Synthesis of Vanillin derivative Schiff's bases from threonine, and valine: Structural study and multiscale computational modeling.

Addressing the escalating global challenge of antimicrobial resistance, this study reports the synthesis, comprehensive structural characterization, and multiscale computational modeling of two novel vanillin-derived Schiff's bases, 1 (from threonine) and 2 (from valine). Spectral analyses confirmed their successful formation. Antimicrobial assays revealed significant activity against a panel of Gram-negative (Escherichia coli, Pseudomonas aeruginosa), Gram-positive (Bacillus subtilis, Staphylococcus aureus), and fungal (Candida albicans, Aspergillus flaves) strains. Notably, compound 1 exhibi…

Steam explosion pretreatment coupling high-temperature short-time sterilization facilitating cellulose degradation and sporulation-regulatory gene expression in high-solid fermentation.

Steam explosion coupling high-temperature short-time sterilization (SE-HTST) was exploited to modify cellulosic biomass medium properties an … al thermal sterilization (CTS), respectively. SE-HTST increased the viable cell and spore number of Bacillus subtilis by 1.8 and 1.6 times at 72 h of fermentation compared to CTS. Moreover, the expressions of master transcriptional gene spo0A and the early sigma factors of sigF and sigE genes gradually increased in the SE-HTST medium, showing enhanced sporulation i …

Silica-coated graphene compared to Si-CdSe/ZnS quantum dots: Toxicity, emission stability, and role of silica in the uptake process for imaging purposes.

Quantum dots (QDs) present a special type of nanocrystals (NCs) due to their unique optical and chemical properties. While cadmium-based QDs … emission inside the cytoplasmic portion of the liver hepatocellular carcinoma cell line (HepG2) and Bacillus subtilis (B. subtilis), but they revealed lower photoluminescence (PL) intensity compared to Si-CdSe/ZnS NCs although G-Si QDs are advantageous in other aspects, i.e. possess lower toxicity and higher stability with temperature variations.

Antibacterial effects of antiretrovirals, potential implications for microbiome studies in HIV.

BACKGROUND: Despite being used by more than 18 million people our understanding of the extent of the effects of antiretrovirals on the human body and other organisms remains incomplete. In addition, the direct effect of antiretrovirals on the gut microbiota of HIV-infected individuals has been largely overlooked in microbiome studies concerned with HIV-infected individuals. METHODS: Here we tested 25 antiretrovirals on Bacillus subtilis and Escherichia coli using a broth microdilution assay to assess whether these drugs have an antibacterial effect. RESULTS: We found that several widely used…

BACKGROUND: To evaluate the antibacterial activity of 21 plant essential oils against six bacterial species. METHODS: The selected essential oils were screened against four gram-negative bacteria (Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa, Proteus vulgaris) and two gram-positive bacteria Bacillus subtilis and Staphylococcus aureus at four different concentrations (1:1, 1:5, 1:10 and 1:20) using disc diffusion method. The MIC of the active essential oils were tested using two fold agar dilution method at concentrations ranging from 0.2 to 25.6 mg/ml. RESULTS: Out of 21 e…

Purification and characterization of Heteroscorpine-1 (HS-1) toxin from Heterometrus laoticus scorpion venom.

Crude venom from Thai giant scorpion, Heterometrus laoticus, most commonly found in the northeastern area of Thailand, was evaluated for PD50 (paralytic dose 50) activities from abdominal injection to cricket (Gryllus sp.) and activities against various kinds of microorganisms. It exhibited good results in disc diffusion assay for Bacillus subtilis, Klebsiella pneumoniae, Pseudomonas aeruginosa and Staphylococcus aureus. After purification, toxin showed acceptable results for PD50 determination in entrapped cricket as well as inhibitory activity against B. subtilis, K. pneumoniae, and P. aerug…

RqcH and RqcP catalyze processive poly-alanine synthesis in a reconstituted ribosome-associated quality control system.

In the cell, stalled ribosomes are rescued through ribosome-associated protein quality-control (RQC) pathways. After splitting of the stalled ribosome, a C-terminal polyalanine 'tail' is added to the unfinished polypeptide attached to the tRNA on the 50S ribosomal subunit. In Bacillus subtilis, polyalanine tailing is catalyzed by the NEMF family protein RqcH, in cooperation with RqcP. However, the mechanistic details of this process remain unclear. Here we demonstrate that RqcH is responsible for tRNAAla selection during RQC elongation, whereas RqcP lacks any tRNA specificity. The ribosomal pr…

Exploring the Exclusive Isolation of Pseudomonas syringae in Peltigera Lichens via Metabolite Analysis and Growth Assays.

The specific association of the potentially plant-pathogenic Pseudomonas syringae with Peltigera lichens raises questions about the factors … compounds, such as aminoglycosides, may facilitate P. syringae presence in Peltigera by inhibiting Bacillus subtilis and other antagonists. In addition, compounds absent in Peltigera, like anthracene, might serve as a carbon source inhibitor like Bacillus velezensis.

Anti-influenza and anti-COVID-19 drugs promote bacterial antibiotic cross-resistance through drug-specific adaptive pathways.

The extensive use of antiviral drugs for COVID-19 and influenza has increased their release into clinical and environmental settings, yet their potential to promote bacterial antibiotic cross-resistance remains poorly understood. Here, we investigated the antibacterial off-target effects of four clinically used anti-IFV/COV drugs-baloxavir acid (BXA), favipiravir (FAV), nirmatrelvir/ritonavir (NMV/r), and oseltamivir phosphate (OSV)-and their potential to influence bacterial stress responses and antibiotic cross-resistance in Escherichia coli, Bacillus subtilis, and Pseudomonas aeruginosa PAO1…
