cells expressing high levels of IL-7R, but not T cells expressing low levels of IL-7R, from these mice into recombinase-activating gene-2(-/-) mice induced chronic colitis. Selective eliminat

Role of iron in host-microbiota interaction and its effects on intestinal mucosal growth and immune plasticity in a piglet model.

Iron is an essential trace element for both the host and resident microbes in the gut. In this study, iron was administered orally and parenterally to anemic piglets to investigate the role of iron in host-microbiota interaction and its effects on intestinal mucosal growth and immune plasticity. We found that oral iron administration easily increased the abundance of Proteobacteria and Escherichia-Shigella, and decreased the abundance of Lactobacillus in the ileum. Furthermore, similar bacterial changes, namely an increase in Proteobacteria, Escherichia-Shigella, and Fusobacterium and a reduct

Single-dose infusion of engineered viral receptor binding domain confers rapid and durable protection against viral infection.

Developing both rapid- and long-acting antiviral drugs for single-dose administration can improve medication adherence and protect people at risk of infection. To provide proof of this concept, here, we designed multimerized form of viral receptor-binding domains (RBDs) to immediately occupy viral receptors to block infection and subsequently induce virus-specific protective immunity. We engineered SARS-CoV-2 RBD, enhancing its affinity to ACE2 and immunogenicity through multimerization and Fc modification. A single administration of 4RBD-Fc not only effectively blocked ACE2-dependent SARS-CoV

Influence of absorption enhancers on the pharmacokinetic properties of non-oral beta-lactam-cefpirom using the rabbit (Chinchilla) in vivo model.

The oral application is the application of the first choice for drug administration. A lot of drugs exhibit relatively low bioavailability. This may be caused by binding of the drug in the gastro-intestinal tract, by poor penetration of the intestinal mucose or by highly hydrophilic properties. Therefore, problem drugs were only used for i.v. administration (intravenously) or for i.m. administration (intramuscularly). In the present study, cefpirom was investigated as a model substance. Cefpirom (Cp) is a semi-synthetic amino-2-thiazolyl-methoxyimino cephalosporin. It exhibits highly hydrophil

Inhibition by leukotriene inhibitors, and calcium and platelet-activating factor antagonists, of acute gastric and intestinal damage in arthritic rats and in cholinomimetic-treated mice.

The leukotrienes, platelet activating factor and intracellular calcium have been implicated in the development of gastro-intestinal lesions induced by non-steroidal anti-inflammatory drugs (NSAIDs) but the relative significance of these inflammatory mediators in lesion formation has not been established in sensitive and specific models of gastro-intestinal ulceration. In the present study the effects of drugs affecting 5-lipoxygenase activity, the actions of platelet activating factor and intracellular calcium on the development of gastric and intestinal ulceration induced by NSAIDs were inves
