[Immune system in women with genital inflammatory diseases caused by Chlamydia and virus infection].

Immunologic studies were carried out in 29 women with clinical manifestations of chronic inflammations of the internal genitals. Dot-hybridization with biotin-labeled DNA probes revealed chlamydial DNA in the cervical cells of 14 women, Herpes simplex type 2 and/or cytomegaloviral DNA in 15 women. Cell-mediated immunity was assessed using monoclonal antibodies CD3, CD4, CD8, and CD19 and by Facscan flow cytofluorometer (Becton Dickinson). Immunoglobulins were measured by nephelometry with Abbott kits and TDx device. No appreciable changes in the mean values of the parameters characterizing T-c

Characteristics of gram-stained cervical smear from patients with Chlamydia trachomatis infection.

Cervical discharges from 142 women attending the Public Gynecologic Service of Araraqura (SESA), Brazil were cultured for Chlamydia trachomatis. Gram-smears and plating on semiquantitative sheep blood agar and chocolate agar were also carried out. An isolation rate of 18% was reported. The presence of purulent cervical secretion was observed in 8 (32%) out of 25 women. It was also observed that a substantial proportion of culture-positive women had no symptoms. Our data demonstrate that screening tests should be based on specific diagnostic techniques for Chlamydia trachomatis since the majori

Isolation of Chlamydia trachomatis among women with symptoms of lower genital tract infection.

Isolation of chlamydia trachomatis from the endocervix using cyclohexamide-treated McCoy cells were done in order to estimate the prevalence rate of its infection among gynecologic out patients who had symptoms and/or signs of lower genital tract infection. There were 498 patients from May 1989 to July 1990. Eighty-six per cent of these patients were 25 years old or older. Most of them (63%) were agricultural employees. Ninety-three per cent were married and 78 per cent had less than or equal to seven yrs of education. Of 476 specimens, isolation rate of C. trachomatis was 7.8 per cent. Other

[Studies on otolaryngeal Chlamydia trachomatis infection province (a case report and seroepidemiological study)].

Recently, chlamydial infection patterns have undergone considerable change. Chlamydia trachomatis has, in particular, been described as a major cause of sexually transmitted diseases. Because of the more liberal attitude concerning sexual life-styles and behavior patterns, the oral cavity and pharynx has been increasingly becoming a site of infection for many sexually transmitted diseases. In the otolaryngeal region, because of oro-genital sexual relations, C. trachomatis infection is presently attracting a great deal of attention. The authors report a case of pharyngitis and cervical lymphade

Prevalence of active Chlamydia trachomatis infection at the time of exploratory laparotomy for ectopic pregnancy.

Cultures of the fallopian tube, cul-de-sac, and cervix were obtained from 50 patients undergoing exploratory laparotomy for ectopic pregnancy. Chlamydia trachomatis was cultured from the cervix in four patients (8%). All the cultures from the fallopian tube or cul-de-sac were negative. Immunofluorescent stain of deparaffinized cross-sections of fallopian tube at the site of tubal involvement with the ectopic pregnancy failed to reveal any C trachomatis inclusions.

Adverse foetal outcome in specific IgM positive Chlamydia trachomatis infection in pregnancy.

A study was undertaken to determine the adverse foetal outcome in antichlamydial IgM positive asymptomatic pregnant females. An indirect immunoperoxidase assay was done to detect IgM in 78 apparently normal asymptomatic pregnant women during the third trimester and follow up was done till delivery to study the effect of chlamydial infection on foetal outcome. A total of 28 (35.9%) women were positive for antichlamydial IgM while only 3.33 per cent asymptomatic non-pregnant normal women (controls) were positive. 28.7 per cent IgM positive mothers delivered low birth weight (LBW) babies (P less

Protective immunity in pig-tailed macaques after cervical infection with Chlamydia trachomatis.

Seven pig-tailed macaques were inoculated in the cervix with Chlamydia trachomatis. Cervical infection resulted in all animals. After spontaneous cessation of cervical shedding of C. trachomatis, repeated inoculation either failed to produce infection, or resulted in infection of shorter duration with lower inclusion counts. Thus, cervical infection may lead to protective local immunity in pig-tailed macaques.

Confirmatory assay increases specificity of the chlamydiazyme test for Chlamydia trachomatis infection of the cervix.

Enzyme immunoassays for the detection of chlamydial antigens are commonly used to diagnose Chlamydia trachomatis infection. As is true for all nonculture methods, the specificities of these tests are a concern. A confirmatory blocking assay (Abbott Laboratories, North Chicago, Ill.) was evaluated at four sexually transmitted disease test sites. This assay is designed to confirm true-positive Chlamydiazyme (CZ) specimens and to identify false-positive CZ reactions caused by cross-reacting bacteria. Cervical specimens were collected from 2,891 women. Chlamydia prevalence by tissue culture (TC) w

Diagnosis of Chlamydia trachomatis cervical infection by detection of amplified DNA with an enzyme immunoassay.

A sensitive and specific system for detection of amplified Chlamydia trachomatis DNA from cervical specimens by fluorometric quantitation in an enzyme immunoassay (EIA) format (polymerase chain reaction [PCR]-EIA) is described. The primers selected for PCR-amplified DNA were from the 15 serovars of C. trachomatis and two strains of Chlamydia pneumoniae (TWAR). One strain of Chlamydia psittaci (Borg) was not amplified. One hundred four previously cultured cervical specimens were evaluated. Forty-six culture-positive specimens containing from 1+ to 4+ inclusion bodies were all positive by PCR-EI

[Epidemiological study on Chlamydia trachomatis infection among sexually active female adolescents].

Sexual behaviour in 141 female adolescents, from 15 to 20 (average 17.4) years old, detained in a remedial institution was investigated from an epidemiological viewpoint, with special reference to Chlamydia trachomatis (CT) infection, from April, 1985 to March, 1986. All had experienced sexual intercourse. Nineteen subjects (13.5%) were found to be CT-antigen positive by either cell cultures (16 cases) or immunofluorescence tests (9 cases). The immunotypes of CT belonged to types D-K. CT-antigen was isolated more frequently (25.5%) from girls with uterine cervicitis than from those without inf

Chlamydia trachomatis cervical infection and oral contraceptive use among adolescent girls.

This study examines the relationship between oral contraceptive use and Chlamydia trachomatis cervical infection in women less than 19 years of age. The clinical and epidemiologic data of 73 (19.4% prevalence rate) girls with chlamydial infection were analyzed, with special attention given to contraceptive use. The findings were compared with data from 303 girls who were chlamydia negative to assess the relationship between oral contraceptive use and C. trachomatis infection. Use of an oral contraceptive for 6 months or longer was associated with chlamydial infection (p = 0.005; odds ratio = 2

[Chlamydia trachomatis infection among sexual partners of the patients with non-gonococcal urethritis].

During the past 3 years, 30 sexual partners including 18 married couples and 12 extramarital sexual pairs whose male partners were diagnosed as having non-gonococcal urethritis were examined for Chlamydia trachomatis infection. Twenty-three of the 30 couples (76.7%) had identical results either positive or negative for Chlamydia trachomatis infection. All 3 male partners of the 3 pairs who had the non-identical results for Chlamydia trachomatis infection, male negative and female positive, had history of urethritis or prostatitis. Fourteen of the 17 female partners (82.4%) who were positive fo

The use of intrauterine contraceptive devices, pelvic inflammatory disease, and Chlamydia trachomatis infection.

With the same epidemiologic approach taken in a recent study that suggested that oral contraceptive use may not protect against chlamydial pelvic inflammatory disease, the risks of chlamydial pelvic inflammatory disease were evaluated for intrauterine contraceptive device users. Compared with women using no method of contraception, intrauterine contraceptive device users were not found to be at any higher risk of cervical chlamydial infection. Whether this places intrauterine contraceptive device users at no increased risk of chlamydial pelvic inflammatory disease cannot be ascertained from th

Historical and clinical factors associated with Chlamydia trachomatis genitourinary infection in female adolescents.

Five hundred sixty-eight adolescent female patients receiving routine gynecologic care at urban clinics were screened by culture for Chlamydia trachomatis infection at both the urethra and endocervix. Culture results for 562 were available from either or both sites. Positive cultures were obtained from 139 (25%). Urethral infection was not associated with either urinary tract symptoms or sterile pyuria, but urethral or endocervical infection was associated with cervical friability (P = less than 0.0001), endocervical mucopus (P = 0.0001), cervical erythema (P = 0.0002), and cervical ectopy or

Selective screening for Chlamydia trachomatis infection in nonurban family planning clinics in Wisconsin.

To determine the prevalence of, and identify risk factors for, Chlamydia trachomatis infection, we studied 380 women attending four Wisconsin family planning clinics in October 1985. The patients completed self-administered sexual history questionnaires, were examined by nurse clinicians and had specimens taken for direct fluorescent antibody (DFA) testing for C. trachomatis. Of 335 women with adequate specimens, 10.7 percent had positive DFA tests. Selective screening criteria were developed based on the following risk factors for C. trachomatis: Age less than 20 years and recent exposure to

Systemic Chlamydia trachomatis infection in mice: a comparison of lymphogranuloma venereum and trachoma biovars.

We developed a murine model of systemic infection with Chlamydia trachomatis biovar lymphogranuloma venereum (LGV). The pathological features of this infection resemble those of human LGV infection since both are characterized by granuloma formation. Mice developed resistance to reinfection with LGV, and this resistance was based on cellular immune mechanisms since it was transferable with immune spleen cells but not with immune serum. Resistance required viable organisms for induction. We compared LGV biovar infection with trachoma biovar infection. Trachoma biovar produced similar but less m

Persistent infection of mouse fibroblasts (McCoy cells) with a trachoma strain of Chlamydia trachomatis.

An in vitro model of persistent infection of mouse fibroblasts (McCoy cells) with a trachoma strain (G17) of Chlamydia trachomatis has been developed. Persistently infected cultures were established by infecting McCoy cells with high multiplicities of chlamydiae. After the first cycle of chlamydial replication, the host cells multiplied more rapidly than the parasites, so that the fraction of inclusion-bearing cells declined to less than 1%. However, after 100 days, the proportion of inclusion-bearing cells rose dramatically, and the cultures alternated between periods of massive host cell des
