Ultrastructural morphology and domain structure of a unique collagenous component of basement membranes.

A disulfide cross-linked collagenous fragment (7 S) has been isolated by pepsin solubilization from several tissues rich in basement membranes including bovine lung, human placenta, and the murine EHS tumor. Examination of this material by the rotary shadowing technique indicates that these fragments are similar to but not identical with the 7S collagen described recently [Risteli, J., Bächinger, H.P., Engel, J., Furthmayr, H., & Timpl, R. (1980) Eur. J. Biochem. 108, 239-250]. The central rodlike portion of the particles was found to be similar in length; however, the peripheral four arms of

Pediatric pulmonary hemorrhage observed in non-vascular and vascular Ehlers-Danlos syndrome.

BACKGROUND: Ehlers-Danlos syndrome (EDS) is a heterogeneous group of heritable connective tissue disorders with varying features depending on the EDS subtype. EDS is associated with various respiratory manifestations. Pulmonary hemorrhage has been previously reported in vascular EDS (vEDS); however, this manifestation remains not particularly well-defined in other subtypes of EDS. This study extends the clinical understanding of EDS, particularly non-vascular EDS, and expands etiological spectrum of pulmonary hemorrhage in children. METHODS: We retrospectively analyzed the records of patients

Novel serological neo-epitope markers of extracellular matrix proteins for the detection of portal hypertension.

BACKGROUND: The hepatic venous pressure gradient (HVPG) is an invasive, but important diagnostic and prognostic marker in cirrhosis with portal hypertension (PHT). During cirrhosis, remodelling of fibrotic tissue by matrix metalloproteinases (MMPs) is a permanent process generating small fragments of degraded extracellular matrix (ECM) proteins known as neoepitopes, which are then released into the circulation. AIM: To investigate their potential as plasma markers for detection of PHT. METHODS: Ninety-four patients with alcoholic cirrhosis and 20 liver-healthy controls were included. Clinica

Evolution of the schistosomal hepatic lesions in mice after curative chemotherapy.

Mice with 64-day-old Schistosoma mansoni infection (+/- 27 worms, 8-12 pairs) were treated simultaneously with oxamniquine and hycanthone. The cure rate was 100%, and changes occurring thereafter in the liver were sequentially followed by means of histologic, ultrastructural, and immunofluorescence methods. Soon after treatment, hepatitic changes cleared up and periovular granulomas diminished in size. The predominant Type III collagen in granulomas was reduced, and the Type I showed no apparent increase, whereas Type IV did not seem to participate in the process. Collagen fibrils in periovula

Pulmonary vascular Ehlers-Danlos syndrome with hemoptysis as the main manifestation: CT and histologic findings of lung parenchymal damage.

BACKGROUND: Vascular Ehlers-Danlos syndrome (vEDS) is a rare inherited connective tissue disease caused by mutations in the COL3A1 gene. The disease can cause fatal complications such as rupture of the arteries, uterus, and intestine, as well as pulmonary complications, including spontaneous pneumothorax and hemoptysis. Since hemoptysis in vEDS is rare and often misdiagnosed, this study aims to summarize its clinical features, CT findings and the diagnosis and treatment process, thereby improving clinical understanding. METHODS: Patients with vEDS presenting with hemoptysis treated at the Fir

Is Exon Skipping a Viable Therapeutic Approach for Vascular Ehlers-Danlos Syndrome with Mutations in COL3A1 Exon 10 or 15?

Vascular Ehlers-Danlos syndrome or Ehlers-Danlos syndrome type IV (vEDS) is a connective tissue disorder characterised by skin hyperextensibility, joint hypermobility and fatal vascular rupture caused by COL3A1 mutations that affect collagen III expression, homo-trimer assembly and secretion. Along with collagens I, II, V and XI, collagen III plays an important role in the extracellular matrix, particularly in the inner organs. To date, only symptomatic treatment for vEDS patients is available. Fibroblasts derived from vEDS patients carrying dominant negative and/or haploinsufficiency mutation

The relative amounts of the collagen chains alpha 1(I), alpha 2 and alpha 1(III) in the skin of 31 patients with osteogenesis imperfecta.

1. The relative amounts of type III and type I collagen, determined as the ratio of their constituent alpha 1(III) and alpha 1(I) chains, have been measured by interrupted electrophoresis of pepsin extracts of skin collagen from 31 patients with osteogenesis imperfecta of varying severity, and from six clinically unaffected family members. In 18 patients the ratio of the alpha 1(I) to alpha 2 chains of type I collagen has also been measured. 2. In the 15 patients with osteogenesis imperfecta classified as mild or the 18 with type I disease the ratio alpha 1(III)/alpha 1(I) was significantly in

Aggressive change of a carotid-cavernous fistula in a patient with Ehlers-Danlos syndrome type IV.

The authors report a rare case of a carotid-cavernous fistula (CCF) secondary to Ehlers-Danlos syndrome (EDS) type IV which showed an aggressive angiographical change.A 59-year-old woman presented with headache, right pulsatile tinnitus, and diplopia on the right side. The diagnostic angiography demonstrated a right CCF. Accordingly transarterial embolization of the fistula was attempted 5 days later. The initial right internal carotid angiography showed an aneurysm on the petrous portion of the internal carotid artery (ICA) which was not recognized in the diagnostic angiography. Spontaneous r

Consistent linkage of dominantly inherited osteogenesis imperfecta to the type I collagen loci: COL1A1 and COL1A2.

The segregation of COL1A1 and COL1A2, the two genes which encode the chains of type I collagen, was analyzed in 38 dominant osteogenesis imperfecta (OI) pedigrees by using polymorphic markers within or close to the genes. This was done in order to estimate the consistency of linkage of OI genes to these two loci. None of the 38 pedigrees showed evidence of recombination between the OI gene and both collagen loci, suggesting that the frequency of unlinked loci in the population must be low. From these results, approximate 95% confidence limits for the proportion of families linked to the type I

Congenital nephrosis of the Finnish type (CNF): matrix components of the glomerular basement membranes and of cultured mesangial cells.

Congenital nephrosis of the Finnish type (CNF) is a hereditary renal disease of unknown aetiology manifested by massive proteinuria of the newborn and unresponsive to any treatment. In this study kidney samples and cultured glomerular mesangial cells from five patients with CNF were studied by indirect immunofluorescence microscopy for the presence and location of major basement membrane matrix (GBM) components. Histological changes of glomeruli ranging from mild thickening of basement membranes to total obliteration and sclerosis were seen. Notably, thickening of the subepithelial layer of Bo

Myofibroblast expression in skin wounds is enhanced by collagen III suppression.

Generally speaking, the excessive expression of myofibroblasts is associated with excessive collagen production. One exception is seen in patients and animal models of Ehlers-Danlos syndrome type IV in which the COL3A1 gene mutation results in reduced collagen III but with concurrent increased myofibroblast expression. This paradox has not been examined with the use of external drugs/modalities to prevent hypertrophic scars. In this paper, we injected the rabbit ear wound model of hypertrophic scarring with two doses of a protein called nAG, which is known to reduce collagen expression and to

Health-related quality of life in children with osteogenesis imperfecta: a large-sample study.

UNLABELLED: In this large-sample study, we demonstrated that osteogenesis imperfecta (OI) significantly impaired the quality of life (QoL) in children. Moderate/severe OI patients had worse QoL scores than patients with mild OI. Furthermore, the QoL for OI patients was correlated with the presence of pathogenic gene mutations. INTRODUCTION: Osteogenesis imperfecta (OI) is a hereditary disease characterized by multiple fragility fractures and progressive skeletal deformities. No detailed investigations about the quality of life (QoL) have been carried out in a large sample of patients with OI.

Y-position cysteine substitution in type I collagen (alpha1(I) R888C/p.R1066C) is associated with osteogenesis imperfecta/Ehlers-Danlos syndrome phenotype.

The most common mutations in type I collagen causing types II-IV osteogenesis imperfecta (OI) result in substitution for glycine in a Gly-Xaa-Yaa triplet by another amino acid. We delineated a Y-position substitution in a small pedigree with a combined OI/Ehlers-Danlos Syndrome (EDS) phenotype, characterized by moderately decreased DEXA z-score (-1.3 to -2.6), long bone fractures, and large-joint hyperextensibility. Affected individuals have an alpha1(I)R888C (p.R1066C) substitution in one COL1A1 allele. Polyacrylamide gel electrophoresis (PAGE) of [(3)H]-proline labeled steady-state collagen

Impaired pyridinoline cross-link formation in patients with osteogenesis imperfecta.

Patients with osteogenesis imperfecta (OI) show various degrees of bone fragility. Nevertheless, details of the mechanisms causing bone fragility remain unclear. We hypothesized that differences in pyridinoline cross-link formation at the N-and C-termini in type I collagen molecules partly contribute to bone fragility of OI. To verify this hypothesis, urinary N and C terminal telopeptides of type I collagen (uNTx and ubetaCTx, respectively) and urinary hydroxyproline (uHyp) were measured using second morning void urine samples obtained from OI patients and healthy control children. Ratios of u

Borrelia burgdorferi BmpA is a laminin-binding protein.

The Borrelia burgdorferi BmpA outer surface protein plays a significant role in mammalian infection by the Lyme disease spirochete and is an important antigen for the serodiagnosis of human infection. B. burgdorferi adheres to host extracellular matrix components, including laminin. The results of our studies indicate that BmpA and its three paralogous proteins, BmpB, BmpC, and BmpD, all bind to mammalian laminin. BmpA did not bind mammalian type I or type IV collagens or fibronectin. BmpA-directed antibodies significantly inhibited the adherence of live B. burgdorferi to laminin. The laminin-

The Ehlers-Danlos syndrome, or rather syndromes includes a heterogeneous group of diseases secondary to abnormalities of collagen fibres, for certain of which the specific biochemical defect has been identified. The preponderance of collagen fibers in the body explains the diversity of clinical manifestations which can occur as part of this form of hereditary connective tissue dysplasia. The involvement of the arteries is the most serious manifestation and determine the prognosis of the disease. The rupture of arteries, true vascular catastrophy occurring in young patients, is most characteris
