Corneal irregularity of the anterior and posterior surface in patients with limbal stem cell deficiency evaluated with anterior-segment optical coherence tomography.

PURPOSE: To evaluate corneal anterior and posterior irregularities due to limbal stem cell deficiency (LSCD) based on staging using Fourier harmonic analysis with anterior segment optical coherence tomography (AS-OCT). STUDY DESIGN: Retrospective observational study. METHODS: Patients with LSCD and those without anterior segment disease (controls), examined using AS-OCT, were retrospectively included. Based on Fourier harmonic analysis of the central 3 mm, spherical components, regular astigmatism, asymmetry components, and higher-order irregularities of the anterior, posterior, and total co

Fibrin glue inhibits migration of ocular surface epithelial cells.

PurposeFibrin glue has been used successfully in numerous ophthalmic surgical procedures. Recently, fibrin glue has been used in limbal stem cell transplantation to reduce both operative time and to negate the need for sutures. The aim of this study was to determine the effects of fibrin glue on epithelial cell migration in vitro.MethodsCorneoscleral rims were split to retain the epithelial layer, Bowman's layer, and anterior stroma. Rims were cut into eight equal-sized pieces and were placed directly on culture plates or affixed with fibrin glue. Rims were maintained in culture for 25 days an

Intravenous immunoglobulin for antibody-mediated keratolimbal allograft rejection.

A 33-year-old woman with congenital aniridia presented with decreased vision in her right eye. Slit lamp examination revealed diffuse conjunctivalisation of the ocular surface with mild subepithelial fibrosis consistent with aniridic keratopathy secondary to limbal stem cell deficiency. She underwent limbal stem cell transplantation with cadaver donor tissue (keratolimbal allograft (KLAL) surgery) and received systemic immunosuppression. Despite optimal combination immunosuppressive therapy managed by a renal transplant specialist, 2 weeks after the KLAL, the patient developed intractable eye

Differential regulation of GLUT1 activity in human corneal limbal epithelial cells and fibroblasts.

The corneal epithelial tissue is a layer of rapidly growing cells that are highly glycolytic and express GLUT1 as the major glucose transporter. It has been shown that GLUT1 in L929 fibroblast cells and other cell lines can be acutely activated by a variety agents. However, the acute regulation of glucose uptake in corneal cells has not been systematically investigated. Therefore, we examined glucose uptake in an immortalized human corneal-limbal epithelial (HCLE) cell line and compared it to glucose uptake in L929 fibroblast cells, a cell line where glucose uptake has been well characterized.

Manual limbal markings versus iris-registration software for correction of myopic astigmatism by laser in situ keratomileusis.

PURPOSE: To compare the efficacy and safety of manual limbal markings and wavefront-guided treatment with iris-registration software in laser in situ keratomileusis (LASIK) for myopic astigmatism. SETTING: National Taiwan University Hospital, Taipei, Taiwan. METHODS: Eyes with myopic astigmatism had LASIK with a Technolas 217z laser. Eyes in the limbal-marking group had conventional LASIK (PlanoScan or Zyoptix tissue-saving algorithm) with manual cyclotorsional-error adjustments according to 2 limbal marks. Effect of immunosuppression on survival of allograft limbal stem cells.

PURPOSE: To examine whether the application of immunosuppression enhances the survival of limbal allograft stem cells. METHODS: Wistar (allograft) or Fischer 344 (isograft) rat limbal tissue was transplanted into superior lamellar excision sites in Fischer 344 rats. Allograft-recipient rats received an immunosuppressive agent or vehicle for 8 weeks. Graft-recipient rats were examined by slit-lamp microscopy for clinical signs of rejection, and some recipients were killed for immunohistochemical analysis during rejection. Pterygium surgery with mitomycin C: how much sclera should be left bare?

PURPOSE: To evaluate the effect of covering the sclera with conjunctiva during surgical treatment of pterygium on the rate of postoperative complications, cosmetic outcome, and risk of recurrence. METHODS: A comparative, prospective clinical design was used. The study population consisted of 371 consecutive patients with unilateral primary advanced pterygium. All underwent bare sclera technique for pterygium excision. At completion of surgery, topical mitomycin C 0.02% was applied for 5 minutes. In the first 189 patients (group A), the conjunctiva was sutured to the sclera 2 mm from the limbu

Expression of a mucin-like glycoprotein produced by ocular surface epithelium in normal and keratinized cells.

PURPOSE: We previously characterized a monoclonal antibody (H185) to a mucin-like glycoprotein produced by human ocular surface epithelium. In the current study, we used H185 to investigate the pattern of the mucin-like glycoprotein in normal and keratinized apical cells of the ocular surface epithelium. METHODS: We compared the cell characteristics and the pattern of H185 binding in cytologically and immunohistochemically stained samples of apical cells of conjunctival surface epithelium from 20 normal subjects and six patients before and after treatment for superior limbic keratoconjunctivi

DNA Oxidation and Expression of Repair Enzymes in Organ- Cultured Human Limbal Epithelium.

DNA oxidation damage and its repair are essential for maintaining genomic integrity in the human limbal epithelium, which harbors corneal epithelial stem cells. This study investigated the distribution of the DNA base oxidation 8-oxoguanine (8-oxoG) and the base excision repair (BER) enzymes 8-oxoguanine DNA glycosylase (OGG1) and apurinic/apyrimidinic endonuclease 1 (APE1) in non-cultured and eye-bank organ-cultured human limbal epithelia. Immunohistochemistry was used to assess the localization and staining intensity of 8-oxoG, OGG1, and APE1, evaluated semi-quantitatively and by image analy

miR-138-5p overexpression inhibits limbal epithelial cell proliferation and induces cell cycle arrest, in vitro.

MicroRNAs (miRNAs) play critical roles in ocular surface diseases. In our recent study, through microarray analysis, miR-138-5p was identified as one of the most prominent miRNAs expressed in aniridia patients derived primary limbal epithelial cells (pLECs). Although miR-138-5p expression was significantly elevated in aniridia samples, its specific role in the pathogenesis of aniridia-associated keratopathy (AAK) remained unclear. Therefore, the present study focused on exploring the potential functional impact of upregulated miR-138-5p on limbal epithelial stem cell maintenance and function.

Mechanistic insights into conjunctivalization in limbal stem cell deficiency using a lineage tracing approach.

PURPOSE: Disease that develops on the ocular surface in animals and humans that have their limbal stem cells erased or their niche severely damaged by physical or chemical trauma, is difficult to visually evaluate in a progressive and long-term manner. When limbal stem cell deficiency (LSCD) develops under such circumstances, intravital clinical microscopy or other techniques cannot be used to accurately detail what transpires on the cornea. Here, we investigated the evolution of LSCD and characterize cellular transformations that arise when the limbal niche is lost. METHODS: We employed the
