Enhancing the efficacy of adoptively transferred T cells is essential for successful immunotherapy. The cytotoxic activity of these T cells is closely related to their mitochondrial function, which plays a critical role in T cell differentiation. This study explored the potential of nicotinamide riboside (NR), known to enhance mitochondrial function, to improve the efficacy of adoptively transferred CD8+ T cells against hepatocellular carcinoma (HCC). In subcutaneous and metastatic HCC tumor models, NR treatment significantly improved the effectiveness of adoptive T cell therapy (ACT). NR prom

The recent COVID-19 pandemic left behind the lingering question as whether new variants of concern might cause further waves of infection. Thus, it is important to investigate the long-term protection gained via vaccination or exposure to the SARS-CoV-2 virus. Here we compare the evolution of memory T-cell responses following primary infection with subsequent antigen exposures. Single-cell TCR analysis of three dominant SARS-CoV-2 spike-specific CD4+ T-cell responses identifies the dominant public TCRα clonotypes pairing with diverse TCRβ clonotypes that associated with mild disease at primary

Endometrial immune disorders create an inhospitable endometrial environment for embryonic nidation in endometriosis. CD8 + tissue resident memory T cells (CD8 +TRM) are abundant tissue resident immune cells in endometrium, but the effect of CD8 +TRM on endometrial receptivity of endometriosis remains unexplored. We collected endometrium tissue to explore the number and function of CD8 +TRM in eutopic endometrium of endometriosis and controls through flow cytometry. Then we co-cultured CD8 +TRM with endometrial stromal cells (ESCs) to explore the influence of CD8 +TRM on the expression of endom

BACKGROUND: HIV-1-infected individuals progressively loss CD4(+) T cells leading to immunosuppression and raising the risk of opportunistic infections. CD8(+) T-cells play an important role in the immune response against virus infections through their TCR.
OBJECTIVE: To evaluate the CD8-TCR repertoire and immunologic markers in HIV-1-infected patients.
STUDY DESIGN: Ten chronic HIV-1-infected individuals on prolonged effective antiretroviral treatment (ART) were analyzed at baseline (before treatment interruption), after at least one year of treatment interruption (TI) and after at least one

INTRODUCTION: SARS-CoV-2 is continuing to prevail as an endemic virus, and therefore, we need a next-generation vaccine that prevents SARS-CoV-2 infections, broadly protects against multiple CoVs, and induces long-term local and systemic immunity. To address that need, we have designed a mixed lipopeptide-based pan-coronavirus (LPMix) vaccine based on T and B cell epitopes derived from highly conserved and functional regions of the SARS-CoV-2 spike (S), nucleocapsid (N), and membrane (M) proteins.
METHODS: Male C57BL/6 mice (n=5 per group) were immunized intranasally twice, 14 days apart, wit

BACKGROUND: Tertiary lymphoid structure (TLS) is a neogenized, ectopic lymphoid aggregate found in infected, autoimmune and tumour tissues with an activated adaptive immune response. However, a comprehensive understanding of the pathological role, function, and formation of TLS in allograft rejection remains incomplete.
METHODS: We enrolled two large retrospective cohorts of liver biopsy (LB) after pediatric living donor liver transplantation (LDLT) and developed a deep learning pathomics (DLP) model. Gene expression profiles and corresponding clinical information of 590 cases were enrolled f

Conventional influenza virus vaccines induce antibody responses of limited breadth. Whether mRNA-based influenza virus vaccines can induce a superior germinal center (GC) response in humans remains unclear. Here we assessed B cell responses in an observational study of cohorts of healthy young adults receiving a licensed, split-virion or investigative mRNA-based quadrivalent seasonal influenza virus vaccine over two consecutive seasons. mRNA-based vaccines consistently elicited higher antibody titers and frequencies of memory B cells. In the draining lymph nodes, mRNA vaccination stimulated su

BACKGROUND: Tissue-resident memory T (TRM ) cells are detrimental in allergic contact dermatitis (ACD), in which they contribute to the chronicity and severity of the disease.
METHODS: We assessed the impact of a standard topical corticosteroid (TCS) treatment, triamcinolone acetonide (TA), on the formation, maintenance and reactivation of epidermal TRM cells in a preclinical model of ACD to 2,4-dinitrofluorobenzene. TA 0.01% was applied at different time points of ACD response and we monitored skin inflammation and tracked CD8+ CD69+ CD103+ TRM by flow cytometry and RNA sequencing.

Allergen-specific CD4+ T cells play a pivotal role in the pathophysiology of food allergy and in the induction of tolerance during desensitization. However, their study remains challenging due to the considerable heterogeneity in the frequency and phenotype of food allergy-specific Th2 cells in allergic individuals, together with an extremely low abundance of T cells specific to a given allergen. Furthermore, a major limitation of human studies is their reliance on peripheral blood sampling, whereas higher frequencies of allergen-specific cells may reside in intestinal tissues directly implica

CD57+ CD8+ T cells, also referred as effector memory cells, are implicated in various conditions including tumor immunity, virus immunity, and most recently with autoimmunity. However, their roles in the progression and remission of T1D are still unclear. Here, we noted an increase in peripheral CD57+ CD8+ T cells in a T1D patient harboring an activator of transcription 3 (STAT3) mutation. Our in-depth study on the role of CD57+ CD8+ T cells within a T1D patient cohort revealed that these cells undergo significant compositional shifts during the disease's progression. Longitudinal cohort data

Bacille Calmette-Guérin (BCG), an attenuated vaccine derived from Mycobacterium bovis, is the current vaccine of choice against tuberculosis (TB). Despite its protection against active TB in children, BCG has failed to protect adults against TB infection and active disease development, especially in developing countries where the disease is endemic. Currently, there is a significant effort toward the development of a new TB vaccine. This review article aims to address publications on recombinant BCG (rBCG) published in the last 5 years, to highlight the strategies used to develop rBCG, with a

Immunological correction of cognitive processes impaired due to the action of neurotoxic amyloidogenic forms of proinflammatory protein S100A9, a promoter of the inflammatory-amyloid cascade occurring in Alzheimer's disease, is poorly understood. Chronic intranasal administration of S100A9 fibrils leads to suppression of spatial memory formation in the Morris water maze in 12-month-old C57BL/6J mice and to an increase in activity of the ASCL1 gene involved in neurogenesis at the stage of cell differentiation, in the hippocampus and prefrontal cortex. In the case of combined administration of S

CD4+ T cells orchestrate adaptive immunity against respiratory viruses through functionally distinct subsets, yet the qualitative differences between vaccine- and infection-induced responses remain incompletely understood. This review summarizes current evidence on how CD4+ T cell subsets shape the magnitude, breadth, and durability of immunity to SARS-CoV-2 and influenza virus. mRNA vaccines elicit a potent Th1/Tfh1-biased response that generates high-affinity neutralizing antibodies and cytotoxic CD4+ T cells (CD4-CTLs) with stem-cell memory properties. However, this strong type I bias may c

While the role of breast milk antibodies to protect infants from human cytomegalovirus (HCMV) has been investigated, the role of T-cells has received little attention. We compared the frequency of memory T-cell populations in breast milk between mothers of infants who acquired breast milk HCMV (transmitters) and those with uninfected infants (nontransmitters). Nontransmitter mothers had an increased frequency of CD8+ effector memory T-cells (Tem) in their breast milk. In addition, we describe tissue-resident memory T cells (Trm) in breast milk and demonstrate most were Tem. We present data tha

BACKGROUND: Bystander activation has primarily focused on conventional antigen-specific T cells (TMEM) and other innate immune cell types. However, the role of virtual memory T cells (TVM) has been largely overlooked, despite their numerical superiority and highly cytotoxic phenotype. Bystander activation is particularly relevant in infections caused by intracellular pathogens. In this study, we aimed to compare the bystander activation potential of TVM cells versus TMEM cells during the early days following T. cruzi infection.
METHODOLOGY/PRINCIPAL FINDINGS: Our results demonstrate that TVM

BACKGROUND: LL-37 is the only member of the cathelicidin family of antimicrobial peptides in humans and is an autoantigen in several autoimmune diseases and in acute coronary syndrome (ACS). In this report, we profiled the specific T cell response to the autoimmune self-antigen LL-37 and investigated the factors modulating the response in peripheral blood mononuclear cells (PBMCs) of healthy subjects and ACS patients.
METHODS AND RESULTS: The activation induced marker (AIM) assay demonstrated differential T cell profiles characterized by the persistence of CD134 and CD137, markers that impair

Treatment-free remission (TFR) is a key therapeutic goal for chronic myeloid leukemia (CML) patients in deep molecular response (DMR). While predicting patient outcome remains challenging, different NK cell populations seem crucial. We conducted an immunological sub-study from the Argentina Stop Trial (AST), including 46 patients in 2019 (AST I) and 35 new patients between 2022 and 2023 (AST II). To characterize NK cell subsets in patients attempting TFR, peripheral blood mononuclear cell samples were collected before stopping treatment and phenotype and functional characteristics were assesse

BACKGROUND: Vaccine-induced memory B-cell (MBC) subsets have distinct roles in the establishment of protective immunity; MBCs expressing nonswitched immunoglobulin M (IgM+ MBCs) replenish the MBC pool, whereas MBCs expressing isotype-switched immunoglobulin (sIg+ MBCs) differentiate into plasma cells upon antigen reencounter. We investigated immunogenicity and MBCs induced by combined 13-valent pneumococcal conjugate vaccine (PCV13) and 23-valent pneumococcal polysaccharide vaccine (PPV23) in human immunodeficiency virus (HIV)-infected adults.
METHODS: Forty HIV-seropositive adults receiving

The clinical value of assessing immunoglobulin (Ig)G and IgA subclasses in addition to the isotypes of soluble Igs in serum has been well established. >20years ago, the International Union of Immunological Societies and the World Health Organization performed collaborative studies in order to validate antibody (Ab) clones for the detection of IgG and IgA subclasses for a broad range of laboratory assays, except for flow cytometry. Here we analyzed the performance of commercially available Ab clones to detect IgG and IgA subclasses in memory B-cells and plasma cells (PCs) by flow cytometry. In
