Opposing role of JNK-p38 kinase and ERK1/2 in hydrogen peroxide-induced oxidative damage of human trophoblast-like JEG-3 cells.

Trophoblasts play a crucial role in embryo implantation and maintenance of normal pregnancy. Recently, oxidative stress has been considered as one important factor in the pathogenesis of spontaneous abortion and preeclampsia. Many studies have reported that the plasma levels of hydrogen peroxide (H2O2) are significantly increased in women with preeclampsia, but the mechanisms involved in H2O2-induced cell cytotoxicity in trophoblasts are still not completely explained. Our present study was undertaken to provide a united understanding of the role of oxidative stress generated by H2O2 on human

Constitutively active mitogen-activated protein kinase versions reveal functions of Arabidopsis MPK4 in pathogen defense signaling.

Plant mitogen-activated protein kinases (MAPKs) are involved in important processes, including stress signaling and development. In a functional yeast screen, we identified mutations that render Arabidopsis thaliana MAPKs constitutively active (CA). Importantly, CA-MAPKs maintain their specificity toward known activators and substrates. As a proof-of-concept, Arabidopsis MAPK4 (MPK4) function in plant immunity was investigated. In agreement with the phenotype of mpk4 mutants, CA-MPK4 plants were compromised in pathogen-induced salicylic acid accumulation and disease resistance. MPK4 activity w

Nitric oxide induces extensive apoptosis in endometrial epithelial cells in the presence of progesterone: involvement of mitogen-activated protein kinase pathways.

During trophoblast invasion, luminal and glandular endometrial epithelial cells (EEC) have been found to undergo apoptosis through undetermined mechanisms. We postulate that nitric oxide (NO) and progesterone may mediate apoptosis in EEC because they are produced by trophoblasts at concentrations that can cause apoptosis in non-uterine cells. Using a cultured EEC line, RL95-2, we found that sodium nitroprusside (SNP) or S-nitroso-N-acetylpenicillamine (SNAP), two commonly used NO-releasing agents, caused the death of EEC in a dose-dependent manner and progesterone markedly enhanced NO-induced

Interaction mechanism of low molecular weight fucoidan in targeting SR-A to modulate the MAPK signal pathway for the inhibition of oral leukoplakia.

Oral leukoplakia (OLK) is the most prevalent oral precancerous lesion, with its occurrence and progression linked to local irritants and systemic factors. Among these, chronic inflammatory microenvironment significantly promotes OLK. Conventional drug treatments have notable limitations and a high recurrence risk, underscoring the urgent need to identify reliable molecular markers and to develop effective, safe targeted therapies for OLK. This study identified abnormal overexpression of scavenger receptor A (SR-A) in the oral mucosal epithelium of patients with OLK, and low molecular weight fu

Long-Term Co-Exposure to Nanopolystyrene and Heterocyclic Aromatic Amines Induces Neurotoxicity in Zebrafish via the MAPK Pathway.

Nanoplastics (NPs) are common hydrophobic contaminants in food packaging, readily interacting with other pollutants to produce severe unknown toxicity. Heterocyclic amines (HAAs) are carcinogenic hydrophobic byproducts commonly generated during the thermal processing of meat products. With the widespread adoption of takeout food, coexposure risks to NPs and HAAs are inevitable, yet studies on their combined contamination remain scarce. This study comprehensively evaluated the chronic toxic effects of coexposure to polystyrene nanoplastics (PS-NPs) and 9H-pyridine[3,4-b]indole (Norharman) on ze

SPP1+ macrophages contribute to silica-induced pulmonary inflammation and fibrosis via the ERK/JNK signaling pathway.

Secreted phosphoprotein 1-positive (SPP1+) macrophages play a critical role in the progression of silicosis, but their regulatory mechanisms remain unclear. In this study, we integrated single-cell RNA sequencing and spatial transcriptomics to characterize crucial macrophage subpopulations and their associated regulatory pathways during silicosis. Our results demonstrate that silica exposure significantly promotes the accumulation of SPP1+ macrophages within silicotic nodules. These macrophages contribute to inflammation by releasing pro-inflammatory cytokines such as TNF-α, MCP-1, and IL-6. C

Fermentation with aspergillus costiformis enhances the anti-migratory activity of Aquilaria sinensis leaves with possible involvement of MAPK-related pathways.

Aquilaria sinensis leaves (ALs) are an underutilized plant resource rich in polyphenols and flavonoids. Microbial fermentation has been proposed as an effective strategy to enhance the functional value of plant materials. However, its impact on the bioactivity and mechanisms of ALs remains unclear. This study aimed to evaluate the effects of fermentation with Aspergillus costiformis AHHC on the chemical composition and biological activities of ALs, and to elucidate the molecular mechanisms underlying the fermentation-induced functional changes. Non-targeted metabolomic analysis was performed t

Koumine inhibits osteoclastogenesis and prevents ovariectomy-induced bone loss via suppression of the MAPK signaling pathways.

Excessive osteoclast formation drives osteolytic bone diseases such as osteoporosis. Koumine (KM), an alkaloid derived from Gelsemium elegans, exhibits various bioactivities; however, its role in bone homeostasis remains unknown. This study investigated the effects of KM on RANKL-induced osteoclastogenesis in bone marrow-derived macrophages (BMMs). Cell viability, differentiation (TRAcP staining), and function (F-actin ring formation, bone resorption pit assay) were assessed. The underlying mechanisms were explored using Western blot and qPCR. The in vivo efficacy of KM was evaluated in an ova

Progesterone regulates the secretion of GnRH in bovine endometrial cells via the Erk1/2-ARRB1 pathway.

In ruminants, progesterone promotes uterine development to create a favourable environment for an embryo. However, the mechanisms by which progesterone regulates the function of the endometrial epithelial cells that line the uterus remain unclear. To investigate likely mechanisms, this study used different concentrations of progesterone to treat cells from a bovine endometrial epithelial cell (BEEC) line. The effect of the treatments was assessed by constructing mRNA expression profiles of the cells using RNA sequencing (RNA-Seq). Compared to the control cells (0 ng/mL progesterone), it was re

Melatonin Protects against LPS-Induced Mitochondrial Dyshomeostasis and Ovarian Damage through JNK Signaling Pathway in Mouse Ovary.

Both mitochondrial dysfunction and inflammation are closely associated with the pathogenesis of diminished ovarian reserve (DOR). While melatonin (MT) is known to protect against ovarian injury, its precise mechanism in counteracting lipopolysaccharide (LPS)-induced mitochondrial dysfunction and ovarian reserve impairment remains unclear. This study aimed to explore the effects and underlying mechanisms of MT on LPS-induced ovarian reserve dysfunction. Follicle development in mouse models was assessed using HE staining and follicle counting. Immunofluorescence, Western blotting, and quantitati

Genistein reverses the exacerbating effect of 17β-estradiol on experimental occlusal interference induced chronic masseter hyperalgesia through suppressing ERK1/2 signal pathway in spinal trigeminal nucleus of ovariectomized rats.

BACKGROUND: Temporomandibular disorder (TMD) pain is more prevalent in females than in males, with high estrogen levels potentially being a risk factor. Research indicates that 17β-estradiol (E2) exacerbates experimental occlusal interference (EOI)-induced orofacial hyperalgesia, which can be reversed by genistein. This study aimed to explore the central mechanisms within the spinal trigeminal nucleus (Sp5) related to the pain-exacerbating effect of E2 and the antiestrogenic properties of genistein in a model of EOI-induced chronic masseter pain. METHODS: Female rats underwent ovariectomy (OV

MiR-106a-5p in extracellular vesicles derived from alveolar epithelial cells mediates cognitive dysfunction induced by chronic intermittent hypoxia in mice through MAPK signaling pathway.

BACKGROUND: Obstructive sleep apnea (OSA), characterized by chronic intermittent hypoxia (CIH), is frequently associated with cognitive dysfunction. However, the underlying peripheral-central interplay mechanism remains to be elucidated. Recent years have witnessed the proposal of the “lung-brain axis (LBA)” concept, suggesting that lung tissue can remotely regulate brain function via extracellular vesicles (EVs). This investigation aims to determine whether EVs derived from alveolar epithelial cells (AEC-EVs) mediate CIH-induced cognitive impairment and to delineate the associated molecular m

Ginkgetin delays the progression of osteoarthritis by inhibiting the NF-κB and MAPK signaling pathways.

BACKGROUND: Osteoarthritis (OA) is considered an advancing chronic degenerative joint disease, leading to severe physical functional impairment of patients. Its development is closely related to increased inflammation and oxidative stress within the joint. Ginkgetin (GK), a natural non-toxic chemical, has proven anti-inflammatory, antioxidant, anti-tumor, and neuroprotective effects. METHODS: First, this study utilizes network pharmacology to explore the intrinsic connection between GK and OA. In vitro, SW1353 human cartilage cells were stimulated with Tert-butyl hydrogen peroxide (TBHP), and

DUSP1 interacts with BIP to regulate Staphylococcus aureus-induced apoptosis through the MAPK signaling pathway.

Staphylococcus aureus (S. aureus) is a major human pathogen that causes apoptosis of immune cells during infection. The rate of apoptosis influences the severity and outcome of the disease, and can be fatal in conditions such as sepsis and septicemia. Dual-specificity phosphatase-1 (DUSP1) is a negative regulator of the mitogen-activated protein kinase (MAPK) signaling pathway in the host innate immune response. However, its role in S. aureus-induced apoptosis remains unexplored. In this study, we investigated the function and underlying regulatory mechanisms of DUSP1 in S. aureus-induced apop

Autoinflammation in patients with leukocytic CBL loss of heterozygosity is caused by constitutive ERK-mediated monocyte activation.

Patients heterozygous for germline CBL loss-of-function (LOF) variants can develop myeloid malignancy, autoinflammation, or both, if some or all of their leukocytes become homozygous for these variants through somatic loss of heterozygosity (LOH) via uniparental isodisomy. We observed an upregulation of the inflammatory gene expression signature in whole blood from these patients, mimicking monogenic inborn errors underlying autoinflammation. Remarkably, these patients had constitutively activated monocytes that secreted 10 to 100 times more inflammatory cytokines than those of healthy individ
