Hippocampus in the symptom architecture of schizophrenia spectrum disorders: A network analysis approach.

BACKGROUND: Hippocampal volume reductions are among the most robust neuroanatomical findings in schizophrenia spectrum disorders (SSD). However, how these structural alterations relate to core functional domains such as cognition, sensorimotor functioning, and impulsivity remains largely unclear. To address this gap, we applied network analysis to elucidate the complex interplay between hippocampal volume, cognition, neurological soft signs (NSS) and impulsivity, while controlling for overall psychopathology and global functional status. METHODS: We included 204 SSD individuals (mean age: 38.

Self-similar architectures for pressure-tolerant and mechanically durable superamphiphobic coatings.

Superamphiphobic coatings promise broad utility but continue to face a fundamental challenge: architectures that resist liquid impalement are intrinsically fragile, undermining mechanical durability under realistic service conditions. Here, we report a phase-separation-assisted spray-coating strategy that overcomes this trade-off by constructing self-similar hierarchical micro-/nanostructures. Non-solvent-induced phase separation of a fluorosilicone adhesive, coupled with the self-assembly of fluorinated silica nanoparticles, embeds superamphiphobicity uniformly throughout the coating bulk rat…

Hist2Cell: Deciphering fine-grained cellular architectures from histology images.

Histology images offer a cost-effective approach to predicting cellular phenotypes using spatial transcriptomics. However, existing methods struggle with individual gene expression accuracy and lack the capability to predict fine-grained transcriptional cell types. We present Hist2Cell, a vision graph-transformer framework to accurately resolve fine-grained cell types directly from histology images. Trained on human lung and breast cancer datasets, Hist2Cell predicts cell-type abundance with high accuracy (Pearson correlation over 0.80) and captures cellular colocalization. Moreover, it genera

Stabilizing Cu-Based Photocathodes: From Interfacial Engineering to Advanced Architectures.

Copper-based oxides, including Cu2O, CuO, CuBi2O4, CuFeO2 and CuFe2O4 have emerged as promising photocathode materials for solar-driven photoelectrochemical (PEC) reduction reactions such as hydrogen evolution (HER), carbon dioxide reduction (CO2RR), and nitrogen reduction (NRR). Their appeal lies in the combination of their earth-abundance, low toxicity, and suitable optoelectronic properties. However, the practical deployment of these materials is hindered by their intrinsic instability under operating conditions, primarily due to photocorrosion, interfacial charge recombination, and limited

Genetic architecture of the limbic white matter microstructure in aging and Alzheimer's Disease.

INTRODUCTION: Limbic white matter (WM) abnormalities are prevalent in aging and Alzheimer's disease (AD), but genetic drivers are unclear. METHODS: In 2614 older adults (mean age ± SD: 73.7 ± 9.8 years; 26% cognitively impaired) from seven harmonized cohorts enriched for cognitive impairment, we quantified free-water-corrected diffusion MRI (dMRI) metrics in seven limbic tracts. We estimated single nucleotide polymorphism (SNP) heritability, performed cohort genome-wide association studies (GWASs) with meta-analysis, evaluated shared genetic architecture and enriched pathways, and assessed AD…

RamanOmics decodes the spatial vibrational-molecular architecture of senescence in aging and repair.

Aging and tissue repair involve heterogeneous remodeling across transcriptional, biochemical and cellular dimensions, yet prevailing definitions rely on isolated molecular markers that obscure how these states co-evolve. Here we present RamanOmics, a multimodal framework integrating label-free hyperspectral Raman imaging with single-nucleus RNA sequencing and spatial transcriptomics to link biochemical states with transcriptional programs at single-cell spatial resolution. Applied to young and old mouse lung and skin, RamanOmics reveals tissue-specific programs: lung senescent cells are enrich

Single-cell analysis of the epigenome and 3D chromatin architecture in the human retina.

Most genetic risk variants linked to ocular diseases are nonprotein coding and presumably contribute to disease through dysregulation of gene expression; however, understanding their mechanisms has been impeded by incomplete annotation of transcriptional regulatory elements across retinal cell types. To address this, we carried out single-cell multiomics assays to investigate gene expression, chromatin accessibility, DNA methylome, and three-dimensional (3D) chromatin architecture in human retina, macula, and retinal pigment epithelium/choroid. We identified 420,824 unique candidate regulatory…

The Metabolic Architecture of Glaucoma: A Unified Framework of Cofactor Failure and Kynurenine Dysregulation.

Glaucoma remains a primary cause of blindness, yet its pathogenesis often extends beyond intraocular pressure (IOP). This review integrates four converging lines of metabolic evidence-aqueous humor (AH) metabolomics, kynurenine pathway (KP) activity, tetrahydrobiopterin (H4BIP) biology, and NAD/one-carbon dysfunction-into a testable framework for retinal ganglion cell vulnerability. By utilizing a systematic AH metabolomics atlas covering glaucoma, pseudoexfoliation, and diabetes on a standardized HILIC-LC-HRMS platform, we demonstrate that, while aromatic amino acid elevations are non-specifi

Multivalent C2-alkyl trihydroxypiperidine architectures modulate β-glucocerebrosidase activity.

Multivalent systems based on C2-alkylated (3R,4R,5R)-3,4,5-trihydroxypiperidines, members of the iminosugar family, were developed as modulators of the lysosomal enzyme β-glucocerebrosidase (GCase), whose deficiency causes Gaucher disease and is also related to Parkinson disease. The iminosugar units were successfully multimerized onto tri- and tetrapodal scaffolds and gold nanoparticles, enabling a systematic investigation of the multivalent effects. Both architectures exhibited a marked enhancement in inhibitory potency compared to a suitable monovalent reference compound, proving a strong m…

The application of ultrastructure expansion microscopy reveals the apical microtubule architecture of Babesia.

Tick-transmitted apicomplexan parasites of the genus Babesia infect vertebrate red blood cells and cause babesiosis in humans and animals. Detailed studies of their subcellular organization have been constrained by their small size and intracellular development. In this study, we report the identification of a previously unrecognized apical tubulin rosette (ATRo) in Babesia divergens - a unique rosette-like microtubule structure located at the apical pole of the parasite. The ATRo consists of 6-7 radial spines and represents a distinct architecture that appears morphologically different from t…

Benchmarking deep learning architectures for hyperspectral in-vivo brain tumor segmentation.

BACKGROUND AND OBJECTIVE: Deep Learning (DL) has become increasingly relevant in medical image analysis, especially for tasks such as surgical guidance. In this respect, Hyperspectral Imaging (HSI) has shown encouraging results for in-vivo brain tumor segmentation during surgeries, where DL models can benefit from spectral information. The literature includes a large collection of Neural Network (NN) architectures designed to process spatial and spectral information in HS images. However, most of these architectures have been developed outside the healthcare domain, and comparative studies on …

Unveiling Shared Genetic Architectures and Causality: Intestinal Diseases and Neurological Diseases.

BACKGROUND: The "gut-brain axis" provides a theoretical foundation for the connection between intestinal and neurological diseases, but whether this reflects a shared genetic etiology or causal relationships exist remains unclear. METHODS: We used genome-wide association study summary data from FinnGen and UK Biobank to investigate the genetic correlations and causal relationships between three intestinal diseases and six neurological diseases. RESULTS: We observed positive global genetic correlations between irritable bowel syndrome and epilepsy (rg = 0.429, p = 1.53 × 10-2), and stroke (rg

Linking Cell Architecture to Mitochondrial Signaling in Neurodegeneration: The Role of Intermediate Filaments.

Mitochondrial dysfunction is a pivotal contributor to neurodegeneration. Neurons heavily rely on mitochondrial oxidative metabolism and therefore need highly efficient quality control mechanisms, including proteostasis, mitochondrial biogenesis, fusion-fission dynamics, and mitophagy, to sustain bioenergetics and synaptic function. With aging, deterioration of mitochondrial quality control pathways leads to impaired oxidative phosphorylation, excessive reactive oxygen species generation, calcium imbalance, and defective clearance of damaged organelles, ultimately compromising neuronal viabilit
