Genetic Variants and Clinical Characteristics of Young-Onset Parkinson's Disease in the Hakka Population of Western Fujian.

RESEARCH OBJECTIVE: Young-onset Parkinson's disease (YOPD), defined by symptom onset at or before 50 years of age, has a strong genetic component. The mutation spectra vary markedly across ethnic groups. However, YOPD among the Hakka, a subgroup of the Han Chinese ethnicity, remains uncharacterized. We investigated the genetic and clinical profiles of YOPD in the Hakka population of western Fujian. MATERIALS AND METHODS: A total of 33 unrelated patients with YOPD were included in the study. All patients underwent whole exome sequencing (WES) to screen all known Parkinson's disease (PD)-relate

Genetic Analysis Reveals Relationships Among Populations of Puccinia triticina from Henan Province of China.

Wheat leaf rust, caused by Puccinia triticina Erikss. (Pt), is a major foliar disease that poses a significant threat to wheat production in Henan Province, a major wheat-growing region of China. Elucidating the population genetic structure of Pt is critical for predicting pathogen dispersal trends and guiding disease management. In this study, 384 Pt isolates collected from 13 locations in Henan were genotyped using 11 polymorphic simple sequence repeat (SSR) loci. A total of 204 multilocus genotypes (MLGs) were identified. The Sanmenxia (SMX) population exhibited the highest genetic diversit

Identifying genetic causes and establishing a diagnostic approach for WES-negative pediatric population with neurodevelopmental disorder.

Global developmental delay (GDD) and intellectual disability (ID) are frequently caused by genetic factors, yet many patients remain undiagnosed even after whole exome sequencing (WES). This study aimed to apply Optical Genome Mapping (OGM) and Illumina Complete Long Reads (ICLR) in pediatric patients with unexplained GDD/ID after WES and propose a practical diagnostic strategy for clinical implementation. We conducted OGM and ICLR on 87 pediatric patients with unexplained GDD/ID despite prior WES. Discordant cases underwent further validation using gap-PCR or PacBio long-read sequencing. A mi

Causal Effects of Lifestyle and Dietary Factors on Urinary Stone Risk in a Genetic and Population-Based Study.

This study evaluated how lifestyle habits and dietary factors influence urinary stone risk by integrating Mendelian randomization (MR) analysis with data from the National Health and Nutrition Examination Survey (NHANES). Instrumental variables were obtained from genome-wide association study datasets, while urinary stone outcomes were sourced from the FinnGen database. The primary MR analysis used the inverse-variance weighted approach, and additional MR methods verified the robustness of the findings. NHANES data further assessed the relationships between lifestyle characteristics, dietary p

Global, Population and Genetic Evidence on the Relationships Between Immune-Mediated Inflammatory Disease and Cancer Risk.

BACKGROUND: Immune-mediated inflammatory disease (IMID) and cancer share underlying mechanisms. We aimed to comprehensively evaluate the associations between IMIDs and cancers from global, population and genetic perspectives. METHODS: A triangulation framework was employed to assess the association between IMIDs and cancers, using the Global Burden of Disease Study (2012-2021) to analyse six IMIDs and 33 cancers. The UK Biobank (UKBB) prospective cohort was subsequently used to validate these associations, with hazard ratios (HRs) and 95% confidence intervals (CIs) estimated by Cox proportion

Genetic Diversity and Ecological Distribution of Bulinus Populations in Southern Benin Using PCR-RFLP Analysis.

Schistosomiasis remains a major public health concern in Benin. Freshwater snails of the genus Bulinus serve as intermediate hosts for schistosomes, trematode parasites responsible for bilharzia. The urinary form, caused by Schistosoma haematobium, is the most prevalent and is primarily transmitted by Bulinus truncatus and Bulinus globosus, whereas the intestinal form, caused by Schistosoma mansoni, is transmitted by Bulinus forskalii. Effective control strategies require an understanding of the molecular diversity and genetic structure of Bulinus populations. Bulinus samples were collected fr

BACKGROUND: Dermacentor nuttalli is a dominant tick species in northern China and adjacent regions, where it poses significant threats to public health and the livestock industry through the transmission of diverse zoonotic pathogens. Understanding its population genetic structure is crucial for elucidating dispersal patterns and informing control strategies. However, the lack of high-resolution molecular markers has hindered such investigations. This study aimed to develop microsatellite markers and use them to assess the genetic diversity and structure of D. nuttalli populations across north

Genetic association study between rs2234253 (p.T96K) variant of TREM2 and Alzheimer's disease in a Tunisian population.

BACKGROUND: Alzheimer's disease (AD) is the leading cause of major neurodegenerative cognitive impairment. The risk of developing AD is influenced by a complex interaction of genetic predisposition and environmental factors. Among the genetic risk factors, the APOE ɛ4 allele is the most significant, while variants in the TREM2 (Triggering Receptor Expressed on Myeloid Cells 2) and ABCA7 (ATP-binding cassette transporter A7) genes have also been associated with an increased risk of AD. OBJECTIVE: This study aimed to investigate the association of APOE ɛ4, TREM2 gene variants (rs75932628 [p.R47

Spectrum of genetic alterations in patients with peroxisome biogenesis defects in the Iranian population: a case series study.

Peroxisomal disorders are a group of hereditary metabolic disorders that happen when peroxisomes are defective. Around 80% of individuals affected by peroxisomal disorders are classified within the spectrum of Zellweger syndromes with autosomal recessive inheritance pattern that results from mutations in one of the 13 PEX genes. Clinical exome sequencing plays a vital role in the diagnosis where the symptoms are atypical. In the current study, we used this technique to find the underlying genetic cause in 14 Iranian patients with peroxisomal disorders. PEX1 variants were detected in five patie

Population structure, gene flow and genetic diversity of sheep blowfly (Lucilia cuprina dorsalis) in Australia.

BACKGROUND: Flystrike (cutaneous myiasis) is caused by blowfly larvae of the genus Lucilia. This disease is a major obstacle to sustainable global sheep and wool production. Flystrike control relies primarily on breech modification surgery (mulesing) and insecticidal treatment; however, control is constantly compromised by the emergence and spread of insecticide-resistance. Preventing the spread of resistance is severely hindered by a limited understanding of genetic variation, structure and gene flow within and among Lucilia populations. Australia is one of the world's largest producers of sh
