Membrane protein complexes are critical to many cellular processes and have proven to be good targets for a variety of therapeutics. One such complex is the bacterial chemoreceptor signaling complex, which has potential as a target for novel antibiotics. This complex is responsible for sensing chemical signals in the environment to bias swimming of the bacterial cell toward more favorable conditions. While these complexes are well studied, the mechanisms of signal transduction and kinase control are still not fully understood. We have applied multiple thermal stability measurements and limited

SGK1 (serum/glucocorticoid-regulated kinase 1) is an AGC family serine/threonine kinase with a pronounced inducible activation profile; its expression and activation are governed by hormonal stimuli and cellular stress. When PI3K-AKT signaling is constrained, SGK1 can function as an alternative node that sustains cell survival and stress adaptation. Its dysregulation has been linked to tumor drug resistance, salt-sensitive hypertension, cardiovascular disease, fibrosis, and neurodegenerative disorders, supporting its potential as a therapeutic target. As understanding of SGK1 biology and struc

Barrier tissues-epithelial and endothelial interfaces that compartmentalize the human body-govern molecular exchange, immune surveillance, and organ homeostasis. Their dysfunction is central to disorders ranging from dermatitis to neurodegeneration. Conventional static cultures fail to capture the relevant microenvironment and typically rely on cell lines that overlook patient-specific genetics. Organs-on-chips (OoCs), by contrast, can recapitulate barrier-specific flow, biomechanics, chemical gradients, and a multicellular architecture. Additionally, incorporating primary or induced pluripote

We report a crown-ether-assisted topotactic ion-exchange reaction in the β-pyrochlore oxide KOs2O6, enabling complete extraction of K+ ions under mild aqueous conditions without degradation of the rigid Os-O cage framework. Treatment with 18-crown-6 yields a hydrated protonated phase formally described as (H3O)Os2O6, which is inaccessible by conventional acid-mediated ion-exchange routes. Combined synchrotron X-ray and neutron diffraction analyses show that the cubic Fd3̄m symmetry is preserved after ion exchange. The exchanged phase is characterized by protons bonded to cage oxygen atoms and

Dexmedetomidine has been widely used clinically for perioperative sedation and analgesia, accompanied by the shortcomings of cardiovascular adverse effects and short duration. Although developing a slow-releasing dexmedetomidine formulation is a conventional strategy to address these problems, it is unfortunately limited by the small molecule and high solubility of the drug. In this study, we designed a self-assembling peptide (SAP) by referring to the chemical structure of dexmedetomidine, which contains a pH-responsive imidazole group and a hydrophobic benzene group. By also containing the t

Controlled fabrication of twisted van der Waals heterostructures is essential to unlock the full potential of moiré materials. However, achieving reproducibility remains a major challenge, particularly for air-sensitive materials such as Bi2Sr2CaCu2O8 + δ (BSCCO), where it is crucial to preserve the intrinsic and delicate superconducting properties of the interface throughout the entire fabrication process. Here, a dry, inert and cryogenic assembly method is presented that combines silicon nitride nanomembranes (NMBs) with pre-patterned electrodes and the cryogenic stacking technique (CST) to

Crohn's disease (CD) remains a highly complex disorder, and the progress of preclinical gene therapy for CD has been constrained by several significant challenges. These include the identification of optimal therapeutic gene targets, the difficulty of targeting therapy-resistant cells within a chronic inflammatory microenvironment, particularly in the enteric nervous system (ENS), and the lack of robust animal models that faithfully recapitulate human pathology, as classical models largely rely on toxin-induced colitis. This review synthesizes major preclinical studies on gene therapy for CD a

Cardiovascular diseases are major causes of morbidity and death worldwide, and most cardiac deaths are related to ischemic injury of the myocardium (myocardial infarction). As underlined in the current clinical definition and classification of myocardial infarctions, not all myocardial injuries are due to ischemia: irreversible injury, ending in necrosis, can be induced also by various other factors, such as infections, immune disorders, physical and chemical agents, and trauma. This is supported by clinical studies showing that elevated serum levels of cardiac troponins, as a measure of myoca

This study offers promising insights into the anti-obesity potential of Pleurotus ferulae, an edible mushroom valued in Asian cuisine for its nutritional benefits. A hot water extract of P. ferulae (PWE) administered to high-fat diet-induced obese mice over an 8-week period significantly reduced their body weight gain and fat accumulation. PWE not only improved the body weight metrics but also positively influenced the serum lipid profile of obese mice by lowering their total cholesterol and low-density lipoprotein cholesterol levels. In vitro studies using 3T3-L1 adipocytes showed that PWE in

Chronic myeloid leukemia (CML) is recognized as a classic clonal myeloproliferative disorder. Given the limited treatment options for CML patients in the accelerated phase (AP) and blast phase (BP), there is an evident need to develop new therapeutic strategies. This has the potential to improve outcomes for individuals in the advanced stages of CML. A promising therapeutic target is Wilms' tumor 1 (WT1), which is highly expressed in BP-CML cells and plays a crucial role in CML progression. In this study, a chemically synthesized nucleus-targeting WT1 antagonistic peptide termed WIP2W was iden

Tetrabutylammonium bromide (TBAB) is a widely used industrial reagent and is commonly found in our aquatic ecosystem as an industrial byproduct. In humans, the ingestion of TBAB causes severe neurological impairments and disorders such as vertigo, hallucinations, and delirium. Yet, the extent of environmental risk and TBAB toxicity to human health is poorly understood. In this study, we aim to determine the developmental toxicity of TBAB using zebrafish embryos as a model and provide novel insights into the mechanism of action of such chemicals on neurodevelopment and the overall embryonic pro

Fabry disease (FD), a lysosomal storage disorder, is caused by defective α-galactosidase (GLA) activity, which results in the accumulation of globotriaosylceramide (Gb3) in endothelial cells and leads to life-threatening complications such as left ventricular hypertrophy (LVH), renal failure, and stroke. Enzyme replacement therapy (ERT) results in Gb3 clearance; however, because of a short half-life in the body and the high immunogenicity of FD patients, ERT has a limited therapeutic effect, particularly in patients with late-onset disease or progressive complications. Because vascular endothe

Polycystic ovarian syndrome (PCOS) is a complex multifactorial disorder of unknown pathogenesis in which genetic and environmental factors contribute synergistically to its phenotypic expressions. Endocrine-disrupting chemicals (EDCs), a group of widespread pollutants freely available in the environment and consumer products, can interfere with normal endocrine signals. Extensive evidence has shown that EDCs, environmental contributors to PCOS, can frequently induce ovarian and metabolic abnormalities at low doses. The current research on environmental EDCs suggests that there may be link betw

Short stature is a major skeletal phenotype in osteogenesis imperfecta (OI), a genetic disorder mainly caused by mutations in genes encoding type I collagen. However, the underlying mechanism is poorly understood, and no effective treatment is available. In OI mice that carry a G610C mutation in COL1A2, we previously found that mature hypertrophic chondrocytes (HCs) are exposed to cell stress due to accumulation of misfolded mutant type I procollagen in the endoplasmic reticulum (ER). By fate mapping analysis of HCs in G610C OI mice, we found that HCs stagnate in the growth plate, inhibiting t

Parkinson's Disease (PD) is one of the most common neurodegenerative disorders that affects the motor system, and includes cardinal motor symptoms such as resting tremor, cogwheel rigidity, bradykinesia and postural instability. Its prevalence is increasing worldwide due to the increase in life span. Although, two centuries since the first description of the disease, no proper cure with regard to treatment strategies and control of symptoms could be reached. One of the major challenges faced by the researchers is to have a suitable research model. Rodents are the most common PD models used, bu

Chronic inflammatory diseases such as periodontitis have been associated with increased risk for various medical conditions including diabetes and cardiovascular disease. Endotoxin (lipopolysaccharide, LPS), derived from gram-negative periodonto-pathogens, can induce the local accumulation of mononuclear cells in the inflammatory lesion, increasing proinflammatory cytokines and matrix metalloproteinases (MMPs). This ultimately results in the destruction of periodontal connective tissues including alveolar bone. Curcumin is the principal dyestuff in the popular Indian spice turmeric and has sig

This article is part of a Special Issue "Puberty and Adolescence". Puberty is a critical period for brain maturation that is highly dependent on gonadal sex hormones. Modifications in the gonadal steroid environment, via the use of anabolic androgenic steroids (AAS), have been shown to affect brain development and behavior. Studies in both humans and animal models indicate that AAS exposure during adolescence alters normal brain remodeling, including structural changes and neurotransmitter function. The most commonly reported behavioral effect is an increase in aggression. Evidence has been pr

Allergic contact dermatitis results from a T-cell-mediated, delayed-type hypersensitivity immune response induced by allergens. Skin dendritic cells (DCs) play a central role in the initiation of allergic skin responses. Following encounter with an allergen, DCs become activated and undergo maturation and differentiate into immunostimulatory DCs and are able to present antigens effectively to T cells. The frequency of allergic skin disorders has increased in the past decades. Therefore, the identification of potential sensitizing chemicals is important for skin safety. Traditionally, predictiv

Environmental exposures during pregnancy are increasingly recognized as significant risk factors for the offspring health. Perinatal exposure to environmental toxins, including p-Cresol, may disrupt essential physiological processes during critical developmental windows. The perinatal period is particularly critical for neurodevelopment, making the brain particularly vulnerable to external toxins that can impair neural circuitry and increase the risk of behavioral and neuropsychiatric disorders later in life. p-Cresol, a microbial metabolite produced by the human microbiota and through the bac
