Comparison of Tricaine Methanesulfonate (MS-222) and Alfaxalone Anesthesia in Zebrafish (Danio rerio).

The research use of zebrafish has risen exponentially over the past decade while anesthetic options have remained largely unchanged.6 ricaine methanesulfonate (MS-222) is widely accepted as an anesthetic for routine husbandry procedures, however it has limitations and safety concerns. 11 A greater variety of effective anesthetic options for surgical procedures would be advantageous for the research community. Adult zebrafish were randomly assigned to one of the following groups (n = 10, 5 males and 5 females): 200 mg/L MS-222; 6-, 10-, 13-, and 16-mg/L alfaxalone, and control. All zebrafish in

[The choice of drugs for caudal anaesthesia in children. An overview].

Caudal anaesthesia is the most frequently used regional technique in paediatric anaesthesia. Caudal in combination with general anaesthesia is usually performed in healthy children (i.e. for herniotomy or hypospadias). Therefore every complication of this method is a catastrophe, even when the incidence of these complications is very low. Some of the documented complications of caudal anaesthesia in children are caused by the local anaesthetic solutions and/or by additives. Thus, the choice of substances for paediatric caudal blocks should minimize the risk associated with the substances used

Molecular aspects of inhalational anaesthetic interaction with excitable and non-excitable membranes.

The interaction of three volatile general anaesthetics (halothane, enflurane and methoxyflurane) with erythrocyte membranes at concentrations causing protection of intact erythrocytes against hypotonic lysis was investigated in the hope of deriving fundamental information regarding the membrane perturbational characteristics of these substances as compared with those of local anaesthetics studied previously. The volatile agents increased the susceptibility of membrane proteins and, to a somewhat lesser extent, of phospholipids to trinitrophenylation of picryl chloride or trinitrobenzenesulfoni

Pain Relief by Analgesic Eardrops: Paradigm Shift in the Treatment of Acute Otitis Media?

Acute otitis media is a common middle ear infection in children with the predominant symptoms of hearing impairment and pain. If antibiotics are given, they need time to have an effect on the inflammation, so the focus is on pain control. For pain management local anesthetics have the advantage of lesser systemic side effects but are still subject to scrutiny. In this review the literature between 2000 and 2020 was systematically searched for investigating studies and recommendation in guidelines against the background of the mode of action. 11 clinical studies, 2 guidelines and 5 reports resp

The treatment of chronic recurrent oral aphthous ulcers.

BACKGROUND: Chronic recurrent oral aphthous ulcers are the most common type of inflammatory efflorescence of the oral mucosa, with a prevalence of 2% to 10% in Caucasian populations. To treat them properly, physicians should know their clinical appearance and course, conditioning factors, underlying causes, and differential diagnosis. METHOD: This review is based on pertinent articles that were retrieved by a selective search in PubMed and in the Cochrane Central Register of Controlled Trials. RESULTS: Hard, acidic, and salty foods and toothpastes containing sodium lauryl sulfate should be a

Fallopian canal dehiscences: a survey of clinical and anatomical findings.

This survey investigates fallopian canal dehiscences in order to assess the risk of encountering an unprotected facial nerve during routine ear surgery. In a prospective non-randomized study, the intraoperative appearance of the facial canal in 357 routine ear operations was compared with 300 temporal bone specimens from 150 autopsies. Intraoperatively, a dehiscence was detected in 6.4% (23/357) of the operations, most frequently at the oval niche region (16/23 cases). The incidence increased with the number of operations (P<0.0002). Cholesteatoma surgery had the highest relative risk (RR 4.6)

Microcatheterization of the cervical epidural space via lumbar puncture: technical note.

OBJECTIVE: Deposition of opiates, corticosteroids, or local anesthetics into the epidural space is useful for the management of painful maladies of the cervical and thoracic spine. We describe a novel technique for epidural medication delivery via an angiographic microcatheter inserted at or below the conus and advanced cephalad under fluoroscopic guidance. Unlike commercial kits used by anesthesiologists, this method uses a radiopaque catheter that can be precisely targeted to the levels of interest. The hazards of direct puncture, such as "wet tap" or injury to the cervical cord, are minimiz

Pediatric psychopharmacology and local anesthesia: potential adverse drug reactions with vasoconstrictor use in dental practice.

Pain management is important when dealing with pediatric dental patients. The use of local anesthetics can be especially challenging for children taking psychotropic medications. The purpose of this paper was to identify pertinent information regarding drug interactions between vasoconstrictor/local anesthetic combinations and medications for the management of psychiatric or behavior disorders in children. Many of the reported interactions are controversial, largely theoretical with very limited clinical evidence, and not well defined. However, when considering the potential for significant in

Studies on the peripheral action of opioids in postoperative pain in man.

Using simultaneous measurements of pain in both limbs after bilateral orthopaedic surgery, peripheral and central components of regionally administered drugs can be separated. Perineural local anesthetic action is readily shown in both upper and lower limbs. Perineural morphine had no effect in ankle blocks, although an effect superior to that of local anesthetics has been claimed in man. Similarly, no effect was seen from morphine or buprenorphine given by intravenous regional administration in the upper limbs, although opioids have shown local antinociceptive action in animal models. There i

Effects of different anesthetics on oscillations in the rat olfactory bulb.

Different types of oscillations in the olfactory bulb (OB), including θ (1 to 4 and 5 to 12 Hz), β (13 to 30 Hz), and γ oscillations (31 to 64 and 65 to 90 Hz), are important in olfactory information processing and olfactory-related functions and have been investigated extensively in recent decades. The awake and anesthetized states, 2 different brain conditions, are used widely in electrophysiologic studies of OB. Chloral hydrate, pentobarbital, and urethane are commonly used anesthetics in these studies. However, the influence of these anesthetics on the oscillations has not been reported. I

Methemoglobinemia due to application of prilocaine during circumcision and the effect of ascorbic acid.

Local anesthesia with prilocaine has become a routine part of ambulatory circumcision procedures. Methemoglobinemia is a rare but potentially lethal complication of local anesthetics. We report the case of a 42-day-old boy who presented with cyanosis after receiving local anesthesia with prilocaine. Methemoglobin level revealed severe methemoglobinemia (methemoglobin=44.5%). His cyanosis promptly resolved after intravenous administration of ascorbic acid. Cases of local anesthetic-induced methemoglobinemia in urology are under recognized. Although the association between prilocaine use and met

Inhibition of bacterial growth by lignocaine in propofol emulsion.

Contamination of propofol, in an emulsion formulation, has been associated with infective complications. Local anaesthetics, some of which are known to have antibacterial properties, are frequently added to the solution to reduce pain on injection. We examined the growth rates of E. coli, S. aureus, S. epidermidis and P. aeruginosa in propofol with and without lignocaine 0.1%-2% after incubation for 2, 5 and 24 hours at 37 degrees C. Growth of microorganisms in each solution was compared by counting the number of colony forming units (CFU). Propofol supported the growth of all microorganisms. A

Evidence-based review of clinical studies on pharmacology (non-anesthetic studies).

We have divided the analysis of pharmacology studies into studies on local anesthetics and studies on all other pharmacologic agents. This division reflects the large number of studies focusing on methods for obtaining effective anesthesia in patients and permits an independent focus on those clinical studies evaluating nonanesthetic drugs. In this section, we review that alternative class of agents, with an emphasis this year on clinical studies evaluating oral analgesics.

Diplopia as an uncommon complication of cataract surgery.

Two patients are presented with diplopia after cataract extraction performed under local anesthesia. Since there was no spontaneous recuperation after several months, strabismus surgery was performed. The first patient presented a restriction of the inferior rectus muscle probably caused by the retrobulbar injection of anesthetics. The second patient had a decompensation of a congenital superior oblique palsy. A survey of the different etiologies of diplopia after cataract surgery is given.

Phospholipase C activation by anesthetics decreases membrane-cytoskeleton adhesion.

Many different amphiphilic compounds cause an increase in the fluid-phase endocytosis rates of cells in parallel with a decrease in membrane-cytoskeleton adhesion. These compounds, however, do not share a common chemical structure, which leaves the mechanism and even site of action unknown. One possible mechanism of action is through an alteration of inositol lipid metabolism by modifying the cytoplasmic surface of the plasma membrane bilayer. By comparing permeable amphiphilic amines used as local anesthetics with their impermeable analogs, we find that access to the cytoplasmic surface is ne
