Maple syrup urine disease (MSUD) is a metabolic disorder that is caused by deficiency of branched-chain α-keto acid dehydrogenase complex. Although accumulation of toxic metabolites is associated with neurotoxicity, mechanisms underlying brain damage remain unclear. Aim of this study is to evaluate thiol/disulphide homeostasis as a novel indicator of oxidative stress in MSUD patients under treatment. The effect of prostacyclin on canine hepatic bile flow was evaluated. Utilizing chronic, unanesthetized bile fistula dogs, prostacyclin was found to significantly increase bile flow rates and bile bicarbonate and cyclic AMP concentration and output when compared to control values. PURPOSE: The goals of this investigation were to examine the synthesis of prostaglandins after UVB exposure of lens epithelial cells and to investigate their role in cell proliferation.
METHODS: Cultured rabbit lens epithelial cells (cell line N/N1003A) were exposed to low levels of UV radiation. Prostaglandins were assayed by radioimmunoassay; products of arachidonic acid metabolism were analyzed by thin-layer chromatography and mass spectroscopy.
