Mastomys natalensis pretreated with diethylcarbamazine (DEC), levamisole or centperazine were exposed to the standard inoculum of infective larvae of Brugia malayi. Percentage 'take' of infection, duration of prepatent period, course of microfilaraemia and number of adult worms recovered were compared with those of untreated infected Mastomys (control). DEC and centperazine did not alter the 'take' (DEC/centperazine: 100%; control: 88.24%) and the average prepatent period (DEC: 105.44 days; centperazine: 105.18 days, control: 109.20 days). In levamisole pretreated animals on the other hand, th

Female filariae of the species Brugia malayi and Litomosoides carinii were investigated by means of electron microscopy after in vivo treatment with flubendazole. The earliest fine-structure alteration in both species was the disappearance of microtubuli from the intestinal cells as soon as 6 h after treatment. There was no further disintegration of intestinal cells for several days. Microtubuli disappeared from the outer zone of the hypodermal cytoplasm 24 h after treatment. At this time, marked alterations were also observed in the oogonia and in the embryonic cells. Many of these were swoll

The efficacy of ivermectin (Iv) was evaluated against four species of filariae, Litomosoides carinii, Acanthocheilonema viteae, Brugia pahangi and Brugia malayi in Mastomys natalensis. Animals with patent infections, induced with L3 larvae, by intravenous (iv) infusion of the respective microfilariae (Mf) (5 x 10(4) Mf per animal) or by intraperitoneal (ip) route (2 x 10(4) Mf per animal) were used in this study. A single dose of Iv (100 micrograms.kg-1) given subcutaneously (sc) to Mastomys infected with L. carinii or A. viteae resulted in the disappearance of microfilaremia within 2 h of tre

Dot-ELISA assay was compared with standard Sandwich ELISA in detecting parasite antigen in sera from patients with bancroftian filariasis. The same monoclonal antibody and the same serum samples were used in both assays. With Dot-ELISA, 67 of 70 serum samples from microfilaremic patients were positive at a dilution of 1:50. End titers ranged from 1:80 to 1:1 280. While with Sandwich ELISA, 64 of the 70 serum samples were positive at a dilution of 1:10. End titers ranged from 1:10 to 1:320. The specificity of both assays was over 91%, but their sensitivity was markedly different. Dot-ELISA coul

It is possible to infect jirds with Brugia pahangi by three methods. Infective larvae (L3) can be injected either intraperitoneally (ip), when adults develop in the peritoneal cavity, or sub-cutaneously (sc), when they develop in the lymphatics or the heart and blood vessels associated with the lungs. Alternatively adult worms which have been grown in the peritoneal cavities of jirds can be implanted into the peritoneal cavities of other jirds. This latter system has been widely used for screening for new filaricides. We have compared the activity of 9 macrofilaricidal compounds against these

To evaluate the merit of antigen detection assays as a tool to monitor the efficacy of chemotherapy for lymphatic filariasis, we serially measured antigen levels in sera from jirds infected with Brugia malayi and from humans with bancroftian filariasis. Antigenemia was detected in all animals with parasitologically proven infection and was present in jirds with prepatent or occult filariasis. Antigen levels correlated with worm burdens, and progressively declined in drug-cured animals. Treatment with diethylcarbamazine (DEC) triggered a transient increase in serum levels of filarial antigens b

Transplanted infections of Dipetalonema viteae and Brugia pahangi have been evaluated as tools for experimental chemotherapy. Attempts were made to establish these filariae in similar pharmacokinetic sites within the same host, so that direct comparisons of in vivo drug susceptibilities could be made. Unfortunately, it was not possible to establish B. pahangi in the subcutaneous tissues, the preferred site of D. viteae. Therefore, intraperitoneal B. pahangi and subcutaneously implanted D. viteae in gerbils were used for the study. D. viteae infections were significantly enhanced by concomitant

Infection with filarial nematodes induces lymphatic dysfunction in many individuals in endemic areas. The present morphometric study describes changes in lymphatic endothelial cells of Brugia-infected cats. Transmission electron microscopy revealed a decrease in the number of cytoplasmic vesicles in endothelial cells from lymphatic vessels harboring filarial nematodes (4.9 vesicles per micron 2 cytoplasm) when compared with similar cells from the contralateral uninfected control vessels (9.0 vesicles per micron 2). The size of the vesicles and their location within the cytoplasm was not change

Lymphatic filariasis (LF) is a mosquito-transmitted parasitic disease, which is a main concern in tropical and subtropical countries. LF is the second major cause of chronic and irreversible disabilities worldwide, which include lymphoedema, hydrocele, and elephantiasis. According to the World Health Organization (WHO), an estimated 882 million individuals across 44 countries were reported to be at risk of acquiring LF. The nematode Wuchereria bancrofti is the predominant pathogen which causes LF, accounting for approximately 90% of filarial infections. The drugs albendazole (ALB), ivermectin …

BACKGROUND: Lymphatic filariasis (LF) affects more than 120 million people globally. In Tanzania, nearly six million people are estimated to … ge recommended by WHO. Absence from home during MDA and perceptions of being free from hydrocele or elephantiasis were the major reasons for non-compliance.
CONCLUSION: There is a significant decline in LF transmission in Masasi District after seven rounds of MDA. However, the presence of individuals who are persistently non-compliant may delay elimination of …

BACKGROUND: Chronic manifestations of bancrofti filariasis can be debilitating. There is paucity of description of this disease in the childhood population, yet early detection can prevent disability such as elephantiasis.
OBJECTIVE: The objective of the study was to determine the prevalence and clinical features of this infection among children in a plantation estate in Cross River State, Nigeria.
METHODS: It was a cross-sectional study in subjects aged 1 to 18 years, recruited from the nine camps of the settlement, using multistage sampling technique. The presence of Circulating Filarial A…

BACKGROUND: Podoconiosis, also known as mossy foot or endemic non-filarial elephantiasis, is a preventable form of lower-leg lymphoedema caused by prolonged (typically barefoot) exposure to soil derived from volcanic rocks. Acute adenolymphangitis (also called 'acute attack') is a serious complication of podoconiosis resulting in significant symptoms and worsening disability. Despite the well-known morbidity associated with podoconiosis, to date there have been no studies looking at the impact, or burden, of podoconiosis on caregivers. This study explored the experiences and impact of acute at…

Over a 1-month period, selected providers in these clinics sought hydrocele, lymphoedema, elephantiasis, or acute adenolymphangitis among the patients seeking treatment. Bancroftian filariasis, a mosquito-transmitted disease commonly known as elephantiasis, is caused by infection with the parasite Wuchereria bancrofti. Infection with this parasite can induce a broad array of chronic debilitating and socially stigmatizing conditions, but the pathogenesis of this morbidity remains obscure. Recent evidence indicates that in filariasis-endemic areas the primary lesion is not lymphatic vessel obstruction but, rather, dilatation. To determine the extent to which lymphatic dilatation occurs in the presence of living adult W. bancrofti, we performed longitudinal ultra…

Lymphatic filarial infection in humans is associated with a strong skewing of the immune response towards the TH2 arm, with prominent interl … ted levels of IgG3 antibodies to Bpa-26 and manifestation of chronic obstructive disease was found; elephantiasis patients showed significantly higher levels of IgG3 antibodies to Bpa-26 than microfilaremics and asymptomatic amicrofilaremics. This indicates that an imbalance of isotypes of antibodies to particular filarial antigens might play a role in the path …
