Development of an In Vivo Retrodialysis Calibration Method Using Stable Isotope Labeling to Monitor Metabolic Pathways in the Tumor Microenvironment via Microdialysis.

Microdialysis is a technique that utilizes a semipermeable membrane to sample analytes present within tissue interstitial fluid. Analyte-specific calibration is required for quantitative microdialysis, but these calibration methods are tedious, require significant technical skill, and often cannot be performed jointly with the experimental measurements. Here, we describe a method using retrodialysis with stable-isotope-labeled analytes that enables simultaneous calibration and quantification for in vivo tumor microdialysis. Isotope-labeled amino acids relevant to immuno-metabolism in the tumor

Novel plasma phospholipid biomarkers of autism: mitochondrial dysfunction as a putative causative mechanism.

Autism is a neurological disorder that manifests as noticeable behavioral and developmental abnormalities predominantly in males between the ages of 2 and 10. Although the genetics, biochemistry and neuropathology of this disease have been extensively studied, underlying causal factors to this disease have remained elusive. Using a longitudinal trial design in which three plasma samples were collected from 15 autistic and 12 non-autistic age-matched controls over the course of 1 year, universal and unambiguous alterations in lipid metabolism were observed. Biomarkers of fatty acid elongation a

SOD1 activity and protective factors in familial ALS patients with L84F SOD1 mutation.

METHOD: The activity and amount of SOD1 in erythrocyte lysates and the plasma amino acid content were evaluated in four familial ALS patients bearing the L84F SOD1 mutation (fALS), in an asymptomatic family member with the mutation (L84F(5)), in sporadic ALS patients (sALS) and controls. Three of the fALS patients and the L84F(5) subject were tested once a year for three consecutive years. RESULTS: At the first evaluation SOD1 activity was similar in controls, sALS and fALS; the amount of SOD1 protein was lower (P < 0.01) in fALS. In the subsequent 2 years, 34% and 52% decrease of SOD1 activi

Cocaine use disorder: A look at metabotropic glutamate receptors and glutamate transporters.

Glutamate transmission is an important mediator of the development of substance use disorders, particularly with regard to relapse. The present review summarizes the changes in glutamate levels in the reward system (the prefrontal cortex, nucleus accumbens, dorsal striatum, hippocampus, and ventral tegmental area) observed in preclinical studies at different stages of cocaine exposure and withdrawal as well as after reinstatement of cocaine-seeking behavior. We also summarize changes in the glutamate transporters xCT and GLT-1 and metabotropic glutamate receptors mGlu2/3, mGlu1, and mGlu5 base

Evidence for a glutamate receptor of the AMPA subtype which mediates insulin release from rat perfused pancreas.

1. The effect of L-glutamate has been studied on insulin secretion by the isolated perfused pancreas of the rat. The glutamate receptor subtype involved has been characterized. 2. In the presence of a slightly stimulating glucose concentration (8.3 mM), L-glutamate (5 x 10(-5)-4 x 10(-3) M) induced an immediate, transient and concentration-dependent insulin response. On the other hand, in the presence of a non stimulating glucose concentration (2.8 mM), L-glutamate (10(-3) M) did not modify the basal insulin secretion. 3. The three non-NMDA receptor agonists, kainate (10(-4)-10(-3) M), alpha-a

Investigating metabotropic glutamate receptors as molecular targets for next-generation pharmacotherapies for alcohol use disorder.

INTRODUCTION: Alcohol use disorder (AUD) is a major public health concern with few available pharmacotherapy options. There remains a critical need to identify novel molecular targets for more effective treatments. Evidence suggests that AUD is associated with dysregulated glutamatergic neurotransmission, including a hyperglutamatergic state that contributes to the development and maintenance of alcohol seeking behaviors. AREAS COVERED: Recent advances in characterizing neuronal, glial, and synaptic changes underlying AUD pathophysiology have identified the metabotropic glutamate receptor 2 (

Quantitation of Glutamine Synthetase 1 Activity in Drosophila melanogaster.

Protocols to assay the activity of glutamine synthetase (GS) are presented as they have been used in our laboratory to correlate the expression levels of the gene encoding Drosophila GS1 gene, the GS1 protein levels, and its activity in extracts of larvae and heads from Drosophila melanogaster. The assays are based on the glutamine synthetase-catalyzed formation of γ-glutamylhydroxylamine in the presence of ATP, L-glutamate, and hydroxylamine, in which hydroxylamine substitutes for ammonia in the reaction. Formation of γ-glutamylhydroxylamine is monitored spectrophotometrically in discontinuou

Serotonin model of schizophrenia: emerging role of glutamate mechanisms.

The serotonin (5-HT) hypothesis of schizophrenia arose from early studies on interactions between the hallucinogenic drug LSD (D-lysergic acid diethylamide) and 5-HT in peripheral systems. More recent studies have shown that the two major classes of psychedelic hallucinogens, the indoleamines (e.g., LSD) and phenethylamines (e.g. , mescaline), produce their central effects through a common action upon 5-HT(2) receptors. This review focuses on two brain regions, the locus coeruleus and the cerebral cortex, where the actions of indoleamine and the phenethylamine hallucinogens have been shown to

[Cerebral hypoxic and ischemic damage in newborn infants: cellular mechanisms and role of excitatory amino acids].

Brain damage from hypoxia-ischemia plays a major role in neonatal mortality and morbidity. Different lesional mechanisms have been proposed. The most recent hypothesis involves the excitatory amino acids, especially glutamate. In this review, arguments in favour of a glutamate involvement in the lesional process are presented, with particular attention to the consequences of glutamate fixation on post synaptic receptors, especially N-methyl-D-aspartate receptors. Alterations of the intracellular concentration of calcium may also play a role in the pathogenesis of hypoxic-ischemic lesions. A be

Glutamate release in experimental ischaemia of the retina: an approach using microdialysis.

A rabbit eye model of neural ischaemia is described that uses an increased pressure in the anterior eye chamber to block the capillary supply to the retina. A microdialysis probe placed very close to the retinal surface was used to monitor release of amino acids during ischaemia. A large (two- to threefold) increase in the release of glutamate and O-phosphoserine (twofold), but not of six other amino acids monitored, occurred during initial ischaemia. During reperfusion after release of intraocular pressure, much larger (five- to 10-fold) increases in the release of these amino acids were obse

Pineal gland free amino acids and indoles during postnatal development of the rat: correlations in individual glands.

Free amino acids and indoles were measured by HPLC in single pineal glands of 5-, 10- and 20-day-old rats sacrificed during mid-light and mid-dark at each age. Melatonin was detectable in neonates (5-day-old), but day vs night differences in indole constituents did not occur until 10 days of age. Free amino acid steady state levels were high in neonates and there was a tendency for reversal of day vs night differences coinciding with the onset of circadian rhythmicity in indole biosynthesis. High correlations (r greater than 0.85) existed for taurine vs. glutamate in individual glands regardle

Age-dependent modifications in the mRNA levels of the rat excitatory amino acid transporters (EAATs) at 48hour reperfusion following global ischemia.

This study reports the mRNA levels of some excitatory amino acid transporters (EAATs) in response to ischemia-reperfusion (I/R) in rat hippocampus and cerebral cortex. The study was performed in 3-month-old and 18-month-old animals to analyze the possible role of age in the I/R response of these transporters. The I/R resulted in a reduced transcription of both the neuronal EAAC1 (excitatory amino acid carrier-1) and the neuronal and glial GLT-1 (glial glutamate transporter 1), while the glial GLAST1a (l-glutamate/l-aspartate transporter 1a) transcription increased following I/R. The changes ob

Intermediary metabolism, Na+, the low calcium-response, and acute disease.

The variables carriage of pCD, CO2 tension, exogenous ATP, L-glutamate, Mg2+, Na+, pH, source of energy, and temperature are known to modulate the low calcium response of Yersinia pestis in vitro. The role of these effectors and the basis of their interactions are defined here with emphasis on known Y. pestis-specific missense mutations in glucose 6-phosphate dehydrogenase and aspartase, which preclude use of the hexose monophosphate pathway and prevent efficient catabolism of L-glutamic acid, respectively. A physiological Ca2+-deficient rescue scenario is provided that permits essentially ful…

Alanine catabolism in Klebsiella aerogenes: molecular characterization of the dadAB operon and its regulation by the nitrogen assimilation control protein.

Klebsiella aerogenes strains with reduced levels of D-amino acid dehydrogenase not only fail to use alanine as a growth substrate but also become sensitive to alanine in minimal media supplemented with glucose and ammonium. The inability of these mutant strains to catabolize the alanine provided in the medium interferes with both pathways of glutamate production. Alanine derepresses the nitrogen regulatory system (Ntr), which in turn represses glutamate dehydrogenase, one pathway of glutamate production. Alanine also inhibits the enzyme glutamine synthetase, the first enzyme in the other pathw

Ultrasmall Superparamagnetic Magnetite Nanoparticles as Glutamate-Responsive Magnetic Resonance Sensors.

Glutamate, the primary excitatory neurotransmitter in the central nervous system, plays a pivotal role in synaptic signaling, learning, and memory. Abnormal glutamate levels are implicated in various neurological disorders, including epilepsy, Alzheimer's disease, and ischemic stroke. Despite the utility of magnetic resonance imaging (MRI) and magnetic resonance spectroscopy (MRS) in diagnosing such conditions, the development of effective glutamate-sensitive contrast agents remains a challenge. In this study, we present ultrasmall, citric acid-coated superparamagnetic iron oxide nanoparticles

Stimulated release of exogenous GABA and glutamate from cerebral cortical synaptosomes and brain slices by bis(acetato)tetrakis(imidazole) copper(II) complex.

In these experiments we have tested the effect of bis(acetato)tetrakis (imidazole) copper(II) on the release and uptake of 14C-GABA and 3H-glutamate from brain slices and brain cortical synaptosomes. Cu(OAc)2(Im)4 in concentrations ranging from 1 to 100 microM has increased the release of GABA and glutamate from brain slices and synaptosomal preparations in a dose-related manner when the effect on GABA release is two-fold greater than glutamate and 10-fold greater than alanine. Pretreatment with a GABA uptake inhibitor such as 1-2 mM nipecotic acid has no effect on 14C-GABA release, whereas hy

The distribution of glutamate, GABA and aspartate in the nucleus tractus solitarius of the cat.

This report describes the distribution of glutamate, GABA and aspartate in the nucleus tractus solitarius (NTS), area postrema, dorsal vagal nucleus and hypoglossal nucleus. Glutamate levels were particularly high in the dorsal, medial and commissural nuclei of the NTS. Section of the IXth and Xth cranial nerves caused a bilateral decrease in glutamate (ipsilateral side = 48%; contralateral side = 36%) and ipsilateral decrease in GABA (27%) in the dorsal and medial NTS. The ipsilateral dorsal vagal nucleus showed a 48% decrease in glutamate and a 30% decrease in GABA after axotomy.

Acute effects of ethanol on glutamate receptors.

Several studies have revealed that acute ethanol inhibits the function of glutamate receptors. Glutamate receptor-mediated synaptic plasticity, such as N-methyl-D-aspartate-dependent long-term potentiation, is also inhibited by ethanol. However, the inhibition seems to be restricted to certain brain areas such as the hippocampus, amygdala and striatum. Ethanol inhibition of glutamate receptors generally requires relatively high concentrations and may therefore explain consequences of severe ethanol intoxication such as impairment of motor performance and memory. Effects of ethanol on glutamate
