Targeted determination of volatile fluoroalkyl pollutants and non-targeted screening for environmental monitoring.

Per- and polyfluoroalkyl substances (PFAS) are persistent environmental contaminants known for their toxicity, mobility, and bioaccumulation. Efficient sample preparation and analysis of these compounds are critical for environmental monitoring. In this study, a novel analytical methodology is presented, integrating dynamic headspace extraction (DHS) and thermal desorption (TD) with one-dimensional (1D) and two-dimensional (2D) gas chromatography-time-of-flight mass spectrometry (GC-TOFMS) for the quantification of target volatile and semi-volatile PFAS. Such an approach also enables the non-t

Electrochemiluminescence and electrochemical dual-mode biosensing platform for highly sensitive monitoring of PFOA with RuPCN-224 metal organic framework emitters and aptamer-based sandwich assay.

BACKGROUND: Perfluorooctanoic acid (PFOA) can cause thyroid disorders, immune system dysfunction, and liver diseases and even cancer. Developing sensitive detection methods for trace PFOA in environmental matrices is of vital importance for safeguarding human health. As an emerging technique, electrochemiluminescence (ECL) has been extensively studied for detecting environmental pollutants because of near-zero background and superior detection sensitivity. However, current ECL methods are mainly relied on single-mode readout which can induce false positive and negative results. It is extremely

Perfluorooctanoic acid (PFOA) disrupts cardiac performance of thick-shell mussel by inserting into the binding pocket of proliferator-activated receptor gamma and thereby causing lipid metabolism disorders.

Invertebrates constitute the largest group of animals on Earth, accounting for approximately 97 % of all animal species. Although the heart of invertebrates could be a sensitive target for environmental pollution, potential cardiotoxicity for most contaminants has received little attention. In this study, perfluorooctanoic acid (PFOA) and thick-shell mussels (Mytilus coruscus) were used to investigate the effect of PFOA on cardiac performance and the potential underlying mechanisms. Heart beat monitoring demonstrated that four-week exposure to 0.5 and 5.0 μg/L of PFOA resulted in bradycardia a

Synthesis of Perfluorooctyl-Functionalized Halloysite Nanotubes for Dispersive Solid-Phase Extraction of Antimalarial Inhibitors in Whole Blood.

Quinine, chloroquine, and hydroxychloroquine are effective antimalarial drugs that have shown antiviral activity against SARS-CoV-2 in initial in vitro studies. However, their excessive use can lead to adverse effects in humans. Therefore, rapid and accurate analysis of these inhibitors in whole blood samples is crucial for public health, although this presents significant challenges. In this study, perfluorooctyl-functionalized halloysite nanotubes were synthesized and used as sorbents for dispersive solid-phase extraction. The developed sorbents interacted with inhibitors through various mec

Toxicity assessment of exposure to four short- and ultra-short PFAS using Daphnia magna and Hydra vulgaris as models.

Short- and ultra-short per- and polyfluoroalkyl substances (PFAS) such as perfluorobutanoic acid (PFBA, 4C), perfluorobutane sulfonic acid (PFBS, 4C), trifluoroacetic acid (TFA, 2C) and trifluoromethane sulfonic acid (TFMS, 1C) were detected in various environmental matrices, but studies addressing the effects attributed to exposure are still scarce. This study aimed to (1) investigate acute toxicity of these emerging PFAS using Daphnia magna (immobility, mortality, reproduction, and body size) and Hydra vulgaris (morphological changes and reproduction) and (2) assess the sub-chronic/chronic t

Enhanced Ecological Risk of PFOA Degradation Products: Insights from Concentration-Dependent Transcriptomics, Adverse Outcome Pathways, and Biomarker Verification.

The rapid advancement of perfluorooctanoic acid (PFOA) degradation techniques aims to eliminate PFOA molecules and meet stringent water quality standards. However, the environmental risks of PFOA degradation products have rarely been studied. In this study, PFOA (C8) was photocatalytically degraded with N-CQDs/TiO2, producing C3-C7 perfluoroalkyl carboxylic acids (PFCAs) and fluoride ions. We investigated bioaccumulation and trophic transfer of PFCAs in a model food web of Chlorella sp., Daphnia magna, and Danio rerio (zebrafish). Degradation-induced acidification of the solution increased the

Combining field datasets and mathematical modeling to quantify PFAS leaching and mass discharge at an AFFF-impacted site.

Vadose zones serve as significant reservoirs of per- and polyfluoroalkyl substances (PFAS) at contaminated sites, posing risks to the groundwater underneath. Partitioning of PFAS to the solid-water and air-water interfaces in soils complicates PFAS leaching in the vadose zone. We apply mathematical models representing PFAS-specific retention and transport processes to simulate vadose-zone leaching and mass discharge at a PFAS-contaminated field site. The mathematical models are constrained by detailed datasets collected at the site under both ambient rainfall and artificial flushing conditions

Optimizing electro-oxidation for selective trace micropollutant removal and energy efficiency in secondary effluents.

This study investigated the electro-oxidation (EO) of three priority micropollutants: carbamazepine (CBZ), diuron (DIU), and perfluorooctane sulfonate (PFOS), in secondary-treated wastewater using boron-doped diamond (BDD) anodes. Although BDD anodes generate strong, largely non-selective oxidants; however, their performance at environmentally relevant concentrations, across different classes of micropollutants and under realistic organic-matter loads remains insufficiently characterized, especially for fluorinated and other recalcitrant contaminants. The selected compounds represent persisten

PFOS-induced black liver and hepatic aging in mice: A new paradigm in environmental hepatotoxicology.

Perfluorooctanesulfonic acid (PFOS), a persistent environmental contaminant still imported and widely used in industrial and consumer products, poses poorly defined risks under chronic occupational exposure, with limited research on its role in environmental liver aging. Here, we identify previously unrecognized hepatotoxicity of PFOS in mice exposed to an occupationally relevant dose (10 mg/kg) over subacute (25-day) and chronic (105-day) periods. While both exposures induced mild metabolic-associated steatotic liver disease (MASLD) without fibrosis, only chronic exposure produced a distinct

PFAS-exposed zebrafish embryos show impaired innate immune response after infection with mycobacteria.

The Per- and polyfluoroalkyl substances PFNA, PFOA, PFOS and PFHxS have been regulated in the EU based on their interference with immune system functions. For many more environmental pollutants possible effects on immune function are unknown, so quick screening methods are required to determine which compounds are of highest immunotoxic concern. The aim of the present study was to optimize a test protocol to assess effects of chemicals on the innate immune system using zebrafish (Danio rerio) embryos. In a second step, the protocol was used to determine the immunotoxic effects of 11 PFAS on th

Profiles and transplacental transfer of per- and polyfluoroalkyl substances in maternal and umbilical cord blood: A birth cohort study in Zhoushan, Zhejiang Province, China.

Per- and polyfluoroalkyl substances (PFAS) can pass through the placental barrier and pose health risks to fetuses. However, exposure and transplacental transfer patterns of emerging PFAS remain unclear. Here, 24 PFAS were measured in paired maternal whole blood (n = 228), umbilical cord whole blood (n = 119) and serum (n = 120). Orthogonal partial least-squares discriminant analysis (OPLS-DA) was used to differentiate PFAS between different matrices. The transplacental transfer (TPT) of PFAS was calculated using cord to maternal whole blood concentration ratios. PFOS and PFOA were still the d

Transcriptomic re-analyses of human hepatocyte spheroids treated with PFAS reveals chain length and dose-dependent modes of action.

To identify pathway perturbations and examine biological modes of action (MOAs) for various perfluoroalkyl substances, we re-analyzed published in vitro gene expression studies from human primary liver spheroids. With treatment times ranging from 10 to 14 days, shorter-chain PFAS (those with 6 or fewer fluorinated carbon atoms in the alkyl chain) showed enrichment for pathways of fatty acid metabolism and fatty acid beta-oxidation with upregulated genes. Longer-chain PFAS compounds, specifically PFOS (perfluorooctane sulfonate), PFDS (perfluorodecane sulfonate), and higher doses of PFOA (perfl

Prenatal exposure to per- and polyfluoroalkyl substances, fetoplacental hemodynamics, and fetal growth.

INTRODUCTION: The impact of legacy per- and polyfluoroalkyl substances (PFAS) on fetal growth has been well studied, but assessments of next-generation PFAS and PFAS mixtures are sparse and the potential role of fetoplacental hemodynamics has not been studied. We aimed to evaluate associations between prenatal PFAS exposure and fetal growth and fetoplacental hemodynamics. METHODS: We included 747 pregnant women from the BiSC birth cohort (Barcelona, Spain (2018-2021)). Twenty-three PFAS were measured at 32 weeks of pregnancy in maternal plasma, of which 13 were present above detectable levels

Exploring maternal and developmental toxicity of perfluoroalkyl ether acids PFO4DA and PFO5DoA using hepatic transcriptomics and serum metabolomics.

Production of per- and polyfluoroalkyl substances (PFAS) has shifted from long-chain perfluoroalkyl acids to short-chain compounds and those with ether bonds in the carbon chain. Next-generation perfluoroalkylether PFAS include HFPO-DA ("GenX chemicals"), Nafion Byproducts, and the PFOx homologous series that includes perfluoro-3,5,7,9-butaoxadecanoic acid (PFO4DA) and perfluoro-3,5,7,9,11-pentaoxadodecanoic acid (PFO5DoA). PFO4DA and PFO5DoA have been detected in serum and/or tissues from humans and wildlife proximal to contamination point sources. However, toxicity data are extremely limited

Effects of Perfluorohexane Sulfonate Exposure on Immune Cell Populations in Naive Mice.

Perfluorohexane sulfonate (PFHxS) is a member of the per- and polyfluoroalkyls (PFAS) superfamily of molecules, characterized by their fluorinated carbon chains and use in a wide range of industrial applications. PFHxS and perfluorooctane sulfonate are able to accumulate in the environment and in humans with the approximated serum elimination half-life in the range of several years. More recently, some PFAS compounds have also been suggested as potential immunosuppressants. In this study, we analyze immune cell numbers in mice following 28-d repeated oral exposure to potassium PFHxS at 12, 120

Impact of Gestational Exposure to Individual and Combined Per- and Polyfluoroalkyl Substances on a Placental Structure and Efficiency: Findings from the Ma'anshan Birth Cohort.

Prenatal exposure to perfluoroalkyl and polyfluoroalkyl substances (PFASs) is inevitable among pregnant women. Nevertheless, there is a scarcity of research investigating the connections between prenatal PFAS exposure and the placental structure and efficiency. Based on 712 maternal-fetal dyads in the Ma'anshan Birth Cohort, we analyzed associations between individual and mixed PFAS exposure and placental measures. We repeatedly measured 12 PFAS in the maternal serum during pregnancy. Placental weight, scaling exponent, chorionic disc area, and disc eccentricity were used as the outcome variab
