Astaxanthin and Male Reproductive Health: Mechanisms, Experimental Evidence, and Clinical Perspectives.

Oxidative stress is a primary driver of male infertility, underscoring the critical importance of effective antioxidant defense systems in maintaining reproductive function. Astaxanthin, a xanthophyll carotenoid with potent free radical-scavenging capacity and membrane-stabilizing properties, has garnered interest as a therapeutic agent for male reproductive health. Experimental evidence indicates that astaxanthin modulates cellular redox homeostasis, preserves mitochondrial integrity, and attenuates lipid peroxidation in spermatozoa and testicular cells. Through these mechanisms, it potential

Integration of Bone Remodeling and Damage Accumulation: A Preliminary Computational Framework for Physiological and Pathological Responses.

This study presents a computational model that integrates bone remodeling dynamics with damage accumulation, focusing on both physiological and pathological conditions. Building upon Komarova’s classical model of osteoclast and osteoblast interactions, this work introduces fatigue-induced damage using a stress-life (S-N) approach. By simulating bone responses under sinusoidal and random mechanical loads, the model captures the cyclical nature of bone turnover. The results show that under normal physiological conditions, bone is able to repair microdamage and maintain structural integrity. Howe

A mitochondrial inflammation-oriented nanozyme coordinates dual inflammatory axes to restore alveolar bone homeostasis in periodontitis.

Mitochondrial inflammatory stress is an important pathological contributor to alveolar bone loss in periodontitis, yet strategies capable of effectively modulating its initiation and associated signaling network remain limited. Here, a mitochondrial inflammation-oriented nanozyme, CeO2-DMC@ZIF-8, is rationally designed via in situ integration of the NRF2 agonist DMC into a CeO2@ZIF-8 framework. This platform combines catalytic ROS scavenging with pH-responsive drug release, enabling integrated modulation of mitochondrial stress-associated inflammatory pathways. Under inflammatory stimulation,

Infection of endothelial cells by Streptococcus agalactiae reveals potential role of PI-2b pilus on endothelial barrier dysfunction.

BACKGROUND: Streptococcus agalactiae is responsible for sepsis and meningitis, and the major cause of neonatal morbidity and mortality. However, how S. agalactiae disrupts endothelial barriers is poorly understood. OBJECTIVES: Analyse the influence of endothelial cell (HUVECs) growth under static and shear stress conditions during infection with S. agalactiae, and the role of pilus PI-2b during endothelial barrier disruption and increased endothelial permeability. METHODS: HUVECs under static and shear conditions were infected by S. agalactiae (GBS90356 and GBS90356Δpilus2b) strains in the p

Molecular Neuroprotective Responses to Achyrocline satureioides Extract in Caenorhabditis elegans: Modulation of Conserved FOXO/NRF2 Pathways and Resistance to Neurotoxic Insults.

Achyrocline satureioides is widely consumed as an herbal infusion in South America; however, the toxicological profile of its aqueous extract (ASAE), particularly under conditions of neurotoxic stress, remains incompletely characterized. In the present study, we investigated the safety profile of ASAE and its capacity to modulate chemically induced neurotoxicity by integrating in silico toxicological predictions with in vivo assays in Caenorhabditis elegans. Wild-type and mutant strains deficient in key stress-response regulators (daf-2, daf-16, and skn-1) were exposed to ASAE (10-50 mg/mL). T

Impairment in global protein synthesis uncouples UPR gene induction from HAC1 mRNA splicing in Saccharomyces cerevisiae.

Upon dysfunction of the endoplasmic reticulum (ER), also known as ER stress, eukaryotic cells alter their transcriptomes. This cytoprotective response is called the unfolded protein response (UPR), which is mediated by Ire1 and HAC1 in the yeast Saccharomyces cerevisiae. ER stress induces self-association and activation of the ER-resident transmembrane endoribonuclease Ire1, which catalyzes the splicing of HAC1 mRNA. It is widely accepted that HAC1 mRNA is translated into the nuclear transcription factor Hac1, only after being spliced. To investigate the cellular response to ethanol-induced ER

Colon-targeted EGCG delivery remodels the gut microenvironment to enhance therapeutic efficacy in ulcerative colitis.

Ulcerative colitis (UC) is a chronic intestinal inflammatory disorder characterized by epithelial barrier disruption, oxidative stress, and gut microbiota dysbiosis. Although epigallocatechin-3-gallate (EGCG) exhibits potent anti-inflammatory and antioxidant activities, its clinical application is limited by poor stability and low colonic bioavailability. Herein, EGCG-loaded Eudragit® S100-coated chitosan microparticles (EG@euCS MPs) were developed as a colon-targeted oral delivery platforms integrating mucoadhesive retention with pH-responsive release. EG@euCS MPs effectively protected EGCG d

Heat Shock Proteins in Medically Relevant Fungal Pathogens: From Molecular Chaperones to Virulence Factors and Therapeutic Targets.

Heat shock proteins (HSPs) are highly conserved molecular chaperones that play a key role in maintaining protein homeostasis and cellular survival under stress conditions. Clinically relevant human pathogenic fungi include opportunistic fungi, dimorphic fungi, dermatophytes, Mucorales, and other pathogenic groups. HSPs, including Hsp90, Hsp70, Hsp60, Hsp40, and Hsp110, are essential for the correct nascent protein folding, aggregation prevention, and degradation of misfolded polypeptides. Fungal pathogens frequently encounter environmental and host-imposed stresses, including oxidative stress,

p62/SQSTM1: From an autophagy receptor to a condensate organizer of selective autophagy and stress signaling.

Upon exposure to stress, cells activate a variety of stress-response and quality-control mechanisms to maintain homeostasis. Dysregulation of these processes is implicated in numerous diseases, including cancer, liver disorders, and neurodegenerative diseases. p62/Sequestosome 1 (SQSTM1) is a multifunctional protein that plays a central role in protein homeostasis and stress responses by regulating autophagy and signal transduction pathways. Through its multiple protein-interacting domains, p62 functions both as a scaffold for selective autophagic degradation and as a signaling hub. Since our

Antioxidant chemistry and its role in mitigating oxidative stress in animals.

Oxidative stress is a major physiological challenge in poultry and livestock production, arising from an imbalance between reactive oxygen and nitrogen species generation and the antioxidant defense systems of animals. Stressors such as heat, high stocking density, transport, nutritional imbalances, and disease pressure exacerbate oxidative damage under intensive production conditions, leading to impaired immunity, reduced productivity, compromised meat quality, and increased disease susceptibility. Excessive reactive species negatively affect the immune, muscular, reproductive, digestive, and

A Review of Biological Pathways of Chronic Stress as a Risk Hub for Multiple Psychosomatic Diseases From the Perspective of Clinical Nursing.

Chronic stress has emerged as a critical global public health threat, acting as a risk hub that mediates the onset and progression of diverse psychosomatic diseases (e.g., cardiovascular diseases, mental disorders, metabolic syndrome, and even cancer) through interconnected biological pathways. This review systematically elaborates on the core biological mechanisms underlying chronic stress-induced psychosomatic dysfunction, including hypothalamic-pituitary-adrenal (HPA) axis dysregulation, autonomic nervous system (ANS) imbalance, abnormal inflammatory responses, and excessive oxidative stres

High-temperature stress in crops: male sterility, yield loss and potential remedy approaches.

Global food security is one of the utmost essential challenges in the 21st century in providing enough food for the growing population while coping with the already stressed environment. High temperature (HT) is one of the main factors affecting plant growth, development and reproduction and causes male sterility in plants. In male reproductive tissues, metabolic changes induced by HT involve carbohydrates, lipids, hormones, epigenetics and reactive oxygen species, leading to male sterility and ultimately reducing yield. Understanding the mechanism and genes involved in these pathways during t

Protective effect of mitochondria-targeted antioxidants in an acute bacterial infection.

Acute pyelonephritis is a potentially life-threatening infection of the upper urinary tract. Inflammatory response and the accompanying oxidative stress can contribute to kidney tissue damage, resulting in infection-induced intoxication that can become fatal in the absence of antibiotic therapy. Here, we show that pyelonephritis was associated with oxidative stress and renal cell death. Oxidative stress observed in pyelonephritic kidney was accompanied by a reduced level of mitochondrial B-cell lymphoma 2 (Bcl-2). Importantly, renal cell death and animal mortality were both alleviated by mitoc

Transcriptional and epigenomic changes in response to polyethylene glycol-triggered osmotic stress in Brassica napus L.

Drought hinders growth, development, and productivity of higher plants. While the physiological and molecular background of plant responses to drought has been extensively studied, the role of post-translational modifications of histones or DNA methylation in response to dehydration remains largely elusive. In this study, we deciphered genome-wide changes in transcriptome and histone modifications in response to dehydration in rapeseed (Brassica napus L.). High-throughput transcript profiling (RNA-seq) and ChIP followed by sequencing (ChIP-seq) of polyethylene glycol (PEG)-treated rapeseed pla

Jasmonate signaling modulates root growth by suppressing iron accumulation during ammonium stress.

Plants adapt to changing environmental conditions by adjusting their growth physiology. Nitrate (NO3-) and ammonium (NH4+) are the major inorganic nitrogen forms for plant uptake. However, high NH4+ inhibits plant growth, and roots undergo striking changes, such as inhibition of cell expansion and division, leading to reduced root elongation. In this work, we show that high NH4+ modulates nitrogen metabolism and root developmental physiology by inhibiting iron (Fe)-dependent Jasmonate (JA) signaling and response in Arabidopsis (Arabidopsis thaliana). Transcriptomic data suggested that NH4+ ava

Iron deficiency and toxicity trigger divergent metabolic responses and adaptive plasticity in Ulmus pumila: Insights from integrated transcriptomic and metabolomic analyses.

Iron homeostasis is critical for plant growth; however, the mechanisms underlying responses to iron deficiency and toxicity remain poorly understood. We investigated the adaptive strategies of Ulmus pumila, focusing on leaf physiological, transcriptomic, and metabolomic responses to iron stresses. Both iron deficiency and toxicity impaired chlorophyll biosynthesis, PS II efficiency, and chloroplast ultrastructure, resulting in reduced photosynthetic capacity and etiolation/wilting phenotypes. Iron deficiency reduced antioxidant enzyme activity and ROS levels, while iron toxicity activated the

Mechanobiological Response of Osteocyte TRPV4 Base on the Piezoelectricity of Bone Matrix.

PURPOSE: Osteocytes are force-sensitive cells possessing a complex lacunar-canalicular system (LCS), embedded within a piezoelectric bone matrix. TRPV4, as a key mechanosensor, plays a crucial role in osteocyte mechanotransduction and related bone disorders. However, the mechanisms underlying its force sensing, mechanical signaling pathways, and biomechanical response remain poorly understood. METHODS: To address this issue, this study established a finite element model incorporating multiple mechanosensors-including bone matrix, LCS, osteocytes, and TRPV4-based on the piezoelectric effect. Ecotoxicological effects of cadmium on Echinometra mathaei: Oxidative stress, genotoxicity, and biochemical responses.

Cadmium (Cd), a non-essential heavy metal, poses a persistent threat to marine ecosystems. The edible sea urchin Echinometra mathaei, widely recognized as a sentinel species, was used to evaluate the effects of environmentally relevant Cd exposure. These organisms were exposed to 0, 0.01, and 0.1 mg L-1 Cd for 28 days, and gonadal tissues were analyzed for Ca content, oxidative stress biomarkers (H₂O₂, MDA, SOD, CAT), protein and carbohydrate reserves, and genotoxic endpoints (tail intensity, tail moment, tail factor). Cd accumulated progressively in gonads, coinciding with calcium depletion,

Integrating computational evaluation and mechanical testing to optimize laser-sintered polyether ketone scaffolds for mandibular reconstruction in large animal models.

Critical-sized mandibular defects require reconstruction strategies that restore mechanical function while avoiding the limitations of bone grafting and metallic fixation. This study aimed to optimize the mechanical performance of laser-sintered polyether ketone (LS-PEK) scaffolds using an integrated computational and experimental approach. Three scaffold geometries with different bone-implant interface designs (straight-cut/non-curved, straight-cut/curved, and wedge-cut/curved) were evaluated using image-based finite element analysis and extended finite element method (XFEM) fracture simulati
