Regulation of gastric hormones by systemic rapamycin.

The mammalian target of rapamycin (mTOR), an evolutionarily conserved serine-threonine kinase, is an intracellular fuel sensor critical for cellular energy homeostasis. Gastrointestinal endocrine cells play a vital role in the regulation of energy balance by secreting hormones that inform the brain about energy supply. Here we showed the localization of mTOR signaling molecules in more than 90% of gastric ghrelin cells and 36±3% of gastrin cells, while no somatostatin-positive cell showed phospho-S6K1 immunoreactivity. Inhibition of mTOR significantly stimulated expression of gastric ghrelin m…

Circulating anti-hypothalamus antibodies in celiac patients: tissue transglutaminase friend or foe?

Celiac disease (CD) is an autoimmune disease with inflammatory characteristics, having a condition of chronic malabsorption, affecting appro … thalamic periventricular neurons by immunofluorescence and by a home-made ELISA assay. In addition, ghrelin was measured by ELISA. As control, 45 blood serums from healthy age matched were analysed. Among active CD, all patients resulted positive for anti-hypothalamus autoantibodies and sera showed significantly higher levels of ghrelin. All of the free-g …

Motion Sickness and Resting Energy Expenditure in Chinese Male Adults.

BACKGROUND: Motion sickness can influence energy homeostasis by enhancing thermolysis. This study tested the hypothesis that resting energy expenditure (REE), as the major component of thermogenesis, might also play a role during motion sickness. METHODS: The effect of seasickness on REE at sea was examined in 71 healthy Chinese male volunteers. Change in REE, heart rate variability (HRV), blood ghrelin levels, and leptin levels were observed across baseline, voyage, and recovery stages. Seasickness severity was assessed using the Graybiel motion sickness questionnaire (GMSQ), and the nausea …

Adipokines as novel biomarkers in paediatric systemic lupus erythematosus.

OBJECTIVES: Patients with SLE are at risk for premature atherosclerosis and metabolic syndrome. Cytokines produced by adipocytes, adipokines, are important in glucose/lipid metabolism and the development of atherosclerosis. The objectives of this study were to evaluate leptin, adiponectin and ghrelin concentrations in paediatric SLE (pSLE) and to correlate these concentrations with measures of disease activity, serum lipid concentrations, measures of insulin resistance and serum homocysteine concentrations. METHODS: Two hundred and eight samples from 105 patients with pSLE and 77 samples from…

Autophagy-driven modulation of stem cell dynamics in Helicobacter pylori-induced gastric diseases in mice.

Chronic Helicobacter pylori infection is a major contributor to gastric disease progression, with its involvement in autophagy and stem cell dynamics playing a critical role in disease mechanisms. This study investigated how H. pylori, particularly in combination with the carcinogen N-nitroso-N-methylurea (NMU), disrupted autophagy and stem cell function, driving gastric pathology. H. pylori infection significantly increased autophagy, promoted the epithelial-mesenchymal transition, suppressed Tff2 and Ghrelin expression in mouse gastric organoids, and enhanced stem cell proliferation (organoi…

Anti-emetic Action of the Brain-Penetrating New Ghrelin Agonist, HM01, Alone and in Combination With the 5-HT3 Antagonist, Palonosetron and With the NK1 Antagonist, Netupitant, Against Cisplatin- and Motion-Induced Emesis in Suncus murinus (House Musk Shrew).

UNLABELLED: Ghrelin has well-known activity to stimulate appetite and weight gain. Evidence suggests that ghrelin may also have effects in reducing chemotherapy-induced emesis via growth hormone secretagogue receptors (GHS-R1A) in the brain. However, it is not known whether the stimulation of GHS-R1A has broad inhibitory anti-emetic effects. In the present studies, we used Suncus murinus to investigate the potential of the new and novel orally bioavailable brain-penetrating GHS-R1A mimetic, HM01 (1-[(1S)-1-(2,3-dichloro-4-methoxyphenyl)ethyl]-3-methyl-3-[(4R)-1-Methyl-3,3-dimethyl-4-piperidyl]…

Glucose intake reduces alcohol craving and amplifies habituation to cue-induced brain activation in male patients with alcohol use disorder: A randomized, placebo-controlled crossover study examining male and female patients with AUD.

INTRODUCTION: Evidence suggests a role of appetite-regulating hormones in alcohol use disorder. Reductions in acylated ghrelin levels are associated with reductions in craving and cue-induced brain activity. Ghrelin levels can be physiologically decreased by glucose intake, which therefore could be a treatment reducing craving and cue-induced brain activity in patients with alcohol use disorder, potentially mediated by acylated ghrelin. MATERIAL AND METHODS: 80 males and females with alcohol use disorder participated in the randomized placebo-controlled crossover study, examining glucose inta…

Chronic treatment with either dexfenfluramine or sibutramine in diet-switched diet-induced obese mice.

Dexfenfluramine (DEX) and sibutramine (SIB) are effective antiobesity agents. Their effects on weight control and hormone profile have not been previously studied in diet-switched diet-induced obese (DIO) mice, in which treatment is initiated upon cessation of a low-fat diet and resumption of a high-fat diet. Furthermore, their effects on circulating ghrelin in obese humans or in animal models of obesity have not yet been reported. Male C57Bl/6J DIO mice after 16 wk on a high-fat diet (HF, 60 kcal% fat) were switched to a low-fat diet (LF, 10 kcal% fat) for 50 d. HF diet resumed concurrently w…

Growth Hormone Secretagogues and the Regulation of Calcium Signaling in Muscle.

Growth hormone secretagogues (GHS) are a family of synthetic molecules, first discovered in the late 1970s for their ability to stimulate growth hormone (GH) release. Many effects of GHS are mediated by binding to GHS-R1a, the receptor for the endogenous hormone ghrelin, a 28-amino acid peptide isolated from the stomach. Besides endocrine functions, both ghrelin and GHS are endowed with some relevant extraendocrine properties, including stimulation of food intake, anticonvulsant and anti-inflammatory effects, and protection of muscle tissue in different pathological conditions. In particular, …

Appetite-preserving gastrectomy attenuates early postoperative deterioration of bone quality after surgery in esophagogastric junction cancer.

BACKGROUND: Bone loss after gastrectomy is a clinically important but often underestimated complication, particularly in elderly patients with gastric or esophagogastric junction (EGJ) cancer. Total gastrectomy (TG) has been associated with postoperative deterioration of bone health. Appetite-preserving gastrectomy (APG) preserves the ghrelin-secreting region of the stomach and maintains appetite and lean body mass; however, its impact on postoperative bone health remains unclear. METHODS: This retrospective observational study included 28 patients with EGJ cancer who underwent curative gastr…

Growth Hormone and IGF-1 Actions in the Brain and Neuropsychiatric Diseases.

Growth hormone (GH) is secreted by the anterior pituitary gland under the control of hypothalamic neuroendocrine neurons that express somatostatin or growth hormone-releasing hormone (GHRH). Ghrelin, originating primarily in the stomach, is also an important GH secretagogue. GH stimulates the hepatic secretion of insulin-like growth factor-1 (IGF-1) and the expression of IGF-1 in extrahepatic tissues, including the brain. Many regions of the brain express receptors for GH, IGF-1, and ghrelin. In recent decades, evidence from both human and animal studies has indicated that GH, IGF-1, and ghrel…
