Site-specific O-glycosylation of members of the low-density lipoprotein receptor superfamily enhances ligand interactions.

The low-density lipoprotein receptor (LDLR) and related receptors are important for the transport of diverse biomolecules across cell membranes and barriers. Their functions are especially relevant for cholesterol homeostasis and diseases, including neurodegenerative and kidney disorders. Members of the LDLR-related protein family share LDLR class A (LA) repeats providing binding properties for lipoproteins and other biomolecules. We previously demonstrated that short linker regions between these LA repeats contain conserved O-glycan sites. Moreover, we found that O-glycan modifications at the

A mutation in Ampd2 is associated with nephrotic syndrome and hypercholesterolemia in mice.

BACKGROUND: Previously, we identified three loci affecting HDL-cholesterol levels in a screen for ENU-induced mutations in mice and discovered two mutated genes. We sought to identify the third mutated gene and further characterize the mouse phenotype. METHODS: We engaged, DNA sequencing, gene expression profiling, western blotting, lipoprotein characterization, metabolomics assessment, histology and electron microscopy in mouse tissues. RESULTS: We identify the third gene as Ampd2, a liver isoform of AMP Deaminase (Ampd), a central component of energy and purine metabolism pathways. The cau

[Niacin in therapy].

Niacin (nicotinic acid and nicotinamide) is a vitamin used as a source of the NAD+ and NADP+ coenzymes required for many metabolic processes. Its low dietary levels induce the development of pellagra. Niacin has been used for decades in the treatment of patients with disturbed lipid and lipoprotein metabolism, this being the main cause of atherosclerotic changes in cardiovascular diseases. It is still the most efficacious drug in terms of its ability to increase HDL cholesterol content accompanied by a decrease in all atherogenic lipoproteins (VLDL, LDL, and L(a)) as well as fatty acids and tr

Letting lipids go: hormone-sensitive lipase.

PURPOSE OF REVIEW: Despite their pathophysiological importance, the molecular mechanisms and enzymatic components of lipid mobilization from intracellular storage compartments are insufficiently understood. The aim of this review is to evaluate the role of hormone-sensitive lipase in this process. RECENT FINDINGS: Hormone-sensitive lipase exhibits a broad specificity for lipid substrates such as triglycerides, diglycerides, cholesteryl esters, and retinyl esters and the enzyme is in a wide variety of tissues. The high enzyme activity in adipose tissue was considered rate-limiting in the degra

Protective effects of a soy diet in preventing obesity-linked renal disease.

BACKGROUND: Hypertension, hyperlipidemia, hyperfiltration, hyperinsulinemia, glomerular hypertrophy, and ultimately glomerular injury and renal failure are associated with obesity in the Zucker rat. Evidence from other laboratories suggests that soy protein might offer renal protection. METHODS: At five weeks of age obese rats were placed on diets containing either soy or casein as a protein source and studied until 24 weeks of age. At six weeks of age and every four weeks thereafter, 24-hour urine collections were obtained along with measurements of systolic blood pressure (tail cuff) and bl

[Diet and coronary disease].

The build up of cholesterol in the atheromas is caused by, among other things, a disequilibrium of certain plasmatic lipoproteins. Food components which modify the levels of these lipoproteins can be considered as "atherogenic". Other components influence platelet activity and can be considered "thrombogenic". The saturated fatty acids, C-12, C14 and C-16, are atherogenic because they increase LDL plasmatic levels when consumed in large quantities. Estearic acid would not be included in this group because it is easily transformed into oleic acid in the organism. However, all the above acids ar

Postprandial lipoproteins and progression of coronary atherosclerosis.

The relations between triglyceride-rich lipoproteins, alimentary lipaemia and coronary heart disease (CHD) have remained obscure and much debated. We studied the basal and postprandial plasma levels of chylomicron remnants and very low density lipoproteins (VLDL) of varying particle size in 32 male postinfarction patients (mean (S.D.) age 48.8 (3.2) years) and in 10 age-matched control men. The selective quantification of postprandial intestinal and hepatic lipoproteins was accomplished by determining apolipoproteins B-48 and B-100 in lipoprotein subfractions of Svedberg flotation (Sf) rates >

Increased de novo lipogenesis and delayed conversion of large VLDL into intermediate density lipoprotein particles contribute to hyperlipidemia in glycogen storage disease type 1a.

Glycogen storage disease type 1a (GSD-1a) is a metabolic disorder characterized by fasting-induced hypoglycemia, hepatic steatosis, and hyperlipidemia. The mechanisms underlying the lipid abnormalities are largely unknown. To investigate these mechanisms seven GSD-1a patients and four healthy control subjects received an infusion of [1-(13)C]acetate to quantify cholesterogenesis and lipogenesis. In a subset of patients, [1-(13)C]valine was given to assess lipoprotein metabolism and [2-(13)C]glycerol to determine whole body lipolysis. Cholesterogenesis was 274 +/- 112 mg/d in controls and 641 +

Transfer of dehydroepiandrosterone- and pregnenolone-fatty acid esters between human lipoproteins.

Dehydroepiandrosterone (DHEA)- and pregnenolone (PREG)-fatty acid esters (FA) are formed in plasma high density lipoproteins (HDL), whereas they accumulate in very low density lipoproteins (VLDL), low density lipoproteins (LDL), and HDL. We have hypothesized that these lipoidal steroids could be transferred from HDL to VLDL and LDL by the cholesteryl ester (CE) transfer protein (CETP), which mediates CE transfer activity in human plasma. In this study, we further investigated this hypothesis. Lipoproteins and lipoprotein-deficient plasma (LPDP) were purified and analyzed by Western blots. LPDP

Serum lipids and proteins in lactose malabsorption.

It has been suggested that dietary lactose may reduce the intestinal absorption of fat and protein in individuals with lactase deficiency. On the other hand, it is known that a high carbohydrate diet increases serum lipids. The purpose of this study was to examine whether there are differences in the fasting serum lipid and protein concentrations between people with lactose malabsorption and people with normal lactose absorption. Therefore in the connection of a family study serum lipids and proteins were measured in 409 subjects belonging to 11 families. Of these 288 were relatives of the 11

Beneficial antithrombotic effects of the association of pharmacological oral magnesium therapy with aspirin in coronary heart disease patients.

The use of magnesium in the treatment of acute myocardial infarction remains controversial despite preliminary experimental evidence that magnesium plays a beneficial role as a regulator of thrombosis. The aim of our study was to determine whether oral magnesium treatment inhibits platelet-dependent thrombosis (PDT) in stable patients with coronary artery disease (CAD). In a randomized prospective, double-blind, cross-over and placebo controlled study, 42 patients with stable CAD (37 men, 5 women, mean age 68 +/- 9 years) on aspirin received either magnesium oxide tablets (800-1,200 mg/day) or

Exploring Serum Vitamin D, Sex Hormones, and Lipid Profile Disparities in Women With and Without Polycystic Ovarian Syndrome: A Case-Control Study.

Background Polycystic ovarian syndrome (PCOS) is a common endocrine disorder affecting women of reproductive age. It is characterized by dyslipidemia, hormonal imbalances, and metabolic dysfunctions. Vitamin D deficiency may be implicated in the pathogenesis of PCOS, potentially exacerbating its metabolic syndrome. However, the exact interplay between these factors remains underexplored. Aim This study aimed to evaluate serum levels of vitamin D and its association with modalities of PCOS among women with PCOS and healthy controls.  Methods This was a hospital-based case-control study where 60

Pre and post fibrate supplementation lipoprotein electrophoresis in hyperlipemic dogs with biliary tree disease.

Hyperlipemia is commonly observed in dogs with biliary tree disease, where cholestasis can both cause and result of hyperlipidemia. Fibrate therapy effectively reduce lipemia in dogs with both primary and secondary hyperlipidemia. This study aimed to investigate whether fenofibrate therapy could alter lipoprotein patterns in hyperlipemic cholestatic dogs. Retrospective cohort study on left-over frozen-stored (-80 °C) serum samples of 25 client-owned dogs with chronic biliary disease with hyperlipidemia (hypercholesterolemia and/or hypertriglyceridemia) that underwent fenofibrate therapy at 4-1

Lysophosphatidylcholine as a mechanistic and therapeutic nexus in atherosclerotic cardiovascular disease.

Atherosclerotic cardiovascular disease (ASCVD) remains the leading cause of morbidity and mortality worldwide despite intensive lipid-lowering therapy. Residual cardiovascular risk persists even in patients achieving optimal lipoprotein-cholesterol (LDL-C) levels, indicating that additional lipid mediators contribute to disease progression. Lysophosphatidylcholine (LPC), a bioactive lysophospholipid generated from phosphatidylcholine by cytosolic or lipoprotein-associated phospholipase A₂ (PLA₂), has emerged as a critical mediator linking dyslipidemia, inflammation, and vascular injury. LPC is

MiR-124 Inhibits Lipid Deposition in Mouse Liver by Targeting the Trib3/Hnf4α Pathway.

Non-alcoholic fatty liver disease (NAFLD) arises from dysregulated lipid homeostasis, encompassing imbalances in lipid uptake, synthesis, and catabolism in liver. Despite its global prevalence and clinical impact, effective therapeutic strategies for NAFLD remain elusive. A previous study showed that miR-124 inhibits adipogenic differentiation of murine 3T3-L1 cells by targeting the glucocorticoid receptor (GR). Furthermore, the liver-to-body weight ratio and plasma cholesterol levels were significantly elevated in miR-124 promoter KO mice compared to wildtype controls. Therefore, this study a

Metabolomics revealed novel associations between multiple ambient air pollutants exposure and substance use disorders: Evidence from a prospective cohort study.

BACKGROUND: Substance use disorders (SUDs) constitute a significant global public health challenge. Although air pollution is a recognized risk factor for various mental and behavioral disorders, its prospective relationship with SUDs and the underlying mechanisms involved remain inadequately elucidated. METHODS: This prospective cohort study included 216,737 participants from the UK Biobank. An air pollution score (APS) was constructed based on NO2, NOx, PM2.5, PM2.5-10 and PM10 concentrations. An APS-related metabolic signature was derived using elastic net regression from 168 plasma metabo
