Genetic detection and discrimination of closely related oyster pathogens for biosecurity and shellfish stock management.

Disease outbreaks among shellfish cause significant declines in both wild fisheries and aquaculture production globally. In Aotearoa New Zealand, a long history of flat oyster infections by the haplosporidian parasite Bonamia exitiosa has influenced fishery dynamics in the Foveaux Strait. B. ostreae, which causes high host mortality, was detected in both wild and farmed oysters in the Marlborough Sounds in 2015 and at Stewart Island in 2017, prompting the development of rapid diagnostic tests with high sensitivity and specificity that can be used to simultaneously monitor both Bonamia species.

A co-evolved peptide-GPCR system senses host entry to drive fungal infection.

A successful infection requires pathogens to recognize the specific host environment in order to reprogram their physiology accordingly. One major way in which eukaryotic cells sense their surroundings is via G-Protein Coupled Receptors (GPCRs), which share a seven-transmembrane architecture and G-protein-mediated downstream signaling. While mammalian GPCRs are well-characterized and represent important drug targets, their fungal counterparts remain poorly understood. Here we uncover a GPCR-based mechanism that allows the corn pathogen Ustilago maydis to sense whether it has entered plant tiss

The Role of Artificial Intelligence and Machine Learning in Revolutionizing Probiotic Research.

The microbiome, as a vast and dynamic community of microbes, is now recognized as a key regulator of host physiology, profoundly influencing health and susceptibility to disease. Accordingly, probiotics are a mainstay of prevention and treatment. But the individual complexity and dynamic specificity of an individual's microbiome make the previous "one-size-fits-all" research model completely invalid. This review systematically analyzes the applications of artificial intelligence (AI) and machine learning (ML) as transformative computational tools necessary to surmount these challenges. In this

Fermented Dairy Products as Precision Modulators of Gut Microbiota and Host Health: Mechanistic Insights, Clinical Evidence, and Future Directions.

Dairy products-encompassing yogurt, kefir, cheese, and cultured milk beverages-are emerging as versatile, food-based modulators of gut microbiota and host physiology. This review synthesizes mechanistic insights demonstrating how live starter cultures and their fermentation-derived metabolites (short-chain fatty acids, bioactive peptides, and exopolysaccharides) act synergistically to enhance microbial diversity, reinforce epithelial barrier integrity via upregulation of tight-junction proteins, and modulate immune signaling. Clinical evidence supports significant improvements in metabolic par

Shigella flexneri evades LPS ubiquitylation through IpaH1.4-mediated degradation of RNF213.

Pathogens have evolved diverse strategies to counteract host immunity. Ubiquitylation of lipopolysaccharide (LPS) on cytosol-invading bacteria by the E3 ligase RNF213 creates 'eat me' signals for antibacterial autophagy, but whether and how cytosol-adapted bacteria avoid LPS ubiquitylation remains poorly understood. Here, we show that the enterobacterium Shigella flexneri actively antagonizes LPS ubiquitylation through IpaH1.4, a secreted effector protein with ubiquitin E3 ligase activity. IpaH1.4 binds to RNF213, ubiquitylates it and targets it for proteasomal degradation, thus counteracting

A New Species of Unibarra (Monogenoidea, Dactylogyridae) Parasite of Oxydoras niger From Juruá River, State of Acre, Brazil and New Data for U. paranoplatensis.

Monogenoids are ectoparasites that have a simple one-host lifecycle, high species diversity, and a relatively high host specificity. During studies on the helminth fauna of fishes from the Jurua River, in Acre State, Brazil, a new species of the monotypic genus Unibarra Suriano & Incorvaia, 1995 was found parasitizing Oxydoras niger Valenciennes, 1821. Unibarra juruaensis n. sp. is allocated in the genus based on the presence of a single haptoral bar, marginal hooks similar in shape and size, gonads partially overlapping, and a conspicuous filament which connects the base of the male copulator…

The type I macrophage scavenger receptor binds to gram-positive bacteria and recognizes lipoteichoic acid.

Macrophage scavenger receptors exhibit unusually broad binding specificity for polyanionic ligands and have been implicated in atherosclerosis and various host defense functions. Using a radiolabeled, secreted form of the type I bovine macrophage scavenger receptor in an in vitro binding assay, we have found that this receptor binds to intact Gram-positive bacteria, including Streptococcus pyogenes, Streptococcus agalactiae, Staphylococcus aureus, Enterococcus hirae, and Listeria monocytogenes. Competition binding studies using purified lipoteichoic acid, an anionic polymer expressed on the su

Detection of CC17 Enterococcus faecium in dogs and a comparison with human isolates.

Enterococcus faecium strains of clonal complex (CC) 17 were isolated from domestic dogs. The strains were more prevalent in infectious isolates than in colonized isolates, suggesting that strains of the CC17 lineage may have an advantage in causing infections in dogs. The pulsed field gel electrophoresis patterns of some dog and human isolates were over 90% similar. However, antimicrobial resistance patterns and virulence factors were not identical, which might reflect different use of antimicrobials in veterinary medicine or in host specificity.

Phylogenetic and biologic evidence that Babesia divergens is not endemic in the United States.

The causative agent of human babesiosis in a Kentucky case, which was first identified as Babesia divergens, is identical to a parasite of eastern cottontail rabbits on Nantucket Island, Massachusetts based on piroplasm size, morphology, and ribosomal RNA sequence analysis. Studies showing differential infectivity for cattle, host erythrocyte specificity in vitro, parasite size and morphology in vitro, and ribosomal RNA sequences clearly demonstrate that the parasite from the rabbit (conspecific with the human Kentucky agent) is not the same organism as B. divergens.

Cryptosporidium excystation and invasion: getting to the guts of the matter.

Cryptosporidium parvum excystation and host cell invasion have been characterized in some detail ultrastructurally. However, until recently, the biochemical and molecular basis of host-parasite interactions and parasite- and host-specific molecules involved in excystation, motility and host cell invasion have been poorly understood. This article describes our understanding of Cryptosporidium excystation and the events leading to host cell invasion, and draws from information available about these processes in other apicomplexans. Many questions remain but, once the specific mechanisms are iden

Parasitic castration of a vertebrate: Effect of the cymothoid isopod, Anilocra apogonae, on the five-lined cardinalfish, Cheilodipterus quinquelineatus.

Parasitic castration, the specific blocking of host reproductive output by an individual parasite, is a host-parasite interaction common to many invertebrates, particularly crustaceans, echinoderms and molluscs. It can reduce host density, alter host population dynamics and the evolution of host life history traits. Here we show that parasitisation by a single female cymothoid isopod, Anilocra apogonae, castrates its vertebrate host, the five-lined cardinalfish, Cheilodipterus quinquelineatus. Parasitised male fish fail to mouthbrood their young. The gonads of parasitised fish are smaller and

Bridging two hosts: how intracellular environments shape flaviviral infection.

Mosquito-borne flaviviruses replicate in physiologically and biochemically distinct host environments in humans and mosquitoes, providing a unique window into conserved and host-specific mechanisms shaping viral infection efficiencies and outcomes. This review focuses specifically on intracellular factors, including proteins, metabolites, innate immune effectors, and stress sensors in human and mosquito cells that collectively regulate the flaviviral life cycle and host cell survival, with specific emphasis on dengue virus. We discuss both conserved dependencies and species-specific difference

The origins and potential cross-species transmission of paramyxoviruses and other RNA viruses between native snakes and invasive Burmese pythons in the Florida Everglades.

UNLABELLED: During November-December 2020, a disease outbreak involving eastern mudsnakes (Farancia abacura abacura) was documented in the Greater Everglades Ecosystem, Florida, USA. Moribund snakes were observed writhing on riverbanks with concomitant frothy oral discharge. Pathological examination revealed necrotizing enteritis and pulmonary edema, and a reptilian ferlavirus phylogenetically related to anaconda paramyxoviruses was isolated. While ferlaviruses are well-described pathogens of snakes in zoological and private collections worldwide, this represented the first outbreak of ferlavi

A genomic catalog of the mouse gut virome reveals features associated with ageing.

The gut virome is emerging as a key contributor to host physiology, yet it remains underexplored relative to the gut bacteriome. Despite comprehensive viral catalogs for the human gut, comparable resources for mice, the principal model organism in biomedical research, remain lacking, limiting our understanding of virome-host interactions and their implications for host health and disease. Here, we present Mouse Reference Gut Virome (MRGV), a comprehensive catalog comprising 109,778 viral genomes with ≥50% completeness and representing 28,824 species, expanding known mouse gut viral diversity b

AI-enabled viral genomics: from virus discovery to host prediction and emerging variant forecasting.

The rapid expansion of metagenomic sequencing has generated vast repositories of viral sequence data that far outpace our capacity to interpret them using conventional approaches. Highly divergent sequences, sparse functional annotation, and taxonomically uneven sampling present fundamental challenges for reference-dependent methods, which lose sensitivity precisely for novel and understudied viruses with high public health relevance. Artificial intelligence (AI) provides a new avenue to address these challenges by enabling predictive inference from viral genomes and proteins while reducing de
